Project description:The role of microRNAs (miRNAs) during mouse early development, especially in endoderm germ layer formation, is largely unknown. Here, using miRNA profiling, we discovered that miR-124a negatively regulates endoderm lineage commitment in mouse embryonic stem cells (mESCs). We showed that miR-124a inhibits endoderm differentiation in vitro through targeting the 3’ untranslated region (UTR) of Sox17 and Gata6, revealing the existence of an interplay between miR-124a and Sox17/Gata6 transcription factors in hepato-specific gene regulation. Besides, we proposed an in vivo system that utilizes teratoma to evaluate the functional role of miRNA in lineage specification. We demonstrated that ectopic expression of miR-124a in teratomas suppressed endoderm and mesoderm lineage differentiation while augmented the differentiation of ectoderm lineage. Collectively, our findings suggested that miR-124a plays a significant role in mESCs lineage commitment.

Project description:OBJECTIVE: To identify the changes in gene expression elicited by miR-27b-3p mimic transfection of human osteoarthritis (OA) primary fibroblast-like synoviocytes (FLS) obtained from patients undergoing knee arthroplasty. Cells were cultured, and treated for 48 h with miR-27b-3p mimic or Cel-miR-39-3p control mimic. Total RNA was then extracted, and mRNA libraries were prepared using Illumina's TruSEQ stranded total RNA library preparation kit, pooled together, and sequenced on Illumina's NextSeq550.

Project description:Tumor progression is accompanied by an altered myelopoiesis that causes the accumulation of cells inhibiting anti-tumor T lymphocytes. We previously reported that immunosuppressive cells can be generated in vitro from bone marrow cells (BM) after four days GM-CSF and IL-6 treatment. Here, we describe that miR-142-3p down-regulation directs macrophage differentiation and determines the acquisition of their immunosuppressive function in cancer. Enforced miR over-expression impaired monocyte to macrophage transition both in vitro and in vivo. Conversely, forced miR down-regulation promoted the generation of immunosuppressive macrophages even during G-CSF-induced granulocytic differentiation. To identify how miR-142-3p regulates MDSC generation and activity, we analyze the gene expression of BM cultures transfected with either CTRL- or miR 142-3p mimic oligo -transfected before four days GM-CSF and IL-6 treatment. Keywords: Expression profiling by array BM cells were transfected either CTRL- or miR 142-3p mimic oligo before GM-CSF and IL-6 treatment to generate in vitro MDSCs during enforced miR over-expression. A triplicate of each sample was considered.Total RNA from obtained in vitro BM-differentiated MDSCs was isolated by Trizol reagent, and cRNA samples were hybridized to the Affymetrix GeneChip MOE430 2.0.

Project description:Although the miR-124a-3p knockdown mouse exhibited neuronal dysfunction and dysmaturation by disinhibition of Lhx2, its downstream responses in mouse retinal development after birth were still relatively deficient at the transcriptome level. In our previous study, miR-124-3p exhibited a significantly increasing trend after birth, and its related pathways predicted by bioinformatics analysis were associated with biological processes that may play crucial roles in mouse retinal development. To gain insight into its downstream processes, RNA-Seq was used to obtain the expression profile of mRNA transcripts after the knockdown of miR-124-3p.
