Project description:Cancer stem cells (CSCs) are proposed to be unique subsets of clones within a tumor, attributed with tumor progression, resistance to chemotherapy, and metastases. However, little is known about the molecular mechanisms regulating the acquisition and maintenance of CSC properties in cancers, especially in rare cancers, such as osteosarcoma (OS). In this study, we generated OS cells with CSC properties following transduction of defined factors. This method enabled us to obtain abundant OS CSC material. The results of our study could facilitate a better understanding of the mechanisms of OS progression, and aid in the development of new therapies targeting CSCs in patients with OS.

Project description:Differentiated cells can be reprogrammed to an embryonic-like state by transfer of their nuclear contents into oocytes or by fusion with embryonic stem (ES) cells. Little is known about the factors that induce this reprogramming. Here we show that the combination of four factors, Oct3/4, Sox2, c-Myc, and Klf4, can generate pluripotent-like stem cells directly from mouse embryonic or adult fibroblast cultures. Unexpectedly, Nanog was dispensable in this process. These cells, which we designated iPS (induced pluriopotent-like stem) cells, exhibit the morphology and growth properties of ES cells and express ES cell marker genes. Subcutaneous transplantation of iPS cells into nude mice resulted in tumors containing a variety of tissues from all three germ layers. Following injection into blastocysts, iPS cells contributed to mouse embryonic development. These data demonstrate that pluripotent-like cells can be directly generated from fibroblast cultures by the addition of only a few defined factors. Keywords: cell type comparison

Genomic Acquisition of Virulence Factors in Natural Isolates of Burkholderia

Project description:An important, but rarely performed, test of Koch’s molecular postulates involves evaluating the capacity of candidate virulence genes to confer pathogenicity in otherwise non-virulent species. Unbiased genomic surveys of avirulent natural isolates might reveal rare variants possessing specific virulence features, which might prove useful in testing their functional sufficiency. Using a custom pan-genome array, we analyzed a panel of avirulent Burkholderia thailandensis (Bt) isolates related to Burkholderia pseudomallei (Bp), the causative agent of the often fatal human and animal disease melioidosis. We report the discovery of variant Bt isolates exhibiting isolated acquisition of a capsular polysaccharide biosynthesis gene cluster (BpCPS), long regarded as an critical species-specific virulence factor essential for Bp mammalian virulence. BpCPS-expressing Bt strains exhibited certain pathogen-related phenotypes including resistance to human complement binding, but did not exhibit enhanced virulence when assessed in two different in vivo animal infection models. Phylogenetic analysis revealed that the BpCPS-expressing Bt strains likely reside within an evolutionary subgroup distinct from the majority of previously-described Bt strains. Our findings suggest that BpCPS acquisition alone is unlikely to fully explain the ability of Bp to colonize humans and animals, highlighting the importance of other collaborating factors in the pathogenesis of mammalian melioidosis. Super-enhancer-driven MLX mediates redox balance maintanance via SLC7A11 in osteosarcoma

Project description:A systematic analysis of super-enhancers identified MLX as a potential oncogene in osteosarcoma. Knockdown of MLX impaired tumor aggressiveness in vitro and in vivo, suggesting oncogenic properties of MLX. Mechanistically, silencing of MLX downregulates SLC7A11, a key gene encoding glutamate/cystine antiporter, to attenuate the uptake of cystine and interrupt the redox balance, leading to ferroptotic cell death. Pharmacological inhibition of SLC7A11 triggered massive ferroptosis and caused impaired tumor growth, providing a promising approach for osteosarcoma treatment.
