Atonal and Senseless perturbations in Drosophila melanogaster eye-antennal imaginal discs.

Project description:The goal of this study is to identify co-expressed genes downstream of Atonal and Senseless. These gene lists are used as candidate target genes (technically: as foreground sets) in computational predictions of cis-regulatory elements using the cisTargetX method (http://med.kuleuven.be/cme-mg/lng/cisTargetX). Together, the gene expression results and cis-regulatory predictions, yield a gene regulatory network underlying the early events in retinal differentiation. Predicted cis-regulatory interactions have been validated extensively in vivo using enhancer reporter assays and genetic perturbations. Six samples were analyzed for Atonal gain-of-function (GOF) in the eye-antennal imaginal discs of third instar larvae of Drosophila melanogaster, namely three biological repeats for two different Gal4 drivers (Gal4/7 and AtoGal), crossed with UASato. Three samples were analyzed for Atonal loss-of-function (LOF) in the eye-antennal imaginal discs, namely three biological repeats of ato-/- larvae. Three samples were analyzed for Senseless GOF, namely three biological repeats of atoGal4 crossed with UASsens. Finally, eight control samples were analyzed, namely two biological repeats of four different lines (atoGal4, Gal4/7, UASato, and cantonS wild type).

Sequencing of methylated GATC DNA fragments (Targeted DamID) from seven olfactory neuron populations in Drosophila melanogaster

Project description:This project’s aim was to compare the transcriptional profiles of olfactory sensory neurons in Drosophila melanogaster in order to identify novel genes that specify neuron-specific functions/phenotypes or may otherwise be involved in the development of the olfactory system. The isolation of sufficient numbers of intact olfactory sensory neurons (OSN) from the antenna of Drosophila melanogaster has so far limited single-cell transcriptomic approaches being applied to the adult fly antenna. Targeted DamID (TaDa) provides an alternative approach for profiling transcriptional activity in a cell-specific manor that bypasses the need for isolating OSN. Using the Gal4/UAS system, we applied TaDa to seven OSN populations and compared differences in Pol II occupancy for genes across these datasets.

Developmental hotspots drive transcriptional variability and convergence in the Drosophila olfactory system

Project description:The goal of this study was to identify the preponderance of developmental hotspots in the Drosophila olfactory system. Using RNASeq-derived transcriptome profiles of the developing and adult antennae for six different Drosophila species, we show that a few highly variable transcription factors may drive, likely in a combinatorial fashion, high variability in a few select olfactory receptor neuron (ORN) developmental lineages (i.e. developmental hotspots) in an otherwise mostly conserved background. Furthermore, the high variability of these few ORN lineages leads to a correspondingly high probability that they will recruited during the adaptation of the Drosophila olfactory system to specific ecological pressures, one potential example being the remarkably convergent antennal transcriptome profiles in two Drosophila species that have independently evolved the ability to specialize on specific host plants.

Project description:In this study we use Tag-sequencing in eye-antennal and wing imaginal discs across Drosophila species to determine a set of conserved eye-specific developmental genes. Next, we perform motif discovery analysis using the tool i-cisTarget, to depict the core gene developmental network underlying compound eye photoreceptor. The Glass position weight matrix appears as the most highly overrepresented motif, thus positioning Glass as a master regulator in compound eye photoreceptor development. Differential gene expression analysis by RNA-seq in D.melanogaster wild-type eye-antennal versus glass mutant [gl 60j] shows that the majority of our predicted Glass targets show strong downregulation in the glass mutant. Project description:Environmental temperature directly affects the concentration of chemicals in the gas phase. Therefore, if the olfactory system does not adapt to environmental conditions may provide inadequate information about distance to or direction of odor sources. Previous reports in Drosophila melanogaster show that temperature induces changes in olfactory sensitivity in complete individuals. Electrophysiological analysis demonstrates that these changes initiate at the olfactory receptor organs. In this experiment we try to identify particular genes responsible for olfactory adaptation to increasing temperatures using receptor organs of Drosophila after exposure to high temperatures. With this aim, wild-type Canton-S flies were subjected to 48-hour treatments at 30 degrees C. We use a direct approach selecting the specific tissue and making 'a priori' the gene election for the microarray analysis. Differential gene-expression analyses at the third antennal segments are performed comparing control and treated flies.
