Project description:To evaluate the potential role of Hes1 in regulation of type I IFNs and the downstream ISGs expression, we generated the bone marrow-derived macrophages (BMDMs) from Hes1+/+ Cre-ERT2 and Hes1fl/fl Cre-ERT2 mice. RNA-sequencing analysis of Poly(I:C) [(polyinosinic: polycytidylic acid), a mimic of viral double-stranded RNA]-treated BMDMs revealed that 49 genes were induced more than two-fold by Poly(I:C) for 3 hrs in Hes1-deficient BMDMs relative to their expression in Hes1+/+Cre-ERT2 cells. Notably, 67.3% of these upregulated genes were ISGs by screening the Interferome database, suggesting that Hes1 inhibits expression of TLR-mediated ISGs,these results indicated that Hes1 negatively regulates TLR3-mediated induction of ISGs in macrophages.

Naïve and LCMV Cl13 infected liver tissue of ERT2 AlbCre Ifnar1 fl/fl mice

Project description:Age- and sex-matched (male, 2-3 months old) ERT2 AlbCre Ifnar fl/fl and ERT2 AlbCre Ifnar +/+ mice were injected intraperitoneally with 40 mg/kg tamoxifen for 5 consective days. Mice were then intravenously infected with 2x10^6 focus forming units of LCMV Cl13 and liver tissue of either genotype was harvested 1.5 days post infection and analyzed for transcriptomic changes (n = 3). Liver tissue of uninfected animals of both genotypes was harvested as control (n = 3).

Project description:Non-alcoholic fatty liver disease (NAFLD) is the most common chronic liver disorder in developed countries, with extremely limited therapeutic options. MYC is a highly pleiotropic transcription factor with broad biological functions. However, the effect of intestinal MYC on metabolic diseases has not been studied. Here, an unexpected role for intestinal MYC in NAFLD was uncovered. MYC was increased in ileum biopsies from obese people and positively correlated with body-mass index. Intestine-specific reduction of MYC in mice improved high-fat diet (HFD) -induced obesity and hepatic steatosis. Mechanistically, reduced expression of MYC in the intestine promoted glucagon-like peptide-1 (GLP-1) production and secretion via upregulating carbohydrate-responsive element binding protein (ChREBP) and solute carrier family 2, member 2 (GLUT2)/solute carrier family 5, member 1 (SGLT1), respectively. Furthermore, 10058-F4, a MYC inhibitor, had therapeutic effects on HFD-induced metabolic disorders, accompanied by increased GLP-1 and reduced ceramides in the serum. This study highlights intestinal MYC as a putative drug target against NAFLD.

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are to compare NGS-derived incisor transcriptome profiling (RNA-seq) between Runx2fl/fl and Gli1-CreERT2;Runx2fl/fl mice to identify the potential target of Runx2. Methods: Incisor mRNA profiles from one week after induction of one-month-old Runx2fl/fl and Gli1-CreERT2;Runx2fl/fl mice were generated by deep sequencing, in triplicate, using Illumina NextSeq500. Results: Using an optimized data analysis workflow, we mapped about 68 million sequence reads per sample to the mouse genome ( mm10) and identified 80,214 transcripts in th incisors of Runx2fl/fl and Gli1-CreERT2;Runx2fl/fl mice with Partek E/M workflow. Five hundred and eleven differentially regulated genes were identified (＞2-fold, p＜0.05), of which 299 were upregulated and 212 were downregulated.

Comparing the transcriptoms of Mettl14fl/fl and Mettl14 fl/fl; Olig2-Cre oligodendrocyre precursor cell (OPCs) and mature oligodendrocytes using Next Generation Sequencing

Project description:The goal of the experiment is to determine the transcriptome alternation of Mettl14 gene inactivation in different oligodendrocyte lineage cells(OPCs and mature oligodendrocytes), by comparing the Mettl14 fl/fl; Olig2-Cre(mutant) and Mettl14fl/fl(control) OPC and mature oligodendrocyte transcriptoms. Method: RNA collected from Mettl14fl/fl control and Mettl14fl/fl;Olig2-Cre OPC and oligodendrocytes was collected by standard protocols, sequenced using next generation high throughput sequencing technology, and processed in triplicate to produce a transcriptome profile deliminating the variable gene expression in the mutant cells. Results: We identified 11809 transcripts present in the OPC transcriptome, 586 were significantly upregulated and 177 were significantly downregulated in the mutant cells; Among 12,542 transcripts present in mature oligodendrocytes, 1388 transcripts were upregulated and 1247 were downregulated in the mutant cells(significance: q-value<0.001; fold changes (FC) that exceed 2.0 fold (log2 |FC|>1) in expression and an expression level that exceeds two counted transcripts per million (log2 |CPM| >1)) Conclusions: Mettl14 ablation differentially alters the OPC and oligodendrocyte transcriptomes
