Project description:The objetive of this study was to evaluate the impact of acetate pre-treatment in alveolar macrophage like cells (MPI) stimulated with Streptococcus pneumoniae. We observed that acetate induce a high modulation of gene expression. Acetate led to similar numbers of up and down-regulated genes and in a enrichment analysis, acetate was shown to regulate different pathways, such as locomotion, cell proliferation and immune processes.

Project description:We report the application of RNA sequencing technology for high-throughput profiling in mouse bone marrow-derived macrophages pre-treated with MB-3(50μM) or DMSO, and stimulated with LPS(100ng/ml) for 4 hours. The goal of this study is to search differential gene in indicated groups. Through differential gene analysis, we can find out the specific mechanism of MB-3 inhibits LPS-induced pro-inflammatory gene transcription but promotes the expression of gene involved in NRF2 pathway and anti-oxidation acitivity.

Notch signaling is activated in LPS/IC-stimulated macrophages and inhibition of Notch signaling reduced IL-10 production. This study investigated the effect of gamma-secretase inhibitor (GSI) that suppresses Notch signaling pathway, on gene expression profiles in macrophages activated by LPS/IC.

Project description:The data is supplementary to the RNA-seq analysis of LPS and palmitate stimulation of THP-1 macrophages (E-MTAB-6064), where palmitate for the cell treatment was dissolved in sodium hydroxide and coupled with BSA at a molar ratio 7.5:1. Here we stimulated THP-1 macrophages with the corresponding concentration of BSA (4%) and NaOH (0.4 mM) in the presence of 10 nM phorbol 12-myristate 13-acetate (PMA) for 24 hours added to the standard RPMI 1640 medium with 10% fetal bovine serum (FBS) to estimate the effects and compared them with unstimulated THP-1 macrophages cultured in RPMI 1640 medium with 10% FBS and 10nM PMA.
