Project description:Purpose: The aim of this study was to compare transcriptome profiling (RNA-seq) of rice panicle tissue from resistant and suceptible lines at different time intervals infected with M.oryzae. The results were validated using Semi-quantitative PCR. Methods: Panicles from resistant and suceptible rice lines were inoculated with suspension having double distill water, Tween-20 without spores for 0 hour and same suspension with spores was used to inoculate for 48, 72 and 96 hour samples. After collection, samples were frozen on liquid nitrogen, and RNA was extracted using Spectrum™ Plant Total RNA Kit (Sigma). RNA libraries were prepared for sequencing using standard Illumina protocols. Results: Differential Expressed Genes were identified, comparing transcriptome of Control 0 hour vs 48, 72 and 96 hour post infection. Where we get 1070, 2383 and 1080 total significant genes at 48, 72, and 96 hour post infection respectively in HP2216. And, 1564, 1433 and 2263 total significant genes at 48, 72, and 96 hour post infection respectively in Tetep.

Project description:Expression profiles of one-week-old rice seedlings exposed to Pi-deficient solution for 6, 24, 48, 72 hours were monitored by microarray that contains approximately 60,000 rice clones (Affymetrix GeneChip). The probes were prepared from RNAs isolated from rice seedlings exposed to Pi-deficient solution for 6, 24, 48, 72 hours, respectively, and those non-treated controls. For hybridization, two biological replicates were used to extract RNAs from different batches of plants. Rice seedlings were exposed to low phosphate for 6h, 24h, 48h and 72 h, respectively. The Pi-inducible and Pi-repressive genes were taken as the expression ratio (treatment relative to control) greater and less than 2-fold, respectively

Project description:Epidermis is essential for animal survival, providing both a protective barrier and cellular sensor to the external environment. Interestingly, the epidermes of different species show broad morphological and functional diversity yet it is unclear whether this diversity came from modification of an existing gene regulatory network or de novo innovation of new genes. Here we identify the transcriptional regulators underlying the differentiation program of planarian epidermal lineage. We classify Smed-p53 as the most upstream molecule in this transcriptional cascade, suggesting a potentially conserved role for this gene in epidermal differentiation similar to TP63 in vertebrates. Moreover, we find that homologs of Sox and Pax family transcription factors, Smed-soxP-3 and Smed-pax-5, act cooperatively to activate the expression of epidermal markers. Together, these data show that planarian epidermal differentiation is regulated by a combination of conserved elements (p53/p63), recruitment of a non-conventional transcription module (soxP-3/pax-5), and novel genes (prog); they also suggest that specialized adpatations, such as epidermal mucus-secretion, arise from complex changes to gene networks. The purpose of this experiment is to identify genes associated with neoblasts (stem cells). Worms are exposed to 6k rad of gamma radiation, allowed to recover for 24, 48, 72, or 96 hours, and compared to the pre-cursor un-exposed population by RNA Seq to identify genes with decreased expression following radiation exposure.

The global gene-expression profiles of LD13 flow-cell biofilm were compared after 24, 48, 72, 96, and 144 hr, respectively, as well as with those of LD13 planktonic cultures.
