Project description:metR (SPD_0588) mutant (TH9197) cannot grow in chemically defined medium (CDM) and had significant growth defect in CDM with 1 µg/ml methionine. With the increasing concentration of mehtionine, the growth of TH9197 was restored and the growth of metR mutant is as same as D39 WT (TH4306) when the concentration of methionine reaches 50 µg/ml. SPD_0588 is a putative transcriptional regulator and is homologous to MetR in Streptococcus mutans (78.9 % amino acid identity), which regulates both methionine synthesis and transport. We compared the transcriptome of WT and metR mutant in CDM with 1 µg/ml methionine by RNA-seq to see if MetR regulates the metabolism of methionine in Streptococcus pneumoniae D39. We compared the transcriptome of WT and metR mutant in CDM with 50 µg/ml methionine by RNA-seq to see if the regulation of MetR is methionine concentration-dependdent.We also want to identify the regulon of SPD_0588 in the two RNA-seq.

To verify our hypothesis, we firstly construct a strain with metR over-expression in D39 and with 3×flag tag and culture this strain in chemically defined medium (CDM) without methionine. Then collect bacteria for ChIP-seq to identify the genes that can be directly transcriptionally regulated by MetR.

Methionine starvation caused reprogrammed metabolism in Streptococcus pneumoniae

Project description:In our study, deletion of methionine synthesis gene metE (SPD_0510) impaired the growth of Streptococcus pneumoniae strain D39 significantly when bacteria were cultured in chemically defined medium with a low concentration of methionine. Bacteria undergo methionine starvation in this condition. We aimed to know how S. pneumoniae responses to methionine starvation, particularly at the transcriptional level. RNA-seq results show that 804 genes (41.2 % of whole genome) were differentially expressed under methionine starvation. Among them, 258 genes (32.1 %) are related to cell metabolism, with 75 genes involved in sugar and carbon metabolism, 88 genes for synthesis of metabolic products and 95 genes for uptake of amino acids and sugars. Combined with other experiments data, this showed the reprogrammed metabolism in S. pneumoniae under methionine starvation.

Methionine-mediated gene expression and characterization of the CmhR regulon in Streptococcus pneumoniae

Project description:In this study, we investigated the transcriptomic response of Streptococcus pneumoniae D39 to methionine. Transcriptome comparison of the S. pneumoniae D39 wild-type grown in chemically defined medium (CDM) with 0mM to 10mM methionine revealed the elevated expression of various genes/operons involved in methionine synthesis and transport (fhs, folD, gshT, metA, metB, metEF, metQ, tcyB, spd-0150, spd-0431, and spd-0618). We further demonstrated by β-galactosidase assays and quantitative RT-PCR studies that the transcriptional regulator, CmhR (SPD-0588) acts as a transcriptional activator of the fhs, folD, metB, metEF, metQ, and spd-0431 genes. We identified a putative regulatory site of CmhR in the promoter region of CmhR regulated genes and this CmhR site was further confirmed by promoter mutational experiments.

Comparison of the Streptococcus pneumoniae D39 wild-type to the D39 galR mutant [GalM17]

Project description:We explored the regulatory mechanism of Leloir pathway genes in Streptococcus pneumoniae D39. Here, we demonstrate that the expression of galKT is galactose dependent. By microarray analysis and quantitative RT-PCR, we further show the role of the transcriptional regulator GalR, present upstream of galKT, as a transcriptional activator of galKT in the presence of galactose. Moreover, we predict a 19-bp regulatory site (5'-GATAGTTTAGTAAAATTTT-3' ) for the transcriptional regulator GalR in the promoter region of galK, which is also highly conserved in other streptococci. Growth comparison of Î galK with the D39 wildtype strain grown in the presence of galactose shows that galK is required for the proper growth of S. pneumoniae on galactose.
