Project description:To identify the expression profiles of zebra mussel byssus unique genes with the regeneration of the byssal threads, a time-course study was performed. The RNA samples from the feet of fully attached zebra mussels (maintained attachment for more than 4 weeks) were extracted as common reference. The detached mussels were allowed to re-attach on underwater surface. Then the RNA samples from feet of the mussels at different time points post detachment were taken, namely, 12 hours, 1 day, 2 days, 3 days, 4 days, 7 days, and 21 days post detachment. The comparisons of the genes expression profiles were made between different time points and reference, as well as the any two connected time points (expect for 7 days and 21 days).

Project description:Mussels synthesize an interesting class of biological materials with unique properties to adhere onto virtually any solid surface. Inter- and cross-species analyses have revealed that mussel byssus fabrication is influenced by environmental factors. In this study, proteins expressed specifically byssus and in the byssal-producing organ (foot) were examined for two mussel species from the Mytilidae family using liquid chromatography coupled with high-resolution tandem mass spectrometry (LC-MS/MS). The main goal was to describe which proteins are exclusively expressed in the mussel's foot gland, which is responsible for byssus secretion. Proteins uniquely found in the foot samples have been found by comparing to a control tissue (mantle). The overlap with proteins found in byssus samples were also investigated. A restricted list of these proteins have been highlighted as byssus-related, believed to contribute to the unique properties of byssus, such as securing adhesion and toughness, and plasticity based on environmental factors (including promoter regulation). This work may provide further directions on thread biofabrication and additional information on its regulation, as well as contribute to a more complete description of the mussel proteome.

Project description:The winged pearl oyster, Pteria penguin, is a widely distributed and economically important bivalve with unique and primitive byssus. The byssogensis of this species is largely unknown. Here we report the lable free quantification analysis on different tissues of P. penguin.Through the analysis, we found that the process of byssogenesis requires ATP to provide energy among the differential proteins; and seven (Tenascin-X, laminin, papilin, fibrillin-1, spondin, protease inhibitors and Mucins) of the 31 proteins identified in the byssus were derived from ECM. Functions of extracellular matrix proteins can be correlated with the formation and the nature of the byssus.

Project description:Many bivalve species produce groups of strong proteinaceous byssal threads to rigidly attach to underwater substrates. Fibres like these have potential applications as biomedical materials due to their unique mechanical characteristics. The byssus and byssal thread producing glands of Pinctada maxima have not yet been characterised. RNA was isolated from P. maxima foot and byssal stem region tissues and sequenced using the Illumina platform. A de novo reference transcriptome comprising 34,281 contiguous sequences was assembled, and tissue replicates were mapped against the reference for quantitative analysis. Tryptic digests of byssal threads were analysed by LC-MS/MS. The resultant peptides were matched to 62 protein sequences derived from our reference transcriptome. Components of the byssus were identified for further characterisation, including a highly expressed perlucin-like foot protein (Pmfp1) and a recently identified protein that we refer to herein as glycine-rich thread (GRT) protein. This work provides principal knowledge on the molecular components of the byssus for P. maxima and the foot ultrastructure involved in the creation of byssal threads. This study advances our knowledge of byssus biosynthesis in non-mytilids, providing a platform for the design of new marine biopolymers.

Project description:To identify genes involved in nutrition metabolism of Macrobrachium nipponense, two independent cDNA libraries from hepatopancreas and muscle were constructed through high-throughput next-generation sequencing techniques. A total of 112,548,142 and 90,140,774 high quality reads were generated in the two cDNA libraries, respectively. Clustering and assembly of these reads produced a nonredundant set of 98,560 unique sequences, with an average unigene length of 793 bp. The unigene sequences were subjected to GO, COG and KEGG functional classification. A large number of differentially expressed genes were recovered by comparison of the two tissues. The differentially expressed and their functions were predicted by KEGG pathway mapping. Furthermore, 17,971SSRs and 32,935 high-confidence SNPs were identified in the two EST datasets.
