Project description:We reported the hepatic XBP1s cistrome (starting at CT0 and proceeding every four hours for three complete 12-hour cycles) in the liver of wild-type mice and XBP1 liver-specific knockout mice; we also chose liver samples at CT0 and CT8 (four hours before the dusk 12-hour rhythmic peak) for comprehensive XBP1s ChIP-Seq to interrogate the genomic binding profiles of hepatic XBP1s in vivo.

Project description:Age- and sex-matched (male, 2-3 months old) ERT2 AlbCre Ifnar fl/fl and ERT2 AlbCre Ifnar +/+ mice were injected intraperitoneally with 40 mg/kg tamoxifen for 5 consective days. Mice were then intravenously infected with 2x10^6 focus forming units of LCMV Cl13 and liver tissue of either genotype was harvested 1.5 days post infection and analyzed for transcriptomic changes (n = 3). Liver tissue of uninfected animals of both genotypes was harvested as control (n = 3).

Project description:Epithelial mesenchymal plasticity (EMP) is a complex cellular reprogramming event that plays a major role in tissue homeostasis to infections and injury. Recently we elucidated a mechanism wherein the unfolded protein response (UPR) triggers EMP through the inositol-requiring protein 1 (IRE1α)–X-box-binding protein 1 (XBP1) axis, enhancing glucose shunting to protein N glycosylation producing ER stress. To better the genomic targets for the IRE1-XBP1 pathway that activate compensatory metabolic changes in EMP, we systematically identified the genomic XBP1 targets using Cleavage Under Targets and Release Using Nuclease (CUT&RUN) of a FLAG-epitope tagged XBP1 in RSV infection. CUT&RUN identified 7,086 enriched binding sites mapped to 4,827 genes; of these XBP1 binds to 2,119 sites within 1 kb of the transcription start sites. Interestingly, XBP1 binds to 322 superenhancers associated with RHO GTPase signaling that were associated with largely inert gene expression. By contrast, XBP1 binds to proximal promoters inducing coordinate mRNA expression of hexosamine biosynthetic enzymes encoding sequential steps in UDP-GlcNAc biosynthesis through AGCTCA motifs. We demonstrate that IRE1-XBP1 signaling is necessary and sufficient to activate core HBP enzymatic pathway by recruiting transcriptional elongation-competent phospho-Ser2 CTD modified RNA Pol II (pSer-PolII).

Gene expression profiles of livers from male LMNA+/+, LMNA flx/+, and LMNA flx/flx; Alb-Cre+ C57BL/6 mice fed normal chow or high fat diet

Project description:To investigate the molecular basis for male-specific steatohepatitis in Lamin A/C-deficient livers, microarray gene expression analysis was performed on total liver RNA isolated from 26- to 39-week-old, male LMNA+/+, LMNA flx/+, and LMNA flx/flx; Alb-Cre+ C57BL/6 mice fed either normal chow (NC) or high fat diet plus carbohydrate-supplemented water (HFD). Lamins are nuclear intermediate filament proteins that comprise the major components of the nuclear lamina in metazoan cells. Mutations in LMNA, which encodes lamins A/C, cause diseases termed laminopathies, including lipodystrophy, cardiomyopathy, and premature aging. The lamin A/C mutation-associated Dunnigan familial partial lipodystrophy is typically accompanied by fatty liver disease. The role of lamins in the liver is unknown, and it is unclear whether laminopathy-associated liver disease is due to primary hepatocyte defects or systemic alterations. To address these questions, mice carrying a hepatocyte-specific deletion of Lmna (KO mice) were generated. KO hepatocytes manifested abnormal nuclear morphology, and KO mice developed spontaneous male-selective hepatosteatosis, with increased susceptibility to high fat diet-induced steatohepatitis and fibrosis. The molecular mechanism by which liver-specific Lamin A/C deficiency induces male-specific steatohepatitis is unknown. The microarray data presented here demonstrates that hepatic Lmna deficiency is associated with upregulated expression of fatty acid transporters, lipid biosynthetic enzymes, lipid-droplet associated proteins, and interferon-regulated genes and other pro-inflammatory mediators.
