Project description:The objective of the study was to investigate how pristine CuO modulates allergic lung inflammation and whether surface modifications can influence its reactivity. CuO materials exacerbated the existing allergic airway inflammation by eliciting dramatic pulmonary neutrophilia. Transcriptomic analysis showed that CuO, CuO COOH and CuO NH3 commonly enriched neutrophil-related biological processes in lungs with and without the pre-existing condition. In contrast, CuO PEG had a significantly lower potential in triggering pulmonary changes revealing that surface PEGylation suppresses the effects triggered by the pristine material.

Project description:This study investigated the pulmonary toxicity of Copper oxide nanoparticles (CuO NPs) surface modified with polyethylenimine (PEI) or ascorbate (ASC), was investigated. Rats were exposed nose-only to a fixed exposure concentration of ASC or PEI coated CuO NPs for 5 consecutive days. On day 6 and day 27 post-exposure, pulmonary toxicity markers in bronchoalveolar lavage fluid (BALF) were analyzed and histopathological evaluation of the lungs was performed, along with microarray analyses on whole lung tissue samples.

Pulmonary effects of ozone and allergen exposure in the absence of surfactant protein-D in mice.

Project description:Ozone and allergen inhalation causes pulmonary inflammation and impairs lung function in mice. These effects are heightened in the absence of the immunosuppressive surfactant protein-D. Increased expression of inflammatory mediators is critical for the initiation of inflammation and impairment of lung function following ozone and allergen inhalation. An Affymetrix microarray was used to understand global changes in gene expression that occurred after ozone inhalation and/or allergen sensitization and challenge in the abscence of surfactant protein-D. Identification of the set of genes up and down regulated in response to ozone and/or allergen led us to study specific cellular inflammatory pathways. This study has important implications for the study of lung diseases like asthma.

Foxf1 enchances pulmonary inflammation and mastocytosis

Project description:The Forkhead Box f1 (Foxf1) transcriptional factor (previously known as HFH-8 or Freac-1) is expressed in endothelial and smooth muscle cells in the embryonic and adult lung. To assess effects of Foxf1 during lung injury, we used CCl4 injury model. Foxf1+/- mice developed severe airway obstruction and bronchial edema, associated with increased numbers of pulmonary mast cells and increased mast cell degranulation following injury. Pulmonary inflammation in Foxf1+/- mice was associated with diminished expression of Foxf1, increased mast cell tryptase and increased expression of CXCL12, the latter being essential for mast cell migration and chemotaxis. Foxf1 haploinsufficiency caused pulmonary mastocytosis and enhanced pulmonary inflammation following chemically-induced lung injury, indicating an important role for Foxf1 in the pathogenesis of pulmonary inflammatory responses. Serpinb3a contributes to early skin inflammation following allergen exposure

Project description:Serpinb3a knockout mice show attenuated Transepidermal Water Loss (TEWL) and S100A8 expression (an early inflammatory marker) following treatment with Aspergillus fumigatus extract. The aim of the RNAseq experiments was to provided mechanistic insight for the role of Serpinb3a by identifying early differences in gene expression following allergen exposure. Wild type Balb/c and Serpinb3a null mouse backs were shaved and 24 hours later a patch containing 200ug of Aspergillus fumigatus extract was applied to the skin. After 3 days the patch was removed and the skin was harvested after an additional 24 hours.
