Molecular profiling of vascular beds in vivo by DISDIVO

ABSTRACT: We developed a new technique named as Differential Systemic Decellularization in vivo (DISDIVO) for the molecular profiling of vascular beds throughout the body. This method can by applied to murine models. Thus, the data provided here correspond to mice vascular beds obtained in vivo by DISDIVO.

Vascular bed systematic profiling unravels endothelial and glycocalyx dysregulations in experimental high salt hypertension

Project description:Objective: Considerable evidence links dietary salt intake with the development of hypertension, left ventricular hypertrophy, and increased risk of stroke and coronary heart disease. Despite extensive epidemiological and basic science interrogation of the relationship between high salt (HS) intake and blood pressure, it remains unclear how HS impacts endothelial cell (EC) and vascular structure in vivo. This study aims to uncover the HS-induced vascular pathologies using a differential systemic decellularization in vivo (DISDIVO) approach.Approach and Results: We performed systematic molecular characterization of the endothelial glycocalyx (eGC) and EC proteomes in mice with HS (8%) diet-induced hypertension versus healthy control animals. Isolation of eGC and ECs compartments was achieved using the DISDIVO approach. Altered protein expression in hypertensive compared to normal mice was characterized by liquid chromatography tandem-mass spectrometry (LC-MS/MS). Proteomic results from eGC fractions revealed a significant downregulation of eGC and associated proteins in HS diet-induced hypertensive mice. Among 1696 proteins identified in this group, 723 were markedly downregulated while 168 were upregulated, bioinformatic analysis indicates critical damage and derangement of eGC layer. In the ECs fraction HS-induced hypertension significantly altered protein mediators of contractility, metabolism, mechanotransduction, renal functions, and coagulation cascades. In particular, we observed dysregulation of integrin subunits alpha2, alpha2b, and alpha5, which relay external signals to the actin cytoskeleton.Conclusion: These findings provide novel molecular insight on HS-induced structure changes in eGC and ECs that may increase cardiovascular risk and potentially guide the development of new diagnostics and therapeutic interventions.

Project description:The interspersed anatomic distribution of endothelial cells (ECs) and their dependence on microenvironmental cues have been major challenges in studying EC function and response to (patho)physiological stimuli in vivo. Combining EC-specific translating ribosome affinity purification (EC-TRAP) with RNA sequencing provides an accurate in vivo snapshot of tissue-specific EC expression profiles, and maintains a high degree of sensitivity to detect low abundant transcripts while limiting changes in gene expression profiles due to enzymatic tissue dissociation required to generate single cell suspensions for FACS sorting or single cell analysis. In addition to demonstrating EC heterogeneity under physiologic conditions, the in vivo host response to lipopolysaccharide (LPS) revealed the induction of expression programs associated with a native defense response, with marked differences across vascular beds.

Physiological Vascular Permeability Requires Induction of Endothelial NR4A1 by Progesterone Receptor [ChIP-Seq]

Project description:Vascular permeability is frequently associated with inflammation and it is triggered by chemokines and by a cohort of secreted permeability factors, such as VEGF. In contrast, here we showed that the physiological vascular permeability that precedes implantation is directly controlled by progesterone receptor (PR) and it is independent of VEGF. Both global and endothelial-specific deletion of PR block physiological vascular permeability in the uterus while misexpression of PR in the endothelium of other organs results in ectopic vascular leakage. Integration of genome-wide transcriptional profile of endothelium and ChIP-sequencing revealed that PR induces a NR4A1 (Nur77/TR3) specific transcriptional program that broadly regulates vascular permeability in response to progesterone. This program triggers concurrent suppression of several junctional proteins and leads to an effective, timely and venule-specific regulation of vascular barrier function. Silencing NR4A1 blocks PR-mediated permeability responses indicating a direct link between PR and NR4A1. These results reveal a previously unknown function for progesterone receptor on endothelial cell biology with consequences to physiological vascular permeability and implications to the clinical use of progestins and anti-progestins on blood vessel integrity.
