Project description:Head and neck squamous cell carcinoma (HNSCC) is the tenth most common cancer worldwide. Alcohol and tobacco consumption are the predominant risk factors. Despite a multidisciplinary approach, the 5-year overall survival ranges only from 25% to 60%, depending on the site of origin. EVI1 expression increases with higher grading of HNSC tumors and over-expression was linked to a higher rate of lymph node metastasis. In lung cancer, its expression correlated negatively with patient survival. EVI1 was retrovirally over-expressed in CAL-33 and SCC-25 cells. Over-expression promoted proliferation, migration, and invasion of the cells. In CAL-33 cells, 1690 and 1684 genes were up- and down-regulated, respectively, at an FDR <0.1 in response to expression of EVI1. In SCC-25 cells, 334 significantly up- and 417 significantly down-regulated genes were identified. 71 and 181 genes were up- and down-regulated, respectively, in response to EVI1 expression in both cell lines. Gene ontology analysis revealed, among others, the terms epithelium development, extracellular matrix organisationorganization, cell adhesion, response to growth factor, and epithelial cell proliferation to be significantly enriched among the 252 genes that were regulated by EVI1 in a consistent manner in both cell lines.

CD98 marks a subpopulation of head and neck squamous cell carcinoma cells with stem cell properties

Project description:The application of compounds specifically targeting the cancer stem cells (CSCs), in addition to routinely used therapeutics, would likely improve the clinical outcome of head and neck squamous cell carcinoma (HNSCC). The previously described monoclonal antibody K984 (now found to recognize CD98) was used to identify, select and isolate a specific tumor subpopulation. These cells were molecularly characterized and subjected to serial transplantation experiments to determine the CSC properties. We show that CD98high cells, in contrast to CD98low cells, are able to generate tumors in immunodeficient mice and that the CD98high subpopulation expresses high levels of cell cycle control and DNA repair genes, while the CD98low fraction shows expression patterns that represent the more differentiated cells forming the bulk of the tumor. Transcriptional profiling of CD98high cells (n=4 fractions) of VU-SCC-OE xenograft tumors compared to CD98low cells (n=3 fractions) from the corresponding tumors.

Project description:The differential diagnosis between head & neck squamous cell carcinomas and lung squamous cell carcinomas is often unresolved because the histologic appearance of these two tumor types is similar. In the development of a gene expression profile test (GEP-HN-LS) that distinguishes these 2 cancer types, a collection of poorly differentiated primary and metastatic tumor specimens were used. Here we describe 76 such tumor specimens that were used for validation of GEP-HN-LS. The specimens are either head & neck squamous cell carcinomas or lung squamous cell carcinomas.
