Project description:This experiment was a 7-part time series (or, 2x7 factorial design) examining the effect of rapamycin on gene expression in Saccharomyces cerevisiae. Treated and untreated cultures were compared at 0, 20, 40, 60, 120, 240, and 360 minutes. In the paper, expression ratios (mRNA) at each time point were compared to equivalent ratios (protein) from shotgun proteomics NSAF data. High-expressing mRNA ratios were also clustered and GO-characterized.

FIA plasma induces arthritis in naÃ¯ve mice

Project description:Plasma collected from mice with FIA were pooled, and 0.3 ml was injected intravenously into 6-week-old naÃ¯ve SJL mice on days 0 and 2. Synovial antigen array profiling of plasma from the arthritic recipient mice demonstrated autoreactive B-cell responses against peptides representing native fibrinogen and citrullinated fibrinogen, and further epitope spreading resulting in additional targeting of fibronectin, collagen type V, cartilage gp39, and clusterin. Custom-spotted protein slides were probed with plasma samples from individual mice. Four slides were probed with plasma derived from naÃ¯ve mice and four slides were probed with plasma derived from mice injected with FIA plasma.

Gene expression profiling of flower and leaf of Cold-tolerant rice variety Stejaree45

Project description:To stabilize crop yield under low temperature stress conditions, it is important to improve stress tolerance in crops. Upon exposure to low temperature stress, many genes are induced and their products are thought to function as cellular protectants of stress-induced damages Therefore, responses of global gene expression profiles to cold stress was analyzed at the booting stage using the 60K Rice Whole Genome Microarray. The cold-tolerant rice variety, Stejaree45 was transplanted in 1/5000 pot at 20 days after seeding. Fertilizers were applied at the rate of 11, 7.7 and 9.3 kg/10a in N, P2O5 The samples were selected at the booting stage of rice on the basis of the auricle distance (the distance between auricles of flag leaf and the previous leaf of the main culm). The plants with an auricle distance ranging from -5 ㎝ to 0 ㎝ were exposed up to the flag leaf auricle using cold water (13℃) for 3 days. Glumous flowers and flag leaves of the cold-resistant variety, Stejaree45, were used to examine gene expression profiles.
