Project description:Recent studies suggested that microglia, the primary brain immune cells, can affect circuit connectivity and neuronal function. Microglia infiltrate the neuroepithelium early in embryonic development and are maintained in the brain throughout adulthood. Several maternal environmental factors, such as aberrant microbiome, immune activation, and poor nutrition, can influence prenatal brain development. Nevertheless, it is unknown how changes in the prenatal environment instruct the developmental trajectory of infiltrating microglia, which in turn affect brain development and function. Here we show that after maternal immune activation (MIA) microglia from the offspring have a long-lived decrease in immune reactivity (blunting) across the developmental trajectory. The blunted immune response was concomitant with changes in the chromatin accessibility and reduced transcription factor occupancy of the open chromatin. Prenatal replacement of MIA microglia with physiological infiltration of naïve microglia ameliorated the immune blunting and restored a decrease in presynaptic vesicle release probability onto dopamine receptor type-two medium spiny neurons, indicating that aberrantly formed microglia due to an adverse prenatal environment impacts the long-term microglia reactivity and proper striatal circuit development.

Project description:Dysfunctional humoral and cellular innate immunity are key components in the development and progression of age-related macular degeneration (AMD). Specifically, chronically activated microglia and their disturbed regulatory system contribute to retinal degeneration. Galectin-3, a b-galactose binding protein, is a potent driver of macrophage and microglia activation and has been implicated in neuroinflammation, including neurodegenerative diseases of the brain. Here, we report that galectin-3 is strongly upregulated in reactive retinal microglia of AMD patients and in two related mouse models of retinal degeneration. Specific targeting of galectin-3 by genetic knockout or using the small-molecule inhibitor TD139 blocks microglia reactivity and protects from retinal damage in different models of light-induced retinal degeneration. These data define galectin-3 as potent driver of retinal degeneration and highlight the protein as a drug target for ocular immunomodulatory therapies.

Project description:Neurofibromatosis type 1 (NF1) is autosomal dominant condition caused by germline mutations in the NF1 gene on chromosome 17. Children with NF1 are prone to the development of multiple nervous system abnormalities, including autism and brain tumors, which could result from the effect of NF1 mutation on microglia function. Using Nf1-mutant mice, we previously identified impaired microglia process extension and phagocytosis resulting from defects in purinergic signaling. To determine whether these abnormalities are also observed in human microglia, we leveraged human induced pluripotent stem cell engineering to generate human microglia-like (hiMGL) cells harboring three different NF1 patient-derived NF1 gene mutations. Both NF1-mutant and isogenic control hiMGL cells express classical microglial markers and exhibit similar transcriptomes and cytokine/chemokine release profiles, NF1-mutant hiMGL cells have defects in P2X receptor activation, phagocytosis and motility. We conclude that NF1 mutation impairs some aspects of microglia function in humans, which could contribute to a subset of the neurological abnormalities seen in children with NF1.
