Murine islets identified by TMT-labeled phosphoproteome after short time stimulation with neurotransmitters ACh and NE

Project description:Norepinephrine (NE) and acetylcholine (ACh) are important neurotransmitters of sympathetic and parasympathetic nerves, which play antagonistic roles in regulating different islet hormones secretion. Phosphorylation is reported as a critical post-translational modification participates in neural regulation on various physiological activities in islet. However, the molecular mechanisms of these two transmitters in islet are unknown and whether the neuronal regulation is abnormal in diabetes mellitus also unclear. Quantitative phosphoproteome analysis were performed on murine islet after short time stimulation with neurotransmitters NE and ACh to reveal the key moleculars and phosphorylated proteins in islet which participate in the innervation of nerve signaling.

Transcriptomes Analysis of LHX2-knockdown in KYSE30 and KYSE510 cells

Project description:Purpose: To fully realize the potential molecular mechanism that LHX2 promotes ESCC progression Methods: Total RNA of LHX2-knockdown KYSE30/KYSE510 and control cells was extracted with TRIzol Reagent. RNA libraries were constructed using an Illumina TruSeq RNA Sample Preparation kit according to the manufacturer’s protocol. A total of 150 base paired-end reads were sequenced using the Novaseq 6000 S4 platform. Results: We identified 26008 transcripts in KYSE30 control and KYSE30 LHX2-knockdown cells ,and 25561 transcripts in KYSE510 control and KYSE510 LHX2-knockdown cells. Conclusions: Our study represents the analysis of LHX2-knockdown ESCC cells, generated by RNA-seq.

Non-neuronal acetylcholine as an endogenous regulator of proliferation and differentiation of Lgr5-positive stem cells in mice

Project description:Acetylcholine (ACh) has been considered a neurotransmitter residing in central, parasympathetic and neuromuscular synapses of mammals. Here, experiments using crypt-villus organoids that lack nerve and immune cells in culture led us to suggest that endogenous ACh is synthesized in the intestinal epithelium to evoke growth and differentiation of the organoids through activation of muscarinic ACh receptors (mAChRs). The extracts of the cultured organoids exhibit a noticeable capacity for ACh synthesis that is sensitive to a potent inhibitor of choline acetyltransferase (ChAT). Imaging mass spectrometry reveals distribution of endogenous ACh that is localized in intestinal epithelial layer in the cultured organoids as well as in mouse small intestinal epithelium in vivo, suggesting non-neural resources of ACh. Treatment of organoids with carbachol down-regulates growth of organoids and expression of marker gene for each epithelial cell. On the other hand, antagonists for mAChRs enhances growth and differentiation of organoids, indicating involvement of mAChRs in regulating proliferation and differentiation of Lgr5-positive stem cells. Collectively, our data provide evidence that endogenous ACh released from intestinal epithelium maintains homeostasis of intestinal epithelial cell growth and differentiation via mAChRs in mice. RNA sequencing analysis of OTUD6B-knockdown KYSE30 and KYSE450 cells

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of cellular pathways. The goals of this study are using RNA sequencing analysis to test the effects of OTUD6B-knockdown on the mRNA of KYSE30 and KYSE450 cells. Methods: KYSE30 and KYSE450 cells were transfected with negative control or siOTUD6B for 48 hours in 1640 medium plus 10% serum. Three independent replicates were plated, transfected in parallel for each negative control and siOTUD6B. Results: Log-fold changes of mRNAs between negative control and siOTUD6B group were selected with a significance threshod of p<0.05. Conclusions: Our study determined the mRNA changes of OTUD6B-knockdown KYSE30 and KYSE450 cells.
