Project description:Illumina HiSeq technology was used to generate mRNA profiles from Populus tremula x alba INRA 717-1B4 roots treated with Methyl jasmonate. Samples were harvested after two weeks either from untreated control roots or from Methyl jasmonate treated roots. Paired-end (2X100bp) reads were generated and aligned to the Populus trichocarpa (http://www.phytozome.net/poplar.php) using CLC Genomics Workbench 6. mRNA profiles from Populus tremula x alba INRA 717-1B4 roots treated with Methyl jasmonate as well as from control roots were generated by paired-end (2X100bp) Illumina HiSeq sequencing. Four samples were sequenced per lane, two biological replicates per treatment.

Transcription profiling of Arabidopsis Col-4 plants exposed to methyl jasmonate

Project description:As part of the CAGE consortium, Col-4 plants (1.04, 1.08, 1.12 or 5.10 stage) were exposed to a methyl jasmonate stimulus (or to the ethanol as a control) and leaves 1 and 2 were harvested. Hybridizations were performed using the oligo reference labeled with Cy3. The stress samples can be analyzed together with the basal/normal genomic expression patterns for Col-4 wildtype plants collectively produced by CAGE members.

Jasmonate-responsive genes in rice

Project description:The plant hormone jasmonate (JA) plays important roles in the regulation of defense responses in many plants. To clarify the response to JA in rice at gene expression level, we performed a microarray analysis using the Agilent Rice Oligo Microarray (44k, custom-made; Agilent Technologies, Redwood City, CA, USA). As a result, treatment of JA caused high upregulation of many defense-related genes including pathogenesis-related (PR) genes in rice. However, many of these defense-related genes were not upregulated in JA-insensitive transgenic rice plant overexpressing JAZ8deltaC.

SWATH-MS Proteomics Analysis of Persicaria minor herb upon Methyl Jasmonate elicitation

Project description:Stresses from either biotic or abiotic origins can have significant impact towards plant physiology and molecular regulation. Jasmonate acid (JA) and its derivative, methyl JA (MeJA) are hormonal cues released by plants which signal defensive response to curb the damage from such stresses. In an attempt to study the defensive response, a tropical herbal plant, Persicaria minor (P. minor) which is known for its pungent smell as well as various bioactivities including antimicrobial and anti-cancer, has been treated with MeJA to invoke the stress signaling. Such elicitation has been performed in various plants such as Arabidopsis, rice and hairy root cultures of certain herbs, yet how MeJA directly influenced the proteome of a herbal species particularly P. minor has not been previously elucidated. In this study, P. minor plants was exogenously treated with MeJA and its proteome was investigated using a new proteomics approach called SWATH-MS.

Response of aspen cell cultures fed piperonylic acid to methyl jasmonate.

Project description:To better understand the role of phenylpropanoid pathway perturbation in plant metabolism, Populus tremuloides cell cultures previously established from leaf mesophyll tissue were fed with piperonylic acid (PIP) alone, or in combination with methyl jasmonate (MJ_PIP) approximately 5 days after transfer to fresh medium. Methyl jasmonate is an elicitor that activates a suite of defense responses including phenylpropanoid metabolism, while PIP acts as an inhibitor of cinnamate-4-hydroxylase (C4H). Sample tissues were contemporary with those used to generate Series GSE16773. Samples were harvested 48 hrs after initiation of the experiment. Total RNA was extracted and gene expression measured using Affymetrix poplar genome microarrays.
