Project description:Purpose:We aimed to investigate the role of circulating exosomal microRNAs (e-miRNAs) in recurrent ischemic events in ICAD Methods: consecutive patients with severe ICAD undergoing intensive medical management (IMM) were prospectively enrolled. Those with recurrent ischemic events despite IMM during 6-month follow up were algorithmically matched to IMM responders. Baseline blood e-miRNA expression levels of the matched patients were measured using next generation sequencing Results: a total of 122 e-miRNAs were isolated from blood samples of 10 non-responders and 11 responders. Thirteen e-miRNAs predicted IMM failure with 90% sensitivity and 100% specificity. Ingenuity pathway analysis (IPA) determined 10 of the 13 e-miRNAs were significantly associated with angiogenesis-related biological functions (p < 0.025) and angiogenic factors that have been associated with recurrent ischemic events in ICAD. These e-miRNAs included miR-122-5p, miR-192-5p, miR-27b-3p, miR-16-5p, miR-486-5p, miR-30c-5p, miR-10b-5p, miR-10a-5p, miR-101-3p, and miR-24-3p. As predicted by IPA, the specific expression profiles of these 10 e-miRNAs in non-responders had a net result of inhibition of the angiogenesis-related functions and up expression of the antiangiogenic factors conclusion: This study revealed distinct expression profiles of circulating e-miRNAs in refractory ICAD, suggesting an antiangiogenic mechanism underlying IMM failure.

Project description:Moyamoya disease (MMD) and atherosclerosis (ICAD) are both common causes of chronic intracranial vascular occlusion. Surgical revascularization is the preferred procedure for MMD but is not recommended for ICAD, and may be related to vascular reserve capacity, however, the precise reason is not fully understood. Previous studies showed exosomes could regulate revascularization, so we hypothesize plasma exosomal proteins play critical roles in MMD during revascularization. Quantitative proteomics technique and bioinformatics method were used to identify potential key plasma exosomal proteins for revascularization in MMD.

Project description:Background: Gallstone disease (GSD) is one of the most prevalent and costly digestive system diseases in the world and can lead to pancreatitis, severe biliary tract infection and malignant biliary tract tumors. Gallstones can induce biliary colic, and the pain radiates to the shoulder and back. Severe attacks cause nausea and vomiting, which seriously affect the patient's work and life. Exosomes are important extracellular vesicles, and exosomal microRNAs (miRNAs) can be used as biomarkers and therapeutic targets for a variety of diseases. To the best of our knowledge, exosomal miRNA has not been well evaluated as a biomarker of the progression of cholelithiasis or the onset of biliary colic. Methods: Plasma samples from 10 patients with biliary colic attack were collected for exosome extraction, identification and miRNA sequencing. The exosome miRNA sequencing data of 10 healthy people in the GEO database were used as controls for data analysis. Results: Comparison of the plasma exosomal miRNAs of patients with biliary colic to those of healthy controls showed that 35 exosomal miRNAs were upregulated and 9 miRNAs were downregulated. GO and KEGG analyses showed that differential miRNAs are involved in the inflammatory response. The analysis of the subject operation characteristic curve shows that the expression of plasma exosomal miR-1268a can better reflect the attack characteristics of biliary colic than other miRNAs. miR-146a-3p participates in body inflammation and lipid metabolism processes and can be used as a potential therapeutic target. Conclusion: Plasma exosomal miRNAs can be used as biomarkers during the onset of biliary colic and can also be used as potential molecular targets to inhibit inflammation and analgesia.

Project description:To investigate the potential effect of grazing movement on miRNA circulation in cattle, here we profiled miRNA expression in centrifugally prepared exosomes from the plasma of both grazing and housed Japanese Shorthorn cattle. Microarray analysis of the c-miRNAs resulted in detection of a total of 231 bovine exosomal miRNAs in the plasma, with a constant expression level of let-7g across the duration and cattle groups. Expression of muscle-specific miRNAs such as miR-1, miR-133a, miR-206, miR-208a/b, and miR-499 were undetectable, suggesting the mildness of grazing movement as exercise. Changes in miRNA expression in plasma exosome of cattle was measured at during 0, 1, 2 and 4 months of grazing or housing. Plasma exosome samples prepared from three cattle for each treatment were collected and mixed within the treatment at each time for microarray analysis .

Project description:We conducted this study to determine whether exosome regulation underlies the antimigraine mechanisms of acupuncture. By comparing serum samples from patients with migraine and healthy controls using high-throughput small RNA sequencing technology , we identified 705 exosomal microRNAs that are differentially expressed in patients with migraine, and this set of 705 microRNAs included five that are particularly well characterised (hsa-miR-369-5p, hsa-miR-1268b, hsa-miR-145-5p, hsa-miR-222-5p, and hsa-miR-4488). By comparing serum samples collected from patients with migraine before and after acupuncture treatment, we showed that acupuncture normalised the expression levels of those five well-characterised exosomal microRNAs.

Project description:Purpose: This study aimed to explore the differential expression profiles of exosomal lncRNAs and evaluated their potential utility in the accurate diagnosis of LAA stroke. Methods: LncRNA profiles of exosomes in large artery atherosclerosis stroke and controls were generated by high-throughput sequencing. The sequence reads that passed quality filters were analyzed at the transcript isoform level with Hisat2, Trapnell, STAR. qRT–PCR validation was performed using TaqMan and SYBR Green assays. Results: A total of 1020 differentially expressed lncRNAs were identified in LAA stroke patients. GO and KEGG pathway analyses indicated that their target genes are involved in atherosclerosis-related pathways, including inflammation, cell adhesion, and cell migration. 8 exosomal lncRNAs were confirmed with qRT–PCR.The result showed that the expression trend of differential expressed lncRNAs in validation was consistent with RNA-seq.Conclusion: Our study showed the differential expression of lncRNAs in plasma exosomes and presented related diagnostic potential for LAA stroke for the first time. The results suggested that exosomal lncRNA could be potential diagnostic tools in LAA stroke. Circular RNAs (circRNAs), novel endogenous noncoding RNAs, play diverse roles in ischemic stroke. Recently, the abundance and stability of circRNAs in exosomes have been identified. However, a comprehensive analysis of exosomal circRNAs in large artery atherosclerotic (LAA) stroke has not yet been reported. We performed RNA sequencing (RNA-Seq) to comprehensively identify differentially expressed exosomal circRNAs in five paired LAA and normal controls. RNA-Seq identified a total of 462 circRNAs in peripheral exosomes; there were 25 differentially expressed circRNAs among them. Additionally, circRNA competing endogenous RNA (ceRNA) network and translatable analysis revealed the potential functions of the exosomal circRNAs in LAA progression. Two ceRNA pathways involving 5 circRNAs, 2 miRNAs, and 3 mRNAs were confirmed by qRT-PCR. In the validation cohort, receiver operating characteristic (ROC) curve analysis identified two circRNAs as possible novel biomarkers, and a logistic model combining two and four circRNAs increased the area under the curve compared with the individual circRNAs.
