Angiotensin I and II in Human Plasma and Serum and Mouse Plasma

ABSTRACT: Fast turn around monitoring of Angiotensin I and II in blood. Plasma and Serum samples were digested with an HTA Protease in 20 minutes, spiked with SIL Angiotensin I and II and analyzed by targeted LC-MS. This preliminary data demonstrates that Angiotensin I and II can be rapidly measured and multiplexed with several proteins using the methodology. This methodology allows an unprecedent ability to monitor circulating biomarkers in almost real-time.

Neurotoxin tetrodotoxin TTX withdrawal induced neuronal activity: RNA Pol II is poised for rapid induction of arc and other neuronal VF-IEGs

Project description:Transcription of immediate early genes (IEGs) in neurons is exquisitely sensitive to neuronal activity, but the mechanism underlying the earliest of these transcription events is largely unknown. Here we demonstrate that very fast IEGs (VF-IEGs) such as arc/arg3.1 are poised for rapid transcription by the stalling of RNA Polymerase II (Pol II) just downstream of the transcription start site. RNAi-depletion of two subunits of a mediator of Pol II stalling, Negative Elongation Factor, reduces Pol II occupancy of the arc promoter and compromises rapid induction of arc and other VF-IEGs. In contrast, reduction of Pol II stalling did not prevent expression of other fast IEGs (F-IEGs). These F-IEGs are expressed with comparatively slower kinetics and largely lack promoter proximal Pol II stalling. Taken together, our data strongly indicate that very fast kinetics of neuronal IEG expression require poised Pol II and suggest a role for this mechanism in transcription-dependent learning and memory. TTX withdrawal induced neuronal activity. To study activity-induced gene expression, neurons were treated with TTX for 48 hours and then TTX was washed out either for 15 minutes (W15) or for 45 minutes (W45). Gene expression was measured in these two groups in comparison to TTX treated neurons.

Project description:Angiotensin II (Ang-II) regulates adrenal steroid production and gene transcription through several signaling pathways. Changes in gene transcription occur within minutes after Ang-II stimulation, causing an acute increase in aldosterone production and subsequent increase in the overall capacity to produce aldosterone. Our goal was to compare the Ang-II regulation of early gene expression and confirm the upregulation of selected genes using quantitative real-time RT-PCR (qPCR) across three species: human, bovine, and rat. Keywords: ChIP-chip There are 4 different strains: rap1-sil (without the silencing domain), RAP1 (both from Graham I.R. et al 1999) and tpk1 & tpk2 mutants (Euroscarf). Rate of elongation by RNA polymerase II is influenced by specific gene features and histone modifications

Project description:The rate of transcription elongation plays important roles in the timing of expression of full-length transcripts as well as for the regulation of alternative splicing. In this study we coupled Bru-Seq technology with 5,6-dichlorobenzimidazole 1-M-NM-2-D-ribofuranoside (DRB) to estimate the elongation rates of over 2,000 individual genes in human cells. This technique, BruDRB-Seq, revealed gene-specific differences in elongation rates with a median rate of around 1.5 kb/min. We found that genes with fast elongation rates showed higher densities of H3K79m2 and H4K20me1 marks compared to slower elongating genes. Furthermore, fast elongation rates had a positive correlation with gene length, low complexity DNA sequence and distance from nearest active transcription unit. Features that negatively correlated with elongation rate included exon density and the number of LINE sequences in the gene. The BruDRB-Seq technique offers new opportunities to interrogate mechanisms of regulation of transcription elongation. Measurement of RNA Pol II elogation rate. Normal fibroblasts (HF1 and TM), Cockayne syndrome group B fibroblasts, K562 and MCF-7 cells were exposed to DRB for 60 minutes, after which a washout was performed. Nascent RNA was labeled using bromouridine for 10 minutes immediately after the washout. The genomic region extending from actice Trancription Start Sites was used to determine the gene's elongation rate. Comparison of miRNA profiles between young (6 months) and old (24 months) male mouse vertebral samples

HCV dynamics and cellular gene expression in uPA-SCID chimeric mice with humanized livers during intravenous silibinin monotherapy

Project description:Background/Aim: Legalon SIL (SIL) is a chemically hydrophilized version of silibinin that has exhibited hepatoprotective and antiviral effectiveness against hepatitis C virus (HCV) in patients leading to viral clearance in combination with ribavirin. To elucidate the incompletely understood mode of action of SIL against HCV, we studied viral response kinetics and cellular gene expression during SIL treatment in the absence of an adaptive immune response in uPA-SCID chimeric mice with humanized livers. Methods: Chronically HCV-infected mice were treated with daily intravenous SIL at 469 mg/kg (n=5), 265 mg/kg (n=5) or 61.5 mg/kg (n=5). Serum HCV and human albumin (hAlb) were measured frequently and liver HCV RNA was analyzed at days 3 and 14. Microarray analysis of human hepatocyte gene expression was performed at days 0, 3, and 14 of treatment. Mathematical modeling was used to estimate viral kinetic parameters and SIL effectiveness. Results: While hAlb remained constant, a biphasic viral decline in serum was observed consisting of a rapid 1st phase followed by a 2nd slower (or plateau with the two lower SIL dosing) phase. SIL effectiveness in blocking viral production was similar among dosing groups (median =77%). However, the rate of HCV-infected hepatocyte decline, δ, was dose-dependent. Intracellular HCV RNA levels correlated (r=0.66, P=.01) with serum HCV RNA. Pathway analysis revealed increased anti-inflammatory and anti-proliferative gene expression in human hepatocytes in SIL-treated mice. Conclusions: The results suggest that SIL could lead to a continuous 2nd phase viral decline, i.e., potentially viral clearance, in the absence of adaptive immune response along with increased anti-inflammatory and anti-proliferative gene expression in human hepatocytes.
