The insufficient activation of RIG-I-like signaling pathway in IBRS-2 cells contributes to highly efficient replication of porcine picornaviruses

Project description:Seneca Valley Virus (SVV) or commonly known as Senecavirus A (SVA), is one of picornavirus that is associated with vesicular disease and neonatal mortality in swine herds. However, the pathogenesis of SVV remains largely elusive. Our previous study found that SVV replicates extremely faster in porcine IBRS-2 cells than that in porcine PK-15 cells. However, the underlying mechanism remains unknown. In this study, we comprehensively compared the expression features between IBRS-2 cells and PK-15 cells in response to SVV infection by an unbiased high-throughput quantitative proteomic analysis. We found that the innate immune response-realted pathways were efficiently activated in PK-15 cells but not in IBRS-2 cells during SVV infection. A large amount of interferon-stimulated genes (ISGs) were induced in PK-15 cells. In contrast, no ISGs were induced in IBRS-2 cells after SVV infection. This different expression features in the two cell lines was verified by qPCR analysis. Besides, we determined similar results in the two cell lines during another porcine picornavirus foot-and-mouth disease virus (FMDV) infection. Further study demonstrated that the Janus kinase signal transducer and activator of transcription (JAK-STAT) signaling pathway was functioning properly in both IBRS-2 and PK-15 cells. A systematic screening study revealed that the aberrant signal transduction from TBK1 to IRF3 in the RIG-I-like receptor signaling pathway in IBRS-2 cells was the fundamental cause of the different innate immune response manifestation and different viral replication rate in the two cell lines. Together, our findings determined the different feature of IBRS-2 and PK-15 cell lines which will provide useful guidance for choosing right cell line in porcine picornaviruses-mediated host immune signaling research and could be easily extended to other porcine viruses.

Project description:Lotus (Nelumbo nucifera Gaertn) belongs to the family Nymphaeaceae, and is a popular aquatic vegetable that is rich in nutrients. It is widely cultivated in China, and many varieties of lotus are used for different purposes, suited to different climates, and consumed in different ways. Lotus is commonly produced by asexual propagation, so mutation through hybridization and variation fixed by asexual propagation are the main ways to create new varieties. Therefore, the formation of adventitious roots (ARs) in lotus, which does not have a well-developed principal root, is an important part of growth and development. It would be very useful to control ARs formation for lotus production and breeding.

Beauveria bassiana multiprotein bridging factor 1 (BbMBF1) regulating genes involved in fungal response to the prolonged thermal stress

Project description:BbMBF1 played crucial roles in mediating response the prolonged thermal stress, a determinant to the environmental fitness of fungal entomopathogens. We characterized for the first time that disruption of BbMBF1 reduced the mycelial tolerance to the 9-h thermal stress under 40Â°C. The global transcriptome involved in the response to the thermal stress was analyzed by using high throughput sequencing (RNA-Seq). Our transcriptional profiles revealed that numerous differentially expressed genes (DEGs), of which involved in metabolism, cell transport and cell rescue, were significantly involved in fungal response to the themal stress. 1. Total RNA obtained from BbMBF1 disruption mutant were compared to that of wild type strain under control conditin (free of thermal stress); 2. Total RNA obtained from BbMBF1 disruption mutant were compared to that of WT strain under 9-h thermal stress at 40Â°C.

Human follicular fluid neurotransmitters LC-MSMS

Project description:Pretreated human follicular fluid was prepared for targeted metabolomics experiments. For this purpose, 12-point standard calibration curves in the respective range with reference material were measured, and the limit of detection (LOD) and limit of quantification (LOQ) for each neurotransmitter under investigation were calculated. LC-MS/MS was conducted by an ultra-performance liquid chromatography method using a Waters ACQUITY UPLC BEH C18 column (2.1 mm×100 mm, 1.7 µm, Waters, USA). The Waters acquisition UPLC was coupled to an API 4000 triple quadrupole mass spectrometer (AB SCIEX, USA). The mobile phase A buffer was 10% methanol in water (containing 0.1% formic acid), and the mobile phase B buffer was 50% methanol in water (containing 0.1% formic acid). The LC gradient elution was at a flow rate of 0.3 mL/min at 0-8.0 min and 0.4 ml/min at 8.0-17.5 min, then 10%-30% B at 0-6.5 min; 30%-100% B at 6.5-7 min; 100% B at 7-14min; and 100%-10% B at 14-17.5min. The injection volume was 5 µl and the column temperature was set to 40 °C. Twenty-three metabolites (Ach: Acetylcholine chloride; Gln: Glutamine; Glu: Glutamate; His: L-Histidine; NE: norepinephrine; Tyr:Tyrosine; Trp:Tryptophan; Kyn: Kynurenine; GABA: 4-Aminobutyric acid;HisA:Histamine; PA:Picolinic acid;TyrA:Tyramine;DA:Hydroxytyramine hydrochloride; TrpA:Tryptamine;5-HT:Serotonin hydrochloride;E:Adrenaline hydrochloride;KynA:Kynurenic acid;5-HIAA:5-Hydroxyindole-3-acetic acid;DOPA:Levodopa;XA:Xanthurenic acid;VWA:Vanillymandelic Acid; 5-HTTP:5-Hydroxytryptophan;MT:Melatonine)were simultaneously reported in LC-MS/MS multiple reaction monitoring (MRM) mode. Data analysis was performed with Analyst 1.6 software.

A proteomics of gills approach to understanding salinity adaptation of Scylla paramamosain

Project description:Scylla paramamosain (Crustacea) is a commercially important euryhaline species distributed along the coast of southern China and other Indo-Pacific countries. However, a sudden variation in salinity will cause injury or even death of S. paramamosain. In this paper, we simulated a sudden decrease in salinity due to heavy precipitation in crab ponds. Comparison of gill microstructures of individuals in the control group and decreased salinity group showed gills became shorter and thicker, while the top of the filaments became swollen and then returned to normal after 120 h. A total of 3962 proteins were identified by proteomic sequencing of gills after 120 h under conditions of decreased salinity. 845 proteins were differentially expressed proteins: 371 up-regulated and 474 down-regulated. Of the enriched KEGG pathways, 20 were up-regulated and 14 were down-regulation (p<0.05). Among the significantly enriched up-regulated pathways, six were associated with amino acid metabolism and three were associated with Na+-K+-ATPase enzymatic activities. Pathways associated with redox metabolism and energy metabolism were identified. These results showed that in response to a decrease in salinity, S. paramamosain could adapt to the environment after 120 h. Molecular mechanism of this adaptation involved amino acid metabolism and Na+-K+-ATPase ion transport. Meanwhile, energy metabolism and redox metabolism were critical to the adaptation to a sudden decrease in salinity. This study, for the first time at the protein level, revealed the molecular mechanisms underlying salinity adaption of S. paramamosain and provides theoretical guidelines for the cultivation of S. paramamosain and other marine crustaceans.
