By comparing the transcriptome of CPN_KO cells and the cells expressing CRY proteins (PN_KO), genes affected by CRY were analyzed. Parental cells expressing NR1D proteins (CP_KO) were also analyzed to identify genes regulated by NR1D proteins.

Project description:The circadian clock coordinates energy metabolism, with REVERBα considered a dominant regulator of lipid metabolism. REVERBα is a core clock protein, which demonstrates robust rhythmic expression across metabolic tissues. Here, we explore its role in white adipose metabolism, and propose that REVERBα plays a key part in the response of adipose to obesity. Mice globally lacking Reverbα (Reverbα-/-) display adiposity, increased susceptibility to diet-induced obesity, and up-regulation of the lipogenic proteome in white adipose depots. When Reverbα is deleted selectively in adipose (ReverbαFlox2-6AdipoCre), however, we observe only modest phenotypic and transcriptomic changes in the basal state, with altered regulation of clock and extracellular matrix (ECM) pathways. Thus the adipose-autonomous actions of REVERBα are shaped by context. On high-fat diet challenge, ReverbαFlox2-6AdipoCre mice accumulate more adipose mass than littermate controls, and are spared obesity-related inflammation. Gene expression analysis reveals pathways of lipid handling to be up-regulated. These findings therefore highlight a critical role for REVERBα in the regulation of adipose tissue expansion. Further, when transcriptomic data is integrated with the REVERBα cistrome, we detect strong associations of REVERBα binding sites, not only with the clock and ECM targets de-repressed under normal conditions, but with the broad range of metabolic genes unmasked as REVERBα targets by obesity. Thus our data favour a new interpretation of REVERBα function, not as conferring rhythmicity to lipogenesis, but as buffering against lipogenic cues asynchronous to the normal temporal routine.

Project description:Circadian rhythms are central to optimal physiological functioning and their interruption contributes to the development of several chronic diseases. Alcohol (EtOH) intoxication disrupts circadian rhythms within liver, brain, and intestines, but it is unknown whether alcohol also disrupts components of the core clock in skeletal muscle. Female C57BL/6Hsd mice were randomized to receive either saline (control) or alcohol (EtOH) (5g/kg) via intraperitoneal injection at the start of the dark cycle (ZT12), and gastrocnemius was collected every 4hr from Control and EtOH treated mice for the next 48hr following isoflurane anesthetization. In addition, metyrapone was administered prior to alcohol intoxication in separate mice to determine whether the alcohol-induced increase in serum corticosterone contributed to circadian gene regulation. Finally, synchronized C2C12 myotubes were treated with alcohol (100mM) to assess the influence of centrally or peripherally mediated effects of alcohol on the muscle clock. Alcohol significantly disrupted the mRNA expression of Bmal1, Per1/2, and Cry1/2 in addition to perturbing the clock-controlled genes, Myod1, Dbp, Tef, and Bhlhe40 (p<0.05). Alcohol increased serum corticosterone levels and glucocorticoid target genes, Redd1 and Klf15, in muscle. Metyrapone decreased serum corticosterone in EtOH mice but did not normalize the mRNA expression of Per1, Cry1 and Cry2 and Myod1 which were altered by EtOH. Alcohol did not alter core clock gene expression (Bmal, Per1/2, Cry1/2) at 4, 8, and 12hrs post treatment in synchronized C2C12 myotubes. Therefore, binge alcohol disrupted genes of the core molecular clock independently of elevated serum corticosterone.

Here, we analyzed circadian rhythms of urinary excretion of sodium and potassium in wild-type mice and mice lacking circadian transcriptional activator clock. Analysis of urines collected at hourly intervals over a 24-hour period revealed dramatic changes in rhythms of sodium and potassium excretion in clock(-/-) mice. In parallel, significant differences in circadian pattern of plasma aldosterone levels, but not in the 24-hour mean aldosterone levels, were observed. Microarray-based profiling of renal transcriptomes demonstrated that clock(-/-) mice exhibit dysregulation in multiple mechanisms involved in maintaining sodium and potassium balance by the kidney. The most significant changes were detected in the expression levels of several key enzymes (Cyp4a14, Cyp4a12a and Cyp4a12b) required for the conversion of arachidonic acid to 20-hydroxyeicosatetraenoic acid (20-HETE), a powerful regulator of renal sodium and potassium excretion, renal vascular tone and blood pressure. The 20-HETE levels measured in kidney microsomes of wild-type mice followed a circadian-like temporal pattern. In clock(-/-) mice, the acrophase of this rhythm was shifted by 8 hours and the 24-hour mean levels of 20-HETE were significantly decreased. These results demonstrate that circadian rhythms of urine electrolyte excretion are largely dependent on the circadian clock activity and indicate that circadian oscillations in renal 20-HETE content could be an important mechanism of blood pressure regulation. We examined the temporal profiles of gene expression in mouse whole kidney. Animals were sacrificed for microdissection every 4 hours, i.e. at ZT0, ZT4, ZT8, ZT12, ZT16 and ZT20 (ZT M-bM-^@M-^S Zeitgeber (circadian) time, indicates time of light-on as ZT0 and time of light-off as ZT12). The microarray hybridization was performed in duplicates on pools of RNA composed of equivalent amounts of RNA prepared from teo or three animals at each ZT time-point.
