Project description:CCD18co cells are representative of colonic human fibroblasts. We wanted to understand if there was a stimulus that could induce the cells to acquire the same pro-fibrotic signatures that are found in patients with Ulcerative Colitis or Crohn's Disease. We stimulated the cells with various pro-fibrotic stimuli, and analyzed the signatures by single cell RNA sequencing (scRNA-seq)
Project description:A focussed microarray chip was constructed specifically for studying basal colonic gene expression in patients with Spondyloarthropathy (SpA) and Crohn's disease (CD) (ref: PMID 16476712). These focus microarrays were used 1) to study common changes in gene expression between SpA and CD, providing early markers in the follow-up study of patients with SpA and particularly these patients that evolve into clinically overt CD; and 2) to identify new candidate genes for CD by integrating gene expression data with genetic information. Project description:The entire small intestine was observed by balloon endoscopy. Biopsy specimens were taken from jejunum, ileum and colon, respectively. Four Crohn's disease patients and four healthy controls were compared.
Project description:A feature of Crohn's disease is the extra-intestinal manifestation of creeping fat, defined as expansion of mesenteric adipose tissue around the inflamed and fibrotic ileum. In comparison to healthy controls, we found that the greatest transcriptional changes in creeping fat are functions related to immune response to bacterial products.

Project description:Objective: To evaluate gene expression profiles in patients with advanced cirrhosis compared with healthy controls (HC). Results: Patients with advanced cirrhosis exhibit an unbalanced systemic response involving pro-fibrotic / protective signatures that could be contributing to the pathogenesis of cirrhosis and also to that of the extra-hepatic complications of this disease.
