Project description:Adrenal chromaffin cells and sympathetic neuron are derived from neural crest precursors and both cell types can give rise to childhood cancer, neuroblastoma. However only limited is known about the mechanism of their development. Better understanding of their transcriptomic profiles during development could gives an insight into the cell fates acquisition as well as the origin of neuroblastoma. Yellow fluorescent protein expressing sympathetic neuroblasts and adrenal chromaffin cells were isolated from E12.5 TH-IRES-Cre;ROSA26-EYFP mouse embryos by fluorescence-activated cell sorting. Transcriptomic profiles of sympathetic neuroblasts and adrenal chromaffin cells from embryonic age (E)12.5 TH-IRES-Cre;ROSA26-EYFP mice were generated by RNA sequencing, in four paired biological replicates.

Transcriptomes of lineage-specific Drosophila neuroblasts profiled via genetic targeting and robotic sorting

Project description:A brain consists of numerous distinct neurons arising from a limited number of progenitors, called neuroblasts in Drosophila. Each neuroblast makes a specific neuronal lineage. To unravel the transcriptional networks that underlie the development of distinct neuroblast lineages, we marked and isolated lineage-specific neuroblasts for RNA sequencing. We labeled particular neuroblasts throughout neurogenesis by activating a conditional neuroblast driver in specific lineages using various intersection strategies. The targeted neuroblasts were efficiently recovered using a custom-built device for robotic single cell picking. Transcriptome analysis on the mushroom body, antennal lobe, and type II neuroblasts besides non-selective neuroblasts, neurons, and glia revealed a rich repertoire of transcription factors expressed among neuroblasts in diverse patterns. In addition to those likely pan-neuroblast transcription factors, there exist many transcription factors selectively enriched or repressed in certain neuroblasts. The unique combinations of transcription factors present in different neuroblasts may govern the diverse lineage-specific neuron fates

Transcription profiling of GR-N-91 neuroblasts injected subcutaneously into nude mice and a primary tumor Xenograft (PTX) was isolated whilst metastatic neuroblasts were isolated from Myocardium (MY) and Bone Marrow (BM)

Project description:We have used an IGR-N-91 parental cell line established from an involved bone marrow harvested from a high-risk NB (stage 4-NB, 8 year-old boy). These IGR-N-91 neuroblasts were injected subcutaneously into nude mice and a Primary Tumor Xenograft (PTX) was isolated whilst metastatic neuroblasts were isolated from Myocardium (MY) and Bone Marrow (BM). These malignant neuroblasts were further cultured in vitro on bovine corneal extra-cellular matrix to establish sublines (IGR/PTX, IGR/BM and IGR/MY). Agilent oligo microarray analysis was performed to compare gene expression profiles in BM and MY metastatic versus PTX neuroblasts. This analysis used eight microarrays (two replicates plus two dye-swap experiments for both MY and BM cell lines).

MicroRNA Expression profiles of mature hepatocytes and foetal liver progenitor cells

Project description:Human liver progenitor cells (LPCs) show therapeutic potential, however, their in vitro culture results in inadequate function and phenotypic instability reflecting incomplete understanding of in vivo processes. Foetal LPCs capable of differentiation to a hepatocyte phenotype were isolated and mRNA expression profiling carried out using Exiqon miRCURY microarrays. This was compared to profiles from mature human hepatocytes. Foetal LPCs exhibit a distinct miRNA profile consistent with a stem cell signature, cell division, and some liver-specific functions.

Transcriptome of human foetal heart for comparison with cardiomyocytes from pluripotent stem cells

Project description:Gene expression in the human foetal heart for a comparison with cardiomyocytes derived from human pluripotent stem cells. Human foetal heart samples were collected from the individual chambers of the heart at various stages of development. This provided the opportunity to perform gene expression analysis and identify genes involved in the formation of the heart in each of the four chambers and at different stages of development. The dataset can be used to benchmark human pluripotent stem cell derived cardiomyocytes (hPSC-CMs) in terms of their maturation state by comparing it to the foetal heart samples. Two commercially available reference RNA sets are included in the analysis in order to characterize future sets of hPSC-CMs.
