Effects of Electroacupuncture on the CircRNA of Plasma Exosomes in Diabetic Mice: Based on RNA Sequencing Technique

Project description:Aim: To study the effect of electroacupuncture (EA) on circRNA expression of plasma Exosomes and signal pathway of type 2 diabetic mice (T2DM). Methods: 10 mice were randomly selected as normal group. 20 mice were for T2DM model preparation and then were randomly divided into model and model + EA group. Mice in model + EA group were given EA treatment. The changes of fasting blood glucose (FBG) and the Islet structure were evaluated. Plasma exosomes of mice were subjected to RNA sequencing. Bioinformatics analysis on differentially expressed circRNA was performed. 12 differentially expressed circRNA were confirmed for real-time quantitative PCR (qPCR). Results: EA treatment reduced the FBG, preserved islets structure and reduced the islet β cell apoptotic rate of T2DM mice. RNA-sequencing analysis showed EA treatment lead to significant change of 165 circRNA expressions. GO and KEGG revealed that the effects of EA on T2DM mainly focus on regulating the functions and pathways related to multi-link metabolism, cell growth and organ development. Especially, the thyroid hormone signaling pathway was actively regulated by EA. circRNA/miRNA interactions analysis revealed that that mmu-mir-7092-3p was closely combined with circINPP4B, suggesting phosphatidylinositol signaling pathway was affected by EA. 12 circRNAs were identified to have significant differences by qRT-PCR. Conclusion: EA intervention can significantly protect islet function and improve FBG in T2DM, which may be related to the regulation on thyroid hormone and Phosphatidylinositol signaling pathway.

Project description:Intrahepatic cholangiocarcinoma (ICC) is a liver tumor featured by challenges of non-invasive early diagnosis and a higher prevalence rate in Asian countries. These characteristics necessitate the development of liquid biopsy and immunotherapy methods to improve the prognosis of patients with ICC. Herein, we conducted a pilot study on the transcriptome of tumor tissues, adjacent normal tissues, and plasma exosomes of Asian patients with ICC from northern and southern China. We identified a subgroup of immunogenic Asian ICC, which is different from Caucasian ICC and is characterized by T cell exhaustion and neutrophil extracellular traps. The levels of circ-PTPN22 (hsa_circ_0110529) and circ-ADAMTS6 (hsa_circ_0072688), potential circRNA biomarkers, were elevated in the ICC tumor tissues and plasma exosomes of this subgroup than in the other subgroups and normal controls. These circRNAs were derived from post-transcriptional backsplicing of PTPN22 and ADAMTS6 that were expressed in T cells and endothelial cells, respectively, in the ICC microenvironment. Our results revealed a subgroup of Asian ICC characterized by T cell exhaustion and neutrophil extracellular traps and marked by elevated levels of circ-PTPN22 and circ-ADAMTS6 in tumor tissues and plasma exosomes. This subgroup is potentially detectable by plasma exosomal circRNAs and treatable with immune checkpoint blockade.

The miRNAs in plasma exosomes may serve as markers for segmental vitiligo

Project description:More and more studies have showed that plasma exosomal miRNAs are biomarkers for disease. The aim of the study were to investigate the miRNA profiling in plasma exosomes of patients with segmental vitiligo (SV) and to find biomarkers in plasma exosomes for patients with SV. Plasma exosomes and exosomal RNA of 7 SV patients and 8 health persons were purified by exoRNeasy Serum/Plasma Maxi Kit. The miRNA profiles of the 15 samples were sequenced using HiSeq 2500 (Illumina) and analyzed by Reads Per Million (RPM) values and edgeR algorithm. Some differently expressed miRNAs in plasma exosomes and skin tissues of the two sets were validated by qRT–PCR.A total of 85 miRNAs in plasma exosomes showed differential expression between SV patients and health persons, with a |log2（Fold Change）|≥1 and P-value < 0.05. Several miRNAs were confirmed by qRT–PCR and showed similar expression patterns between plasma exosomes and skin tissues. Our study depict the miRNAs expression profiles in plasma exosomes of SV patients and suggest that several miRNAs in plasma exosomes may serve as biomarkers for SV.
