Project description:We injected LLC1 cells into tibia to construct a murine model to study bone microenvironment in cancer bone metastasis. We used single cell RNA sequencing (scRNA-seq) to analyze the cellular heterogeneity of tumor-infiltrating cells.
Project description:In this experiment we define the transcriptome of the adult mouse skeleton by performing total-RNA transcriptome-sequencing on osteocytes, the critical regulatory cells in bone. Osteocytes from 16 week-old male mice (n=8) were isolated for 4 bone types across the skeleton: the tibia, femur, humerus, and calvaria. All other soft tissues including marrow were removed from samples before sequencing.
Project description:We used single cell RNA sequencing (scRNA-seq) to identify femur and tibia osteoblast cell stress response pathway to misfolding of type I procollagen with Gly610->Cys substitution in the triple helical region of alpha2(I) chain.
Project description:Gli1+ progenitors are considered as metaphyseal mesenchymal progenitors in the distal femur and proximal tibia under the growth plate. We used single cell RNA sequencing (scRNA-seq) to analyze the diversity of Gli1+ progenitors in response to methylprednisolone.
Project description:LLC1 cells were injected into the tibia of DMP1-Cre PhAM mice to study the intercellular mitochondrial transfer in the bone microenvironment. We used single-cell RNA sequencing (scRNA-seq) to analyze the cellular heterogeneity of osteocyte-derived mitochondria recipient cells.
Project description:We used spatially resolved RNA sequencing (srRNA-seq) to identify trabecular osteoblast cell stress response pathway to misfolding of type I procollagen with Gly610->Cys substitution in the triple helical region of alpha2(I) chain (trabecular region of proximal tibia adjacent to the growth plate).
Project description:Affymetrix Chicken Gene 1.0 ST Array (ChiGene-1_1-st) profiles were generated from growth plate cartilage derived from the distal femur and proximal tibia of embryonic chickens at three developmental stages (day 12, 13 and 15) corresponding to stages prior to and immediately after the acqusition of mechano-responsive cartilage growth in the femur but not tibia (which does not respond to mechanical stimuli until later stages). Additionally, array profiles were generated from growth plate cartilage from the distal femur and proximal tibia of E15 chicks which were pharmacologically immobilized by decamethonium bromide treatment, which reduces longitudinal growth of the femur only at this stage.

Project description:To investigate the characteristics of PD-1+ LSCs in T-ALL, we purified PD-1+ and PD-1– T-ALL cells from the bone marrow (femur and tibia) of T-ALL mice, Pten null T-ALL mice, or PDX recipients. RNA sequencing was performed to explore the differential gene expression and pathway enrichment of PD-1+ LSCs in T-ALL.
Project description:Microarray analysis of white adipose tissue (WAT) and bone marrow adipose tissue (BMAT) from 22-week-old or 13-week-old male New Zealand White rabbits. From both cohorts, BMAT was sampled from the distal tibia (dBMAT) and the radius and ulna (ruBMAT), while WAT was sampled from the inguinal (iWAT) and gonadal (gWAT) depots. From the 13-week-old cohort, BMAT was also sampled from the proximal tibia (pBMAT). Sufficient RNA could not be isolated from all tissues for all rabbits, so for some rabbits only a subset of tissues is included.
