Project description:To dissect molecular pathways regulated by LMO1 in neuroblastoma, we performed microarray gene expression profiling in a neuroblastoma cells (SHSY-5Y) after LMO1 knockdown.
Project description:The expression profiling of 245 known and 82 novel imsRNAs (intermediate-size noncoding RNAs, 50-500nt, which were identified from human fetal brain tissue) across different human fetal tissues (brain, heart, liver, lung and spleen at the 24-week gestation stage), different fetal brain gestational stages (13w, 14w, 20w, and 24w) and two human brain tumor cell lines (SHSY-5Y and U251).
Project description:The trace element manganese is essential for normal development for all the organisms. Overexposure of manganese may leads to multiple neuronal disorders such as Parkinson, manganism. To explore the molecular mechanism of manganese induced neurotoxicity, gene expression profiling was performed on human neuroblastoma SHSY-5Y cells. Cells were exposed to sub-lethal concentration of manganese (100 μM) for 24 hrs. Our result demonstrates that manganese alter multiple biological pathways including chromatin assembly, neurogenesis and apoptotic pathways.Cells grown in 75mm2 flask. three replicates for each sample

The analysis was based on the loop design method, in which all combinations of two samples were simultaneously hybridized to a microarray, each of the two sample labeled with a different fluorescent probe (Cy3 or Cy5).
Project description:ENCODE ChIP-chip study using 8 diverse human cell lines; 4 of neural origin (NB69, KELLY, SHSY-5Y, U373) and 4 of non-neural origin (HeLa, HepG2, HEK293 and K562) and anti-REST/NRSF antibody (Santa Cruz Biotech; sc-25398 aka H290) to asses REST-genome interactions across 1% of the human genome used in the pilot phase of the ENCODE project. Additionally the REST binding profile in K562 cells that had been transfected with REST specific siRNA oligos (72h and 96h) were determined revealing genomic in vivo binding affinity hierarchies for REST.
