Physcomitrella patens secreted endogenous peptides

ABSTRACT: We used mass spectrometry to comprehensively analyze the endogenous peptide pools generated from functionally active proteins in the secretome fromthe model plant Physcomitrella patens. Treatment with the plant stress hormones methyl jasmonate and salicylic acid induced significant differential changes in the peptide pool.

Project description:Plant secretome comprise dozens of secreted proteins and peptide hormones. However, little is known about peptide composition of plant secretome as well as proteases responsible for peptide pools generation. While the majority of studies focuses on the target detection and characterization of specific plant peptide hormones, we performed a comprehensive analysis of the whole extracellular peptidome, using moss Physcomitrella patens as a model object. We identified hundreds of modified and unmodified endogenous peptides originated from functional and non-functional protein precursors. Plant proteases responsible for shaping the pool of endogenous peptides were predicted. We showed that salicylic acid (SA) induces peptide production in the secretome.

Since the moss P.patens is unable to perceive the known bacterial elicitors such as flagellin or elf18, we induced the release of potential signaling peptides by chitosan.

Project description:Experiment to identify genes for responding to water stress in the moss Physcomitrella patens using various available water stresses such as mannitol, NaCl, and drought. Treatments were carried out on duplicate tissue samples. RNA isolated independently from each tissue sample and monitored for stress/hormone induction of the PpLea-2 gene by Northern blot hybridisation. Duplicate RNA samples bulked for Cy3/Cy5 labelling and array hybridisation. Two microarrays per treatment hybridised using a reciprocal dye swap (control -vs- treatment).

Genome-wide expression profiling of PpABI1 disruptants

Project description:To investigate the disruption effect of Group A PP2C on global gene expression in Physcomitrella patens, we performed microarray analysis of wildtype plant and PpABI1 disruptants using a custom Physcomitrella oligonucleotide microarray, which carries probes for 33,942 gene models of Physcomitrella genome version 1.1. on a 4 x 44 K Agilent platform. Gene expression in wildtype plant and ppabi1 disruptants were measured at 0, 1, 3, 6 and 12 hours after exposure to 10 M-NM-<M ABA. Three biologically independent experiments were performed at 0 hour. Also to discover the early ABA-responsive genes, gene expression in wildtype was measured at 0 and 3 hours after exposure to 1 M-NM-<M ABA.

Transcriptome of Protoplasts Reprogrammed into Stem Cells in Physcomitrella patens

Project description:We obtained genome-wide digital gene expression tag profiles within the first three days of P. patens protoplast reprogramming. At four time-points during protoplast reprogramming, the transcript levels of 4827 genes changed more than four-fold and their expression correlated with the reprogramming phase. Gene ontology (GO) and pathway enrichment analysis of differentially expressed genes (DEGs) identified a set of significantly enriched GO terms and pathways, most of which were associated with photosynthesis, protein synthesis and stress responses. DEGs were grouped into six clusters that showed specific expression patterns using a K-means clustering algorithm. An investigation of function and expression patterns of genes identified a number of key candidate genes and pathways in early stages of protoplast reprogramming, which provided important clues to reveal the molecular mechanisms responsible for protoplast reprogramming. We identified genes that show highly dynamic changes in expression during protoplast reprogramming into stem cells in P. patens. These genes are potential targets for further functional characterization and should be valuable for exploration of the mechanisms of stem cell reprogramming. In particular, our data provides evidence that protoplasts of P. patens are an ideal model system for elucidation of the molecular mechanisms underlying differentiated plant cell reprogramming. Comparison of wildtype and gr1 knock-out mutants of Physcomitrella patens using quantitative proteomics

Project description:(1) While it is known that redox-regulation in chloroplasts is vital for functional photosynthesis, the interplay of different redox-cascades in balancing reactive oxygen species (ROS) with metabolic regulation via thiol switching is still partly unresolved. We investigate the importance of the glutathione reductase isoform maintaining a highly reducing stromal glutathione redox potential (EGSH). (2) Using the model moss Physcomitrella patens, we knocked-out the plastid/mitochondrial GR isoform. We generated a sensor line expressing plastid-targeted redox-sensitive GFP to monitor stromal EGSH in vivo, and compared protein abundances between wildtype (WT) and Δgr1 knock-out mutants by a metabolic labelling approach. (3) On the organelle level, we find that the absence of PpGR1 leads to a shift of stromal EGSH to less reducing values that is not rescued in the light, whereas WT EGSH was clearly responsive to light. On the plant level, growth and photosynthetic performance were decreased with increasing light intensities while ascorbate and zeaxanthin levels were elevated. Proteomics showed an induction of proteins involved in plastid protein repair and degradation. (4) Our results indicate a limited cross-talk between plastid thioredoxin and glutathione redox systems and show that glutathione reductase is necessary for efficient photosynthesis under stress and non-stress conditions.

Genome-wide transcriptome analysis of gametophyte development in Physcomitrella patens

Project description:Gene expression was analyzed with digital gene expression tag profiling (DGEP) with samples taken from P. patens protonema at 3, 14 and 24 days, and also from or leafy shoot tissues l at 30 days after protoplastes isolated, and from 14-day-old caulonemal and chloronemal tissues. Totally, 4333 genes were identified as differentially displayed. Among them, 4129 were developmental-stage specific and 423 were preferentially expressed in either chloronemal or caulonemal tissues. Most of the differentially displayed genes could be assigned to functions in organic substance and energy metabolism or macromolecule biosynthetic and catabolic process based on gene ontology (GO) description. In addition, some regulatory genes were identified as candidates that might be involved in controlling the developmental stage transition and cell differentiation, namely: MYB-like, HB-8, AL3, zinc finger family proteins, bHLH superfamily, GATA superfamily, GATA and bZIP transcription factors, protein kinases, the genes related to protein/amino acid methylation, and auxin, ethylene, and cytokinin signaling pathways. Examination of 4 different sampling times and 2 different organelles.
