Project description:The zona pellucida plays a crucial role in the process of fertilization to early embryonic development, including cellular arrangement and communication between blastomeres. However, little is known regarding the role of the zona pellucida in pre- and post-implantation embryonic development associated with gene expression. We investigated the effects of the zona pellucida on the development and gene expression of pre- and post-implantation embryos. After the zona pellucida was removed at the 2-cell stage, compaction occurred earlier. In addition, the expression of differentiation-related genes in the inner cell mass (ICM) (Oct4, Sox21) and trophectoderm (TE) (Cdx2, Eomes, and Tfap2c) was significantly altered in both zona free (ZF) morula and blastocyst compared to zona intact embryos. After embryo transfer, the implantation rate and number of live fetuses were lower in ZF embryos than in control embryos, whereas the fetal weight at birth was not different. However, placental weight was significantly increased in ZF embryos. RNA-seq analysis of the placenta showed that a total of 473 differentially expressed genes that significantly influenced the biological process. The present study suggests that zona pellucida removal at the 2-cell stage not only disturbs the expression pattern of ICM/TE-related genes, but affects the post-implantation development of mouse embryos. Overall, this study provides a deeper insight into the role of the zona pellucida during early mouse embryonic development and the viability of post-implantation development. It also provides a valuable basis for future research related to the production of ZF embryos in mammals and the developmental properties of blastomeres.

Project description:Mechanisms of implantation such as determination of the attachment pole, fetal-maternal communication, as well as underlying causes of implantation failure are largely unexplored. Here we performed single-cell RNA sequencing (scRNA-seq) on peri-implantation embryos from both humans and mice to explore trophectoderm development and embryo–endometrium crosstalk. We found that the transcriptomes of polar and mural trophectoderm (TE) diverged after embryos hatched from the zona pellucida in both species, with polar TE being more mature than mural TE. The implantation poles show similarities in cell cycle activities, as well as in expression of genes critical for implantation and placentation. Embryos that either fail to attach in vitro or fail to implant in vivo, show abnormalities in pathways related to energy production, protein metabolism and 18S rRNA m6A methylation. These findings uncover the gene expression characteristics of humans and mice TE differentiation during the peri-implantation period and provide new insights into embryo implantation.

Project description:To determine the effect of Zika virus infection on pre-implantation embryonic development, we performed single blastocyst RNA-Seq on MOCK and ZIKV infected embryos. ZIKV infection results in an increased risk of spontaneous abortion and poor intrauterine growth although the mechanisms underlying fetal loss remain undetermined. Little is known about the impact of ZIKV infection during the earliest stages of pregnancy, or pre- and peri-implantation, because most current studies of ZIKV infection in pregnancy models focus on post-implantation stages. Here, we demonstrate that trophectoderm cells of pre-implantation human and mouse embryos can be efficiently infected with ZIKV, and that trophectoderm can propagate virus causing cell death of neural progenitors. These findings were corroborated by our demonstration that hESC-derived trophectoderm cells are infected by ZIKV in a dose dependent manner. RNAseq of single blastocysts revealed key transcriptional changes in cellular and physiologic functions upon ZIKV infection, including nervous system development and function, prior to commitment to the neural cell lineage. Finally, the pregnancy rate of mice infected pre-implantation was > 50% lower than females infected at E4.5. These results demonstrate that pre-implantation ZIKV infection of trophectoderm leads to miscarriage or spontaneous abortion. Moreover, pre- and peri-implantation ZIKV infects trophectoderm cells that propagate virus over time causing cell death in neural progenitors. Cumulatively, these data demonstrate that vertical pre- and peri-implantation ZIKV infection of trophectoderm impairs fetal development and causes neural progenitor cell death, elucidating a previously unappreciated association of pre- and peri-implantation ZIKV infection and microcephaly.

Project description:Developmental failures occurring shortly after blastocyst hatching from the zona pellucida constitute a major cause of pregnancy losses in both humans and farm ungulates. The developmental events occurring following hatching in ungulates include the proliferation and maturation of extra-embryonic membranes –trophoblast and hypoblast- and the formation of a flat embryonic disc, similar to that found in humans, that initiates gastrulation prior to implantation. Unfortunately, our understanding of these key processes for embryo survival is limited because current culture systems are unable to sustain ungulate embryo development beyond hatching. Herein, we report a culture system that recapitulates most developmental landmarks that characterize ovine development up to gastrulating stages: trophoblast maturation, hypoblast migration, embryonic disc formation, Rauber´s layer removal, epiblast polarization and mesoderm differentiation. In this dataset we compare the transcriptional profile of ovine day (D) 14 in vitro cultured embryos and embryonic day (E) 11 and 12.5 in vivo derived embryos by RNA-seq.

Human ES cells are different than mouse ES cells and share defining features with EpiSCs, yet are derived from pre-implantation human embryos. Here we show that EpiSCs can be routinely derived from pre-implantation mouse embryos. The pre-implantation-derived EpiSCs exhibit molecular features and functional properties consistent with bona fide EpiSCs. These results provide a simple method for isolating EpiSCs and offer direct insight into the intrinsic and extrinsic mechanisms that regulate the acquisition of distinct pluripotent states.
