NDUFAF3 is involved in the assembly of the Q/P module of respiratory complex I in the green microalga Chlamydomonas reinhardtii

Project description:The mitochondrial NADH:ubiquinone oxidoreductase, or complex I, is composed of a hydrophobic arm comprising the P module and a hydrophilic arm comprising the N and Q modules. The assembly of complex I is well characterized in humans and catalyzed by a series of assembly factors that join the Q, P, and N modules sequentially. The complex I of protists and plants contains additional ancestral features, like the ferredoxin bridge that connects the matrix and the membrane arms and the carbonic anhydrase domain, which raises the question of their assembly. In this work, a strain where the assembly factor NDUFAF3 has been tagged with a 3xFLAG at the C-terminal extremity has been investigated in the green microalga Chlamydomonas reinhardtii. Like its human homolog, NDUFAF3 interacts strongly with the classical subunits of the Q and P modules, but also with the carbonic anhydrase domain and C1-FDX, one of the subunits of the ferredoxin bridge. The predicted structural positioning of NDUFAF3 within the Q module suggests a role in the formation of the ferredoxin bridge. In contrast, subunits of the N module are only loosely associated with NDUFAF3. We further demonstrate that the N module is attached at a later stage of assembly, suggesting that Chlamydomonas complex I assembles in a human-like pathway. This contrasts with the situation in Angiosperms where the N and the Q modules are attached together before anchoring to the P module. Altogether, these results highlight a conserved and ancestral role of NDUFAF3 in complex I manufacture.

Loqs2 (AAEL013721) interactome in Aedes aegypti Aag2 cells (FLAG-tag)

Project description:Loquacious-2 (Loqs2) is a recently identified paralog of Loquacious found in Aedes aegypti. Data presented here allowed the proteomic characterization of Loqs2 interactome in Aedes aegypti Aag2 cells. We carried out immunoprecipitation experiments where Loqs2 tagged protein was precipitated along with its partners. Immunoprecipitation of tagged eGFP protein was performed in the same conditions to verify the specificity of these associations. Experiments were performed in biological triplicates.

Targeting chaperone-mediated autophagy inhibits properties of glioblastoma stem cells and restores anti-tumor immunity

Project description:Chaperone-mediated autophagy (CMA), a selective autophagic process, plays a crucial role in maintaining intracellular quality and facilitating cellular responses to stress. While CMA has been implicated in finely regulating the stemness of normal stem cells, its significance in cancer stem cells (CSCs) remains largely undefined. In this study, we demonstrate that CMA is upregulated in patient-derived glioblastoma stem cells (GSCs), characterized by a notable increase in the levels of the CMA receptor, lysosome-associated membrane protein type 2A (LAMP2A). The enhanced stability of LAMP2A within GSC lysosomes is a result of MST4-mediated phosphorylation at residues T40, S97, and T136, which facilitates LAMP2A homotrimer formation and diminishes LAMP2A degradation by obstructing the interaction between LAMP2A and Cathepsin A. The contribution of CMA to the self-renewal and tumorigenic potential of GSCs is mediated by the upregulation of mTORC1 activity through the selective lysosomal degradation of its two negative modulators, TSC1 and TSC2. Furthermore, CMA is implicated in promoting resistance to immunotherapy in GBM by inhibiting the TET3-dependent enhancement of tumor cGAS and STING expression. Finally, the genetic or pharmacological inhibition of CMA exhibits a synergistic effect with immune checkpoint therapy in murine models of GBM. Our findings delineate an MST4-LAMP2A signaling pathway that modulates CMA activity and the malignancy of GSCs, with therapeutic targeting of this axis potentially overcoming immunotherapy resistance in GBM.

Splitting Function enables Dual Feedback Regulation to Control JAK2/STAT5 Signaling for a Wide Ligand Range

Project description:Cellular signal transduction is governed by multiple feedback mechanisms to elicit robust cellular decisions. We combined mathematical modeling and extensive time-resolved data sets in primary erythroid progenitor cells and dissected the roles of the two transcriptional feedback regulators of the SOCS family, CIS and SOCS3 in JAK2/STAT5 signaling. Our model revealed that both feedbacks are most effective at different ligand concentration ranges. To identify the relevant transcriptional feedback regulators that are involved in attenuation of Epo-induced JAK2-STAT5 signaling in addition to CIS, we performed a time-resolved genome-wide expression profiling of murine erythroid progenitor cells at the colony forming unit erythroid (CFU-E) stage up to 24 and 7 hours after EPO stimulation and in control, repectively. The analysis identified SOCS3 as the only further de novo regulated gene upon Epo-induced JAK2-STAT5 signaling. From subsequent mathematical modeling, we calculated the STAT5 response in previously unobserved Epo concentrations, which provided a quantitative link between cell survival and the integrated response of STAT5 in the nucleus. In conclusion, our combined modeling approach revealed novel insights into the orchestrated action of feedback control to regulate STAT5-mediated survival decisions over a broad range of ligand concentrations. Freshly sorted CFU-E cells were starved for 1 h in Panserin 401 supplemented with BSA and 50 µM β-Mercaptoethanol. Subsequently, cells were stimulated with 0.5 U/ml Epo or left untreated and RNA was extracted at different time points (0,1,2,3,4,5,6,7,8,14,19,24 hrs after Epo stimulation and at 0,1,2,3,4,5,6,7 hrs without Epo treatment) using the RNeasy Mini Plus Kit (Qiagen, Hilden, Germany).
