Project description:Distinct subtypes of intracortically-projecting neurons (ICPN) are present in all layers, allowing propagation of information within and across cortical columns. How the molecular identities of ICPN relate to their defining anatomical and functional properties is unknown. Here we show that the transcriptional identities of ICPN primarily reflect their broad axonal projections rather than their date of birth or laminar position. Thus, conserved circuit-related transcriptional programs are at play across cortical layers, which may preserve canonical circuit features across development.

There were 1 case of grade I , 1 case of grade II, 3 cases of grade Ⅲ and 3 cases of grade Ⅳ according to Pfirrmann classfication. RNA seqencing analysis and single-cell RNA sequencing were integrated with proteomics data to identify the hub genes for intervertebral disc degeneration using bioinformatic method.
Project description:We have performed bulk RNA sequencing on colorectal tumors in order to determine their transcriptome-based molecular subtypes. RNA was isolated from fresh frozen human colorectal tumor specimens with Trizol extraction. RNA library preparation was performed with KAPA stranded mRNA sequencing kit, during which mRNA selection was performed with polyT capture beads. Sequencing was performed on Illumina HiSeq4000 as single-end 51 bp reads.
