No evidence for major spacial differences within the microbial communities in anaerobic digestion plants under different feeding and stirring regimes

Project description:In anaerobic digestion plants (ADP) homogenization of the feed, the fermenter content and the microbial communities represents a precondition for effective and robust biogas production,but also a major energy consumer. For a 850 m3 agricultural AD-P equipped with eight sampling ports we investigated whether different feeding and stirring regimes enable a sufficient homogenization of the microbial communities using metaproteome and TRFLP analysis. Systematic comparison of the samples by scatter plots and students t-test revealed only a limited number of slightly changed metaproteins, taxonomies and biological processes, indicating no systematic differences between the microbial communities in center and rim as well as between top and bottom. However, comparison of the amount of shared identified metaproteins between the sample ports showed minor variation, which might be correlated with the applied stirring strategy. In sum, the applied stirring and feeding conditionswere sufficient to homogenize the microbial communities in AD-Ps largely.

RNA-seq of mycorrhizal Medicago truncatula roots under K+ deprivation

Project description:Arbuscular mycorrhizal (AM) associations enhance the phosphorous and nitrogen nutrition of host plants, but little is known about their role in potassium (K+) nutrition. Medicago truncatula plants were co-cultured with the AM fungus Rhizophagus irregularis under high and low K+ regimes for six weeks. We determined how K+ deprivation affects plant development, mineral acquisition, and how these negative effects are tempered by the AM colonization. The transcriptional response of AM roots under K+ deficiency was analyzed by whole genome RNA-seq. K+ deprivation decreased root biomass, external K+ uptake, and modulated oxidative stress gene expression in M. truncatula roots. AM colonization induced specific transcriptional responses to K+ deprivation that seem to temper these negative effects. A gene network analysis revealed putative key regulators of these responses. This study confirmed that AM associations provide some tolerance to K+ deprivation to host plants, revealed that AM symbiosis modulates the expression of specific root genes to cope with this nutrient stress, and identified putative regulators participating in these tolerance mechanisms.

Gene expression in Human Bronchial Epithelial Cells under Differing Oxygen Regimes

Project description:Bronchial epithelial cells line the small airways bronchii and small airways of the lungs, acting to trap and remove contaminants deposited in the lung through the process of inhalation. These cells are sensitive decreased oxygen concentrations, and are invoved in local signaling in hypoxic conditions, such as when airways are occluded. We grew cultured human bronchial epithelial cells for three passes, and treated the fourth pass under three different oxygen concentrations (21%, 15%, 2%). RNA from each of three replicates of each treatment was extracted, processed, and hybridized onto Affymetrix Human Transcriptome Array 2.0 microarrays to examine alterations in gene expression under these oxygen regimes. This data was collected to complement ATAC-seq data from cells aliquotted from each replicate.
