Project description:Using atomic force microscopy imaging and nanoindentation measurements, the effect of the minor capsid proteins pUL17 and pUL25 on the structural stability of the icosahedral Herpes Simplex Virus type 1 capsids was investigated. pUL17 and pUL25 that form the capsid vertex-specific component particularly contributed to the capsid resilience along the 5-fold and 2-fold, but not the 3-fold icosahedral axes. We tested whether deletion of UL17 and UL25 changed the levels of the proteins that assemble in the nucleocapsid by quantitative mass spectrometry.

Sequencing of HSV-1 genomic DNA to identify genomic variation resulting from selective pressure imposed by mutation of the PP1 binding site in the pUL21 gene

Project description:The pUL21 gene is essential for efficient replication and dissemination in herpes simplex virus 1 (HSV-1) but the molecular basis for its function is not well understood. Experimental work revealed that pUL21 is a virus-encoded adaptor of protein phosphatase 1 (PP1). Mutation of the specific motif in pUL21 responsible for PPI recruitment, followed by propagation in cell culture, was performed in order to identify the impact of preventing PP1 recruitment on the viral genome. This allowed identification of compensatory mutations disrupting the viral kinase gene pUS3, which restored viral growth and spread, revealing that pUL21 directly antagonises pUS3. Three mutant strains of HSV-1 were generated, mutating the conserved residues F242 and V243 in the PP1 binding sequence, individually and in combination (pUL21F242E, pUL21V243D, pUL21FV242AA), along with a knockout strain lacking pUL21 (ΔpUL21). These four virus stocks, plus wild-type HSV-1, were propagated in Vero cells for three passages. Genomic DNA was extracted from the P3 viruses and sequenced with Illumina. Reads were trimmed and mapped to an edited version of the HSV-1 genome (based on JQ673480.1) and read depth and variants were quantified.

Herpes simplex virus type 1 modifies the protein composition of extracellular vesicles to promote neurite outgrowth and facilitate neuroinfection

Project description:Herpes simplex virus type 1 (HSV-1) is a highly contagious and prevalent human pathogen that causes many diseases, including encephalitis. During primary infection, HSV-1 infects epithelial cells and then neurites of peripheral neurons, establishing lifelong infection in the neuronal cell body. Neurites are highly dynamic structures that grow or retract in the presence of attractive or repulsive cues, respectively. Whether HSV-1 modulates these cues and thereby neurite outgrowth was not known. Here, we show that HSV-1 glycoprotein G (gG) reduces the inhibitory effect of epithelial cells on neurite outgrowth, facilitating viral infection of neurons through neurite ends. The mechanism of action involves the modification of the protein composition of extracellular vesicles (EV), including increased amount of galectin-1. We show that galectin-1 located in EV produced during HSV-1 infection of epithelial cells promotes neurite outgrowth, and this activity can be partially neutralized by antibodies. This study provides new insights into the neurotropism of HSV-1, identifying for the first time a viral protein that modifies the protein composition of EV to increase neurite outgrowth and neuronal infection.

Cellular interactome of HSV-1 pUL21

Project description:The herpes simplex virus (HSV)-1 protein pUL21 is essential for efficient virus replication and dissemination. While pUL21 has been shown to promote multiple steps of virus assembly and spread, the molecular basis of its function remained unclear. Here we identify that pUL21 is a virus-encoded adaptor of protein phosphatase 1 (PP1). pUL21 directs the dephosphorylation of cellular and virus proteins, including components of the viral nuclear egress complex, and we define a conserved non-canonical linear motif in pUL21 that is essential for PP1 recruitment. In vitro evolution experiments reveal that pUL21 directly antagonises the activity of the virus-encoded kinase pUS3, with growth and spread of pUL21 PP1-binding mutant viruses being restored when pUS3 activity is disrupted. This study shows that virus-directed phosphatase activity is essential for efficient herpesvirus assembly and spread, highlighting the fine balance between kinase and phosphatase activity required for optimal virus replication.

Whole-cell and plasma membrane proteomics of herpes simplex virus-1 infection reveals cell surface remodelling via pUL56-mediated GOPC degradation

Project description:Herpesviruses are ubiquitous in the human population and they extensively remodel the cellular environment during infection. Multiplexed quantitative proteomic analysis over the time-course of herpes simplex virus (HSV)-1 infection was used to characterize changes in the host-cell proteome and the kinetics of viral protein production. Several host-cell proteins are targeted for rapid degradation by HSV-1, including the cellular trafficking factor GOPC. We show that the poorly-characterized HSV-1 pUL56 directly binds GOPC, stimulating its ubiquitination and proteasomal degradation. Plasma membrane profiling revealed that pUL56 mediates specific changes to the cell surface proteome of infected cells, including loss of IL18 receptor and Toll-like receptor 2, and that cell surface expression of Toll-like receptor 2 is GOPC-dependent. Our study provides significant resources for future investigation of HSV-host interactions and highlights an unanticipated and efficient mechanism whereby a single virus protein targets a cellular trafficking factor to modify the surface of infected cells.

RAW264.7 cells were infected with Vesicular Stomatitis Virus (VSV, MOI=1) or herpes simplex virus 1 (HSV-1, MOI=5) for 6h. Equal amounts of RNA were assayed for gene expression using Affymetrix mouse 430 2.0 arrays.

Identifying host factors associated with DNA replicated during virus infection

Project description:Viral DNA genomes replicating in cells encounter a myriad of host factors that facilitate or hinder viral replication. Viral proteins expressed early during infection modulate host factors interacting with viral genomes, recruiting proteins to promote viral replication, and limiting access to antiviral repressors. Although some host factors manipulated by viruses have been identified, we know little about pathways exploited during infection and how these differ between viruses. To identify cellular processes manipulated during viral replication, we defined proteomes associated with viral genomes during infection with adenovirus, herpes simplex virus and vaccinia virus. We compared enrichment of host factors between virus proteomes and confirmed association with viral genomes and replication compartments. Using adenovirus as an illustrative example, we uncovered host factors deactivated by early viral proteins, and identified a subgroup of nucleolar proteins that aid virus replication. Our datasets provide valuable resources of virus-host interactions that affect proteins on viral genomes.

Time-resolved Global and Chromatin Proteomics during Herpes Simplex Virus (HSV-1) Infection

Project description:Herpes simplex virus (HSV-1) lytic infection results in global changes to the host cell proteome and the proteins associated with host chromatin. We present a system level characterization of proteome dynamics during infection by performing a multi-dimensional analysis during HSV-1 lytic infection of human foreskin fibroblast (HFF) cells. Our study includes identification and quantification of the host and viral proteomes, phosphoproteomes, chromatin bound proteomes and post-translational modifications (PTMs) on cellular histones during infection. We analyzed proteomes across six time points of virus infection (0, 3, 6, 9, 12 and 15 hours post-infection) and clustered trends in abundance using fuzzy c-means. Globally, we accurately quantified more than 4,000 proteins, 200 differently modified histone peptides and 9,000 phosphorylation sites on cellular proteins. In addition, we identified 67 viral proteins and quantified 571 phosphorylation events (465 with high confidence site localization) on viral proteins, which is currently the most comprehensive map of HSV-1 phosphoproteome. We investigated chromatin bound proteins by proteomic analysis of the high-salt chromatin fraction and identified 510 proteins that were significantly different in abundance during infection. We found 53 histone marks significantly regulated during virus infection, including a steady increase of histone H3 acetylation (H3K9ac and H3K14ac). Our data provide a resource of unprecedented depth for human and viral proteome dynamics during infection. Collectively, our results indicate that the proteome composition of the chromatin of HFF cells is highly affected during HSV-1 infection, and that phosphorylation events are abundant on viral proteins. We propose that our epi-proteomics approach will prove to be important in the characterization of other model infectious systems that involve changes to chromatin composition.
