Project description:LRG1 is a novel proangiogenic and profibrotic molecule that is upregulated in pathologies characterised by abnormal angiogenesis such as AMD and cancer. Blocking LRG1 by genetic models or by our in-house developed function blocking antibody leads to vascular normalsation. In this study, we have treated B16F0 melanoma tumours with 15C4, our function blocking antibody, or with IgG as a control. The tumours were collected at day 17 and the endothelial cell population was isolated by FACS sorting the CD31 positive cells. RNA was isolated and used for the preparation of cDNA libraries. Expression profile of the endothelial cell population was assessed by Illumina sequencing. Differenital expression analysis was conducted using the DESeq2 package in R.

Project description:To understand the molecular mechanisms during the maturation of cord blood-derived endothelial cells into blood brain barrier capillary endothelial cells (BCECs), we have employed whole genome microarray expression profiling to identify genes responsible for the maturation process. Hematopoietic stem cells were isolated from cord-blood samples and differentiated into endothelial cells. The endothelial cells were further maturated into BCECs by co-culturing with blood-brain barrier (BBB) specific cells (pericytes) for 3 days and 6 days.

Transcriptome of isolated mouse peritoneal endothelial cells

Project description:To characterize the metabolic profile of peritoneal endothelial cells (ECs) in response to peritoneal dialysis (PD), we performed RNA sequencing of peritoneal ECs isolated from mice treated with PD fluid for 6 weeks (n = 3) and from mice treated with saline for 6 weeks (n = 3). We demonstrated that peritoneal ECs had a hyperglycolytic metabolism in response to PD fluid treatment, which is associated with the development of microvascular alterations and peritoneal dysfunction.

Project description:The goal of this study was to gain insight into the molecular heterogeneity of capillary endothelial cells derived from different organs by microarray profiling of freshly isolated cells and identify transcription factors that may determine the specific gene expression profile of endothelial cells from different tissues. The study focused on heart endothelial cells and presents a validated signature of 31 genes that are highly enriched in heart endothelial cells. Within this signature 5 transcription factors were identified and the optimal combination of these transcription factors was determined for specification of the heart endothelial fingerprint.

Project description:Complete identification of the bone marrow niche remains one of the most progressing fields. Attempts to identify soluble factors involved in stem cell renewal have been less successful. We have previously shown that endothelial cells (EC) can induce the long-term proliferation of hematopoietic progenitor cells (HPC), especially when they had been subjected to an inflammatory stimulus like interleukins (IL) 1. To identify yet unknown growth factors, we compared the expression profile of IL-1 stimulated and non-stimulated endothelial cells. Human endothelial cells were subjected to IL-1 for 4, 8, and 16 hours. Isolated mRNA was analyzed by oligonucleotide microarray chips covering over 46,000 human genes.
