Global lysine acetylome analysis of Picea asperata somatic 1 embryos during partial desiccation

ABSTRACT: By the combination of affinity enrichment and high-resolution LC-MS/MS analysis, large-scale lysine acetylome analysis was performed in picea asperata. Altogether, 1,360 lysine acetylation sites in 717 protein groups were identified. Intensive bioinformatic analysis was then carried out to annotate those lysine acetylated targets, including protein annotation, functional classification, functional enrichment, etc.

Ventral hippocampal CA1 modulates pain behaviors in mice with peripheral inflammation

Project description:Chronic pain is one of the most significant and costly medical problems throughout the world. Recent evidence has confirmed the hippocampus as an active modulator of pain chronicity but the underlying mechanisms remain poorly defined. By means of in vivo electrophysiology together with chemogenetic and optogenetic manipulations in freely behaving mice, we identified a neural ensemble in the ventral hippocampal CA1 (vCA1) that showed inhibitory responses to noxious external stimuli, but not to innocuous stimuli. Following peripheral inflammation, this neuronal ensemble became responsive to innocuous stimuli and causally contributed to sensory hypersensitivity in inflammatory animals. Mimicking this inhibition of vCA1 neurons using chemogenetics in naïve mice induced chronic pain-like behavioral changes, whereas activating these vCA1 neurons in mice with chronic peripheral inflammation resulted in a striking reduction of pain-related behaviors. Pathway-specific manipulation of vCA1 projections to the basolateral amygdala (BLA) and infralimbic cortex (IL) showed that these pathways were differentially involved in pain modulation at different temporal stages of chronic inflammatory pain. These results confirm a crucial role of the ventral hippocampus and its circuits in modulating the development of chronic pain in mice.

Quantitative analysis of the global proteome of heart tissue in septic shock

Project description:Nowadays, sepsis and septic shock have become major public health problems, which are the main cause of death among patients admitted to the intensive care units. Notably, myocardial dysfunction during sepsis, usually called sepsis-induced myocardial depression, is common in septic shock patients whose incidence is about 70% and leads to a high mortality. However, the mechanism underlying the septic myocardial depression is still unclear. Recently, proteomics has become a powerful method for explore protein dynamics and their complex regulatory mechanism, and thus generates a profound impact on precision medicine and the clinical setting. Importantly, the expression patterns of global proteins in heart tissue between sepsis and control group remain unclear. Therefore, we performed the rat models of sepsis-induced myocardial depression and investigated global protein expression profiles in heart tissue between sepsis and control group using 4D label-free proteomic technique.

Proteomic Comparison Between Hyphae and Spores Reveals Pathogenicity Diversity in Mucor Irregularis

Project description:Mucor irregularis poses significant clinical risks, including severe disfigurement, with hyphal forms being more commonly encountered in clinical settings compared to spore forms. However, the pathogenic differences between these forms at the protein level remain poorly understood. This study employed TMT-based quantitative proteomics to investigate the proteomic differences between spores and hyphae of Mucor irregularis. Our analysis revealed that differentially abundant proteins are predominantly expressed during the hyphal stage. Notably, key enzymes such as RhoGEF GTPase, Subtilisin-related Protease, Acyl-CoA Synthetase, and Ribonuclease T2 appear crucial for hyphal morphological development and pathogenic mechanisms. Additionally, myosins play a significant role not only in hyphal growth and virulence but also in influencing sensitivity to antifungal agents. These findings provide new insights into the morphogenesis, development, and metabolic processes of Mucor irregularis, laying a foundation for further research into its pathogenic mechanisms.

E3 ubiquitin ligase Listerin regulates macrophage cholesterol efflux and atherosclerosis by targeting ABCA1

Project description:Atherosclerosis arises from disrupted cholesterol metabolism, notably impaired macrophage cholesterol efflux leading to foam cell formation. Through single-cell and bulk RNA sequencing, we identified Listerin as a regulator of macrophage cholesterol metabolism. Listerin expression increased during atherosclerosis progression in humans and rodents. Its deficiency suppressed cholesterol efflux, promoted foam cell formation, and exacerbated plaque features (macrophage infiltration, lipid deposition, necrotic cores) in macrophage-specific knockout mice. Conversely, Listerin overexpression attenuated these atherosclerotic manifestations. Mechanistically, Listerin stabilizes ABCA1, a key cholesterol efflux mediator, by catalyzing K63-linked polyubiquitination at residues K1884/K1957, countering ESCRT-mediated lysosomal degradation of ABCA1 induced by oxLDL. ABCA1 agonist Erythrodiol restored cholesterol efflux in Listerin-deficient macrophages, while ABCA1 knockout abolished Listerin's effects in THP-1 cells. This study establishes Listerin as a protective factor in atherosclerosis via post-translational stabilization of ABCA1, offering a potential therapeutic strategy targeting ABCA1 ubiquitination to enhance cholesterol efflux.

Deubiquitination of CDC6 by OTUD6A promotes tumour progression and chemoresistance

Project description:CDC6 is an oncogenic protein whose expression level fluctuates during the cell cycle. Although several E3 ubiquitin ligases responsible for the ubiquitin-mediated proteolysis of CDC6 have been identified, the deubiquitination pathway for CDC6 has not been investigated. Here, we report that OTUD6A functions as a deubiquitinase of CDC6. OTUD6A promotes cell proliferation and decreases sensitivity to chemotherapy by upregulating CDC6 protein expression.

Project description:Through the quantitative proteomics research strategy of TMT labeling and phosphorylation enrichment technology and high-resolution liquid chromatography-mass spectrometry, this project carried out the quantitative research of phosphorylated proteomics. In this study, a total of 6227 phosphorylation sites were identified, of which 4501 sites contained quantitative information. In order to ensure the high reliability of the results, we used the standard of localization probability>0.75 to filter the identification data, and finally determined 4842 phosphorylation sites located on 2328 proteins, of which 4,011 sites of 1996 proteins contained quantitative Information, and use the filtered data for subsequent bioinformatics analysis. If 1.5 times is the change threshold, t-test p-value<0.05 is the standard, and after full proteomics normalization, then among the quantified phosphorylation sites, there are 161 in the AC-B4vsAC-MG comparison group The modification level of the locus was up-regulated, and the modification level of 364 sites was down-regulated (for more information about the differences between the comparison groups, please see the main body of the report). Based on the above data, we conducted a systematic bioinformatics analysis of proteins containing quantitative information, including protein annotation, functional classification, functional enrichment, and cluster analysis based on functional enrichment.
