Dynamics of protein expression reveals primary targets and secondary messengers of estrogen receptor signaling in MCF-7 breast cancer cells

Project description:Estrogen receptor-mediated proliferation of breast cancer cells is facilitated through expression of multiple primary target genes, products of which induce the secondary response to stimulation. To differentiate between the primary and secondary target genes of estrogen receptor signaling, we measured dynamics of protein expression induced by estradiol in MCF-7 breast cancer cells. Measurement of the global proteomic effects of estradiol by SILAC resulted in identification of 104 estradiol-regulated proteins, with only 41 corresponding genes having estrogen response elements (EREs). Selected reaction monitoring assays were used to validate 32 proteins and measure the dynamics of their expression within 72 hours upon estradiol stimulation and in the presence of 4-hydroxytamoxifen, thus confirming estrogen receptor-mediated signaling. Dynamics of protein expression revealed immediate early response proteins including TFF1 and CDK1 and delayed response proteins including CDK2 and NAB2. Presence or absence of EREs in the corresponding genes indicated early or delayed expression of proteins identified by SILAC. Finally, we measured accurate dynamics of estradiol-induced protein expression in the sub-network of primary and secondary targets of estrogen receptor. Interestingly, since NAB2 protein is also a repressor of EGR3-induced transcription, siRNA-mediated silencing of NAB2 resulted in the over-expression of some EGR3-induced proteins, such as ITGA2. To conclude, quantitative proteomics revealed secondary targets of estrogen receptor signaling potentially overlooked by genome-wide profiling of EREs and provided dynamics of protein expression in the network of transcription factors with a negative feedback loop.

Human endothelial gene expression under estradiol and oxLDL treatments

Project description:DNA microarrays were used to investigate global gene expression patterns in cultured human umbilical artery endothelial cells (HUAECs) exposed to 1 nmol/L estradiol and/or 100 µg/ml oxidized low density lipoprotein (oxLDL) for 24 hours compared to control cells. HUAECs from 15 separate cultures were exposed to control (0.1% ethanol), 1nmol/L estradiol, 100 µg/ml oxLDL, or 1nmol/L estradiol + 100 µg/ml oxLDL treatments for 24 h. Total cellular RNA was extracted. Equal amounts of RNA extracted from 3 control cells or 3 estradiol-treated cells obtained from three different cultures were pooled, achieving five biological replicates of the control, five replicates that were treated with estradiol, five replicates that were treated with oxLDL and five replicates that were treated with estradiol+oxLDL . Therefore, a total number of 20 microarrays were developed.

Transcription profiling of human umbilical vein endothelial cells after pulsed or continuous estradiol exposure

Project description:The experiment aims to investigate differences in the transcriptional response of human umbilical vein endothelial cells to estradiol treatment associated with the mode of administration. Human umbilical vein endothelial cells were treated for 5 days with a 1h pulse of 2.4 x 10-8 M 7β-estradiol (E2) once every day or with continuous 10-9M E2, ensuring cells received the same 24h E2 exposure (concentration x time) of two treatments. Control cells were treated with ethanol vehicle. For pulsed treatment, cells were washed and fresh medium was added after the 1h estradiol pulse. For continuous daily treatments, cells were incubated for 24h with media containing estradiol following the same scheme of media changes. The experiment was repeated at least three times.
