The Microarray gene expression was conducted on NSCs and hipocampus from LPS depression induced depression mice (LPS group), LPS depression induced depression mice treated with luteolin(LPS+L group), Normal mice (PBS group) and Normal mice treated with luteolin (PBS+L).

Project description:This experiment aimed at characterising the modulatory role of murine induced Neural Stem Cells (iNSCs) and Neural Stem Cells (NSCs) on macrophages (MPs) exposed to LPS in vitro. NaÃ¯ve MPs were polarized into an M1-like phenotype with LPS, and then co-cultured with 1:1 ratios of iNSCs in a trans-well system that avoids cell-to-cell contacts. NaÃ¯ve MPs, LPS-stimulated MPs and LPS-stimulated MPs co-cultured with NSCs were used as controls.

Gene expression data from mouse ES cell-derived primitive NSCs and definitive NSCs

Project description:Primitive neural stem cells (NSCs) could be derived from pluripotent mouse embryonic stem (ES) cells, and then differentiate into definitive-type neural stem cells which resemble NSCs obtained from the central nervous system. Hence, primitive NSCs define an early stage of neural induction and provide a model to understand the mechanism that controls initial neural commitment. In this study, we performed microarray assay to analyze the global transcriptional profiles in mouse ES cell-derived primitive and definitive NSCs and to depict the molecular changes during the multi-staged neural differentiation process. Primitive NSCs derived directly from ESCs in Lif (p-NSC_L), primitive NSCs that were sub-cultured in the presence of Lif and FGF (p-NSC_LF), as well as definitive NSCs derived from primitive NSCs in medium containing FGF and EGF, were collected for RNA extraction and hybridization on Affymetrix microarrays. Mouse ESCs and NSCs obtained from mouse embryonic brain (E11.5) were included for controls. For each cell type, we collected two biological replicate samples for microarray analysis.

Project description:We used microarrays to assess gene expression differences between proliferating adult NSCs/neural progenitors with and without active SIRT1. Adult NSCs/neural progenitors were isolated from 8 week old Sirt1lox/lox and NestinCre;Sirt1lox/lox mice (129SV strain), cultured for one passage in growth factors (EGF and bFGF) before isolation at early passage 2 for RNA extraction and hybridization on Affymetrix microarrays. Gene expression data were adjusted for background and normalized using RMA.
