Project description:Osteoarthritis (OA) is a highly prevalent and morbid disease affecting over 30 million Americans. Effective drugs and treatments to halt the progression or reverse OA pathogenesis are not available, creating a need for translational research in the field. The A2AR null mouse is an important animal model for the understanding of disease pathogenesis and the development of novel therapies. We have previously shown that these mice develop OA spontaneously by age 16 weeks and that liposomal adenosine is able to reverse established OA in animal models of post-traumatic OA and overbearing/overweight OA. We here illustrate how the A2ARKO transcriptome shows early compromise of cell viability and compares to that of human OA chondrocytes.

Transcriptional profiling of Adenosine receptor (AdoR) null mutant in Drosophila melanogaster imagoes

Project description:Dysregulation of adenosine (Ado) homeostasis has been observed in both rodent models and human patients of Huntington’s disease (HD). However, the underlying mechanisms of Ado signaling in HD pathogenesis is still unclear. In this study we examined influence of Ado signaling on Drosophila HD model. We further examined the transcription profile of AdoR mutants by microarray analysis to identified a downstream target of AdoR signaling, which mediates the AdoR effects on HD pathology. Our findings have important implications for the crosstalk between Ado signaling and pathogenic effects of HD as well as other human diseases associated with polyglutamine aggregation.

Metabolomics of the interaction between PPAR-alpha and age in the PPAR-alpha-null mouse (NMR assay)

Project description:Regulation between the fed and fasted state in mammals is partially controlled by peroxisome proliferator activated receptor-alpha (PPAR-alpha). Expression of the receptor is high in liver, heart and skeletal muscle, but decreases with age. A combined 1H NMR spectroscopy and GC-MS metabolomic approach has been used to examine metabolism in liver, heart, skeletal muscle and adipose tissue in PPAR-alpha null mice and wild type controls during ageing between 3-13 months. For the PPAR-alpha-null mouse multivariate statistics highlighted hepatic steatosis, reductions in the concentrations of glucose and glycogen in both liver and muscle tissue, and profound changes in lipid metabolism in each tissue, reflecting known expression targets of the PPAR-alpha receptor. Hepatic glycogen and glucose also decreased with age for both genotypes. These findings indicate the development of age related hepatic steatosis in the PPAR-alpha-null mouse, with the normal metabolic changes associated with ageing exacerbating changes associated with genotype. Furthermore, the combined metabolomic and multivariate statistics approach provides a robust method for examining the interaction between age and genotype. </p> The GC-MS assay for this study can be found in the MetaboLights study MTBLS314.

Gene expression profile in the heart of wild type and Adenosine Receptor A2a over expressing mice

Project description:Adenosine binds to 4 G protein-coupled receptors located on the cardiomyocyte (A1-R, A2a-R, A2b-R and A3-R) and modulates cardiac function during both ischemia and load-induced stress. While the role of adenosine receptor-subtypes has been well defined in the setting of ischemia-reperfusion, far less is known regarding their roles in protecting the heart during other forms of cardiac stress. We characterized the gene expression profiles of heart from A2a-R over-expressing mice subjected to transverse aortic constriction to study the potential of A2a-R activation to protect from pressure-induced heart failure.

Project description:Microglial activation often accompanies the plastic changes occurring in the brain of patients with neurodegenerative diseases. A2A and A3 adenosine receptors have been proposed as therapeutic targets to combat neurodegeneration. RNAseq was performed using samples isolated from lipopolysaccharide/interferon-γ activated microglia treated with SCH 58261, a selective A2A receptor antagonist and 2-Cl-IB-MECA, a selective A3 receptor agonist.

Project description:We have used an agnostic approach to identify drug combinations by using combination high throughput screening (cHTS) technology and make the surprising discovery that adenosine A2A and beta-2 adrenergic receptor agonists are highly synergistic, selective and novel agents that enhance glucocorticoid activity in B-cell malignancies. We used the microarray study to understand the synergistic mechanism between dexamethasone and A2A receptor agonist or Beta-2 Adrenergic receptor agonist in a multiple myeloma cell line, MM1S. MM1S cells were treated with CGS-21680 or Salmeterol alone, or in combination with dexamethasone for six hours for RNA extraction and hybridization on Affymetrix microarrays.
