Project description:The endocrine changes and gene regulation patterns responding to sexual maturation and spawning have not been clarified in fish species. To identify the potential metabolites and genes regulating the ovarian development and spawning, we employed the approach integrating the metabolic and RNA-seq techniques to investigate the metabolites and candidate genes participating in the sexual maturation and spawning of female blunt snout bream Megalobrama amblycephala. This data descriptor provides metabolic and transcriptomic information for the blood tissue at different developmental stages of female M. amblycephala. With UPLC-MS/MS method, a total of 763 and 173 differential ions were detected from the pairwise comparisons of the four groups in positive and negative mode, respectively. Meanwhile, 191.65 Mb raw reads were generated using illumina Hiseq platform with 50 bp single-end strategy. We then analysed the differential metabolites and differentially expressed genes and their functions among the four groups. This data descriptor will contribute to understanding the dynamics of metabolites and gene regulation in ovarian development and spawning and reuse to support physiological and genetic researches in aquaculture.

Plasma metabolomics profiling for fish maturation in blunt snout bream

Project description:We investigated the comprehensive metabolic profiles of plasma among immature females, mature females ready to spawn, as well as already spawned breeders of blunt snout bream (Megalobrama amblycephala). The purpose of this study was to screen out potential biomarkers for sexual mature female M. amblycephala compared to immature female individuals and already spawned breeders. The three groups were set up in this study, including one year old immature females, 2 years old sexually mature females ready to spawning and successfully spawning females of M. amblycephala. The plasma samples were collected to investigate the comprehensive metabolic profiles through UPLC-MS/MS based metabolomics analysis method. According to multivariate and univariate statistical analysis, plasma metabolite profiles of three groups were obviously separated, and the plasma metabolite profiles of immature female M. amblycephala were much more different from mature females ready to spawn as well as already spawned breeders. The differential plasma metabolites from three hormone related pathways including GnRH signaling pathway, steroid hormone biosynthesis and steroid biosynthesis, were further analyzed. A total of 29 metabolites were identified as differential biomarkers associated with the female maturation status

Identification and characterization of microRNAs involved in growth of blunt snout bream (Megalobrama amblycephala) by Solexa sequencing

Project description:The present study provides the first large-scale characterization of miRNAs in M. amblycephala and miRNA profile of M. amblycephala with different growth performance. The discovery of miRNA resource from this study is expected to contribute to a better understanding of the miRNAs roles playing in regulating the growth biological processes in fish and the study of miRNA function and phenotype-associated miRNA identification in fish. To investigate the functions of miRNAs associated with growth of M. amblycephala, we adopted the Solexa sequencing technology to sequence two small RNA libraries prepared from four growth related tissues (brain, pituitary, liver and muscle) of M. amblycephala using individuals with relatively high and low growth rates.
