Systems-level Responses to Salinity and Temperature Variations in the Marine Bacterium Vibrio natriegens

Project description:Vibrio natriegens strain ATCC 14048 is a salt marsh isolate with a notably rapid growth rate that has garnered considerable interest for biotechnological applications. We used systems-level tools (i.e. metabolomics, transcriptomics, proteomics) to characterize its physiological response to different salinities and temperatures that are relevant not only to its natural environment but also to planned scalable culturing processes. We found organic osmolyte synthesis and membrane transporters were most responsive to changes in salinity. The osmolytes glutamate, glutamine and ectoine responded to salinity across temperature treatments. However, when media was supplemented with choline, glycine betaine appeared to replace ectoine. These results provide a baseline data set for metabolic activity under a variety of conditions that will influence future basic and applied V. natriegens research.

Project description:Both high and low water temperature stresses are major environmental threats of growth and productivity in aquatic animals. However, the mechanism of response to temperature stress of scallop remains unclear. In this study, Argopecten irradians concentricus treated with high and cold temperature stress were analysed with the transcriptomes, metabolomes and integrated network analysis. Transcriptomic analyses revealed that there were many differentially expressed genes enriched calcium ion, kinase activity, phosphatase activity, and lipid related pathways between high temperature stress group and the control group. Although similar results were found in the low temperature stress group versus the control group, the genes enriched in these pathways were different. Moreover, much more genes were enriched in transcription factor activity in the low temperature stress group versus the control group

Influence of temperature fluctuations during cryopreservation on vital parameters, plasticity, and transgene expression of placental multipotent stromal cells

Project description:In this study we performed comprehensive analysis of changes in cell survival, vital parameters, plasticity, as well as transgene expression of placental MSCs after temperature fluctuations within the liquid nitrogen steam storage, mimicking long-term preservation in practical biobanking, transportation, and temporal storage. It was shown that viability and metabolic parameters of placental MSCs did not significantly differ after temperature fluctuations in the range from -196ºC to -100ºC in less than 20 cycles in comparison to constant temperature storage. However, increasing of the temperature range to -80ºC as well as increasing the number of cycles, leads to significant lowering of these parameters after thawing. The number of apoptotic changes increases depending on the number of cycles of temperature fluctuations. Besides, adhesive properties of the cells after thawing are significantly compromised in the samples subjected to temperature fluctuations during storage. Plasticity of placental MSCs was not compromised neither after cryopreservation with constant end temperatures nor with temperature fluctuations. However, regulation of various genes after cryopreservation procedures significantly varies. Alterations in structural and functional parameters of placental MSCs after long-term preservation should be considered in practical biobanking due to potential temperature fluctuations in samples. At the same time, plasticity and transgene expression are not compromised during studied storage conditions, while certain gene regulation is observed.

Project description:The cell cycle coordinates with growth to ensure suitable genome inheritance. The CsdA protein is a highly conserved, DEAD-box RNA helicase and assists RNA structural remodeling at low temperature. The absence of CsdA yields a severe growth defect manifested by increased doubling time, more extensive cell size, or slower colony growth at low temperature. Nevertheless, it is unknown how the lack of CsdA at low temperature affects the cell cycle and the causes of the yield of severe growth defect phenotypes. This work shows that the lack of CsdA results in incoordination of the cell cycle to growth at low temperature. We performed microarray analyses to compare the transcriptome profiles of ΔcsdA mutant with WT cell before and after rifampicin-treatment in LB to understand how the lack of CsdA at low temperature affects the cell cycle and the causes of the yield of severe growth defect phenotypes.
