Gene Expression Patterns in Peripheral Blood Mononuclear Cells Associated with Asthma Exacerbation Attack

Project description:A large, prospective, non-interventional study was designed to study gene expression changes in peripheral blood mononuclear cells (PBMCs) associated with asthma exacerbations over the course of a year. PBMC samples were collected from subjects at the time of the study visits defined as 1) Quiet: during stable disease at 3 month intervals, 2) Exacerbation: during a 14 day period of deteriorating asthma and 3) Follow-up: within 14 days after cessation of an exacerbation. Gene expression levels during stable asthma, exacerbation, and two weeks after an exacerbation were compared. This series of samples comprises of peripheral blood mononuclear cells (PBMCs) collected during the following study visits (a) quiet (N=394 samples from 118 subjects), (b) exacerbation (N=166 samples from 118 subjects) and (c) follow-up (N=125 samples from 102 subjects)

SINGLE-CELL CHARACTERIZATION OF A MODEL OF POLY I:C-STIMULATED PERIPHERAL BLOOD MONONUCLEAR CELLS IN SEVERE ASTHMA

Project description:Background: Asthma has been associated with impaired interferon response. Multiple cell types have been implicated in such response impairment and may be responsible for asthma immunopathology. However, existing models to study the immune response in asthma are limited by bulk profiling of cells. Our objective was to Characterize a model of peripheral blood mononuclear cells (PBMCs) of patients with severe asthma (SA) and its response to the TLR3 agonist Poly I:C using two single-cell methods. Methods: Two complementary single-cell methods, DropSeq for single-cell RNA sequencing (scRNA-Seq) and mass cytometry (CyTOF), were used to profile PBMCs of SA patients and healthy controls (HC). Poly I:C-stimulated and unstimulated cells were analyzed in this study. Results: PBMCs (n=9,414) from five SA (n=6,099) and three HC (n=3,315) were profiled using scRNA-Seq. Six main cell subsets, namely CD4+ T cells, CD8+ T cells, natural killer (NK) cells, B cells, dendritic cells (DCs), and monocytes, were identified. CD4+ T cells were the main cell type in SA and demonstrated a pro-inflammatory profile characterized by increased JAK1 expression. Following Poly I:C stimulation, PBMCs from SA had a robust induction of interferon pathways compared with HC. CyTOF profiling of Poly I:C stimulated and unstimulated PBMCs (n=160,000) from the same individuals (SA=5; HC=3) demonstrated higher CD8+ and CD8+ effector T cells in SA at baseline, followed by a decrease of CD8+ effector T cells after poly I:C stimulation. Conclusions: Single-cell profiling of an in vitro model using PBMCs in patients with SA identified activation of pro-inflammatory pathways at baseline and strong response to Poly I:C, as well as quantitative changes in CD8+ effector cells. Thus, transcriptomic and cell quantitative changes are associated with immune cell heterogeneity in this model to evaluate interferon responses in severe asthma.

Project description:Peste des Petits Ruminants (PPR) is a highly infectious disease caused by a virus of the genus Morbillivirus (PPRV), infecting mainly sheep and goat. Susceptibility of host can vary widely with host breed and virus strain. The mechanisms underlying this variability are not well understood. Here, we carried out the first comparative in vitro study on goat peripheral blood mononuclear cells (PBMCs) infected with PPRV strains of different virulence, vaccine strain (N75/1), low- (IC89), and high-virulent strain (MA08). Goat PBMCs were infected by the different strains and stimulated with Concanavalin A (Con A), and proteome changes of these cells were evaluated during the infection. The level of PPRV replication was assessed first by RT-qPCR quantification of viral N mRNA and by labelling the viral N protein inside the cells with specific antibodies and analysed by flow cytometry. The dynamics of PBMC sub-populations were also evaluated by flow cytometry. Our results showed that viral replication is critical for PPRV inhibitory effect on PBMCs proliferation. Highly virulent MA08 strain reached a higher level of replication in ConA-stimulated PBMCs and induced higher mortality compared to other strains. Low-virulent IC89 strain showed the lower replication level and cell proliferative inhibitory capacities. Differences in immune genes expression levels were assessed using RNAseq analysis and protein expression was compared using mass spectrometry. Analysis of these data provided the transcriptional and proteome landscape of PBMCs infected with these strains. The possible association between the transcriptional and proteome landscape and changes in immune cell subpopulations dynamics was explored.

LncRNAs involves in numerous functionally different biological and physiological processes. The aim of this study is to identify the differentially expressed lncRNAs and mRNAs in patients with asthma and to further explore the functions and interactions between lncRNAs and mRNAs. Ten patients with asthma and 9 healthy controls were enrolled in this study. We used microarray analysis to identify lncRNA and mRNA expression in their PBMC.

Transcriptome analysis of differentially expressed genes by tryptophanyl-tRNA synthetase 1 in human peripheral blood mononuclear cells

Project description:Tryptophanyl tRNA synthetase 1 (WARS1) is classically known as an essential housekeeping enzyme that ligates tryptophan to its cognate tRNA for protein synthesis. Interestingly, in the presence of pathogenic infection, WARS1 is immediately released from monocytes into the extracellular space. Secreted WARS1 promotes the innate inflammatory response by activating toll-like receptor (TLR) 2 and TLR4/myeloid differentiation factor-2 (MD-2) signaling. The purpose of this study was to investigate the expression profile of genes associated with the inflammatory response in WARS1-stimulated human peripheral blood mononuclear cells (PBMCs). As a result, WARS1 significantly upregulated the expression of genes associated with TREM1, IL-8, and IL-6 signaling in PBMCs.
