Project description:In the present study, we used RNA sequencing on mouse splenocytes to describe the immune recall in the days following pneumococcal lung infection. Mice were sacrificed at days 1, 2, 4, and 7 after Streptococcus pneumoniae (TIGR4 serotype 4) intranasal infection and splenocytes were cultured in the presence or absence of the same inactivated bacterial strain to access the transcriptomic profile.

Project description:The objective of this study was to compare recall responses to vaccine antigens at 3 months and 9 months of age in infants who were vaccinated at birth or at 1 month. PBMC were obtained at 9 months of age from infants who were vaccinated at birth (n=25) or 1 month (n=25). The PBMC were cultured in the presence or absence of CRM197 antigen for 72 hours. At the termination of the cultures, total RNA was extracted from the PBMC. The RNA was pooled into groups of n=5 subjects per group, following by labeling and hybridization to Affymetrix microarrays.

Trancriptional comparison of Zbtb32-deficient and -sufficient NP-specific memory B cells at day 7 of recall response [BMPC]

Project description:Memory B cell responses are more rapid and of greater magnitude than are primary antibody responses. The mechanisms by which these secondary responses are eventually attenuated remain unknown. We demonstrate that the transcription factor ZBTB32 limits the rapidity and duration of antibody recall responses. ZBTB32 is highly expressed by mouse and human memory B cells, but not by their naÃ¯ve counterparts. Zbtb32-/- mice mount normal primary antibody responses to T-dependent antigens. However, Zbtb32-/- memory B cell-mediated recall responses occur more rapidly and persist longer than do control responses. Microarray analyses demonstrate that Zbtb32-/- secondary bone marrow plasma cells display elevated expression of genes that promote cell cycle progression and mitochondrial function relative to wild-type controls. BrdU labeling and adoptive transfer experiments confirm more rapid production and a cell-intrinsic survival advantage of Zbtb32-/- secondary plasma cells relative to wild-type counterparts. ZBTB32 is therefore a novel negative regulator of antibody recall responses. CD45.2 wild type and Zbtb32-/- splenocytes from NP-CGG-immune donors were transferred into CD45.1 recipients and challenged with NP-CGG. CD45.2 donor NP-specific plasma cells B cells were isolated from the bone marrow 7 days later. 6 biological replicates of each genotype were performed.

Project description:The aim of this experiment was to investigate differential gene expression in splenocytes stimulated with BCG from naïve and BCG vaccinated mice. The differences between naïve and BCG vaccinated mice might indicate the mechanisms by which BCG vaccination confers an enhanced ability of splenocytes from BCG vaccinated mice to inhibit growth of BCG in splenocyte cultures as compared with splenocytes from naive animals. Splenocytes from 4 naïve and 4 BCG vaccinated female C57bl/6 mice were cultured for 12 hours with BCG (MOI 1:1) or without (unstimulated control). Each animal had a stimulated and an unstimulated sample hybridised to a beadchip.

Influenza-induced inflammation drives pneumococcal otitis media

Project description:Influenza A virus (IAV) predisposes individuals to secondary infections with the bacterium Streptococcus pneumoniae (the pneumococcus). Infections may manifest as pneumonia, sepsis, meningitis or otitis media (OM). It remains controversial as to whether secondary pneumococcal disease is due to the induction of an aberrant immune response or IAV induced immunosuppression. Moreover, as the majority of studies have been performed in the context of pneumococcal pneumonia, it remains unclear how far these findings can be extrapolated to other pneumococcal disease phenotypes. Here, we demonstrate that the viral hemagglutinin (HA) mediates bacterial OM by inducing a pro-inflammatory response in the middle ear cavity in a replication-dependent manner. Importantly, our findings show that it is the inflammatory response that mediates pneumococcal replication; not viral suppression of the immune system or epithelial damage. This study provide the first evidence that HA induced inflammation drives pneumococcal replication in the middle ear cavity, which has important consequences to the treatment of pneumococcal OM.

The analysis of gene expression profiles of splenocytes from wild type and UBL4A/GdX-/Y mice upon LPS stimulation

Project description:Purpose: To compared the gene expression profiles of splenocytes from wild type and UBLA4/GdX (Ubiquitin-like protein 4A, also named GdX) knockout mice upon LPS stimulation. Methods: GdX+/Y and GdX-/Y mice were injected with LPS. Their splenocytes were collected at different time (90min or 6 hrs), and then performed RNA-sequencing analysis. We sequence the samples used BGISEQ-500 platform. Results: The average mapping ratio with reference genome is 93.42%, the average mapping ratio with gene is 83.40%; A total of 17,920 genes were detected.A number of NF-κB targeting genes, such as IL-6, IL-1a and IL-12, have dramatic higher expressions in the splenocytes of GdX+/Y mice than that of GdX-/Y mice. Gene ontology (GO) analysis of GdX function in LPS-treated mice showed one of the most significantly enriched biological processes is related to inflammatory response. Conclusions: Our study revealed the gene expression profiles of the splenocytes from LPS-treated wild type and GdX-/Y mice.
