Project description:Systems responses of mature leaves from 4 reference cultivars of a larger collection of European potato cultivars (Solanum tuberosum L.) are investigated by metabolome profiling and RNA-Sequencing. The chosen reference cultivars, Milva, Alegria, Desiree, and Saturna, vary in ascending order in regard to drought tolerance. Systems analyses are based on 3 independent field trials and 3 paralleled greenhouse trials. Robust responses across all cultivars and conditions to natural seasonal drought stress comprise proline, raffinose, galactinol, arabitol, arabinonic acid, chlorogenic acid, and 102 transcripts which consist to a high proportion of heat shock proteins and genes with signaling or regulatory functions, such as a homolog of abscisic acid receptor PYL4. Constitutive differences of the tolerant cultivars, Desiree and Saturna, compared to the sensitive cultivars include arbutin (hydroquinone-beta-D-glucopyranoside), octopamine (p-hydroxyphenylethanolamine), ribitol and 248 differential transcripts. Many of these transcripts are disease related, receptor kinases, or regulatory genes, for example a homolog of the Arabidopsis FOUR LIPS MYB-regulator of stomatal cell proliferation. Functional enrichment analyses imply that heat stress is a major acclimation component of potato leaves to agronomical relevant drought stress. Enhanced leaf heat stress is a result of drought caused by loss of transpiration cooling. This effect and CO2-limitation are the main dilemmas of drought- or ABA-induced stomatal closure. Constitutive differences between tolerant and sensitive cultivars indicate partially synergistic interactions of drought and biotic stress responses. We suggest that drought tolerance of the potato reference cultivars may be caused by general resistance mechanisms which are part of previously selected pathogen tolerance. Project description:Two cultivars of sorghum were examined from a study performed at the Kearney Research Station in California; 1) a pre-flowering drought tolerant, post-flowering drought susceptible cultivar RTx430 and 2) a pre-flowering drought susceptible, post-flowering drought-tolerant cultivar BTx642. These data are the methanol/water metabolite fraction obtained from a Folsh extraction from Sorghum bicolor leaves and roots. The metabolites were derivatized and analyzed via GC-MS at PNNL. Analyses represent the metabolite profile collected at 2, 3, 4, 6, 8, 9, 10, 11, 13, 15, 17 weeks post soil emergence and under a perturbation of either well-watered (control), pre-flowering drought, or post-flowering drought.

Project description:Drought is among the most limiting factors for sustainable agricultural production. Water shortage at the onset of flowering severely affects the quality and quantity of grain yield of bread wheat (Triticum aestivum). Herein, we measured oxidative stress and photosynthesis-related parameters upon applying transient drought on contrasting wheat cultivars at the flowering initiation stage of ontogenesis. The sensitive cultivar showed ineffective water management and a more severe decline of photosynthesis. Apparently, the tolerant genotype used photorespiration to dissipate excessive light energy. The tolerant cultivar sooner induced superoxide dismutase and showed less inhibited photosynthesis. Such protective effect resulted in less affected yield and spectrum of seed proteome. The tolerant cultivar had a more stable gluten profile, which defines bread-making quality, upon drought. Drought caused the accumulation of medically relevant proteins: (i) components of gluten in the sensitive cultivar and (ii) metabolic proteins in the tolerant cultivar. We propose specific proteins as markers of drought tolerance for guiding efficient breeding: thaumatin-like protein, 14-3-3 protein, peroxiredoxins, peroxidase, FBD domain protein, and Ap2/ERF plus B3 domain protein.

Project description:A heat and drought tolerant rice cultivar (N22) was grown in the field under control and drought conditions during the dry season in 2013. Drought was applied during early grain filling and resulted in simultaneous heat stress, leading to reduced grain yield and quality. Total RNA was extracted from developing seeds under stress and control (fully flooded) conditions and RNA-seq analysis was performed. These samples are a part of a bigger experiment analysing the responses of three contrasting rice cultivars (N22, Dular, Anjali) to combined heat and drought stress including different organs (developing seeds, flag leaves, flowering spikelets) and developmental stages (early grain filling, flowering) at the transcriptomic level.

A liquid-based bottom up proteomics analysis was performed using an iTRAQ labeling multiplexing approach to determine relative abundances. After the peptides were digested with trypsin and iTRAQ labeled, they were fractionated into 12 fractions offline using a C-18 column under high-pH solvent conditions Each fraction was further separated on a low pH C-18 column with direct infusion into a ThermoScientific Q-Exactive mass spectrometer. Project description:Drought is one of the main climate threats for crop plant production limitation. Climate change models predict constant global warming accompanied by strong reduction in water availability, especially for agricultural needs. Potato belongs to crop plants that are considered as sensitive to water shortage. Global estimation analyses show that drought may decrease potato yield by 18-32% in the period of 2040-2069 (Hijmans, 2003, Obidiegwu et al. 2015, Front in Plant science). Crop models predict that potato yields may reduce by ~30% as a result of water deficit in Poland (http://www.climateadaptation.eu/poland/agriculture-and-horticulture/). Genetic variability between potato cultivars has been described regarding their tolerance to drought (Soltys-Kalina et al. 2016). To diminish the effect of forecasted potato harvest losses, it is crucial to identify as many as possible potato plant strategies to withstand long drought periods during vegetative season. For this reason, we decided to analyse the expression differences in transcriptomes independently in two selected pairs of potato cultivars, Gwiazda/Oberon and Tajfun/Owacja. Cultivars in each pair are closely related to each other (having one parent in common or one grandparent in common, respectively) but differ in their sensitivity to drought conditions. In this paper, we identified at least 24 top selected genes whose expression profiles differ significantly during drought period when closely related studied cultivars are compared. Moreover, all but one of selected potato genes have their homologues in Arabidopsis plant genome. We found that A. thaliana mutants with mostly downregulated expression of seven selected homologous genes differ in their response to drought. To our knowledge, all of these genes were until now not reported as drought-related. Thus, our original approach and obtained results allowed to identify new players in plant response to drought.

Project description:In many potato cultivation regions, production is constrained by abiotic stresses such as drought and high temperatures which are often present in combination. We aimed to identify key mechanisms and processes underlying single and combined abiotic stress tolerance by a comparative analysis of tolerant and susceptible cultivars. Physiological data supported cultivars Desiree and Unica as being abiotic stress tolerant, while Agria and Russett Burbank were stress susceptible. This was indicated by the stronger impact of abiotic stress on photosynthetic carbon assimilation in the susceptible cultivars. Similarly, susceptible cultivars exhibited a lower leaf transpiration rate following stress, particularly combined heat and drought stress. Transcript profiles using microarrays were highly divergent both between genotypes and following the application of stress treatments. However, relatively few transcripts or metabolites exhibited genotype specific responses to abiotic stress treatment. Furthermore, apart from a decrease in the abundance of transcripts associated with PSII, particularly the light harvesting complex in both Desiree and Unica, there were very few changes that were consistent across stress susceptible or stress tolerant genotypes following stress treatment.

Project description:Water deficit stress between the booting and grain filling stages significantly affect grain yield and quality of hard red winter wheat. Several stress tolerant cultivars with different adaptation mechanisms have been released and are widely cultivated on the Southern Great Plains of the US. However, the physiological, molecular, and genetic basis of adaptation to drought stress for these cultivars remains unknown. Use of transcriptome profiling to identify drought responsive genes in hexaploid wheat is a challenging process given the quantitative nature of drought stress, genome complexity, and the intricacy of interaction effects. If the information generated from functional genomics studies is to be used in molecular breeding programs for cultivar development, it is highly desirable to use cultivars better adapted for the region. In the current study we used two well-adapted, drought-tolerant, high-yielding, cultivated varieties, TAM 111 and TAM 112, which appear to have different adaptation mechanisms, to identify drought stress induced transcripts during heading and early dough stages. A set of 24 Affymetrix GeneChip wheat genome arrays (2 cultivars; 2 water treatments; 2 sampling stages; 3 biological replicates) from plants subjected to water deficit stress under controlled glasshouse conditions. Differentially expressed genes were identified using a ANOVA (p<0.01) controlling false discovery rate (FDR, q<0.01) using Benjamini Hochberg approach.
