Expression of proteins by a marine Brevibacterium casei strain in response to elevated levels of Cadmium

ABSTRACT: A marine bacterial isolate, Brevibacterium casei was grown in LB broth in presence of 50ppm Cadmium (provided in the form cadmium chloride, with final concentration of Cadmium at 50ppm). The cells were harvested at different time intervals and proteins extracted from these using indigenous protocol developed in laboratory. Similarly, the strain was grown in absence of Cadmium and same steps were followed, which was treated as control. The proteins extracted from test as well control set were digested using trypsin following standard protocol. Trypsin digests were injected in LCMS QToF for obtaining spectral library which was used to identify the proteins against NCBI database

Project description:A marine bacterial isolate, Brevibacterium casei #NIOSBA88 was grown separately in Luria Bertini broth supplemented with 750 mg L-1 Arsenic (III) and 100 mg L-1 Chromium respectively. Similarly, the strain was grown in absence of Arsenic and Chromium, which served as an experimental control. The cells were harvested at different time intervals and proteins were extracted from the cell pellets using in-house protocol (Shah and Damare 2018). The extracted proteins were digested using trypsin following standard protocol. Tryptic digests were injected in LCMS QToF for obtaining spectral library which was used to identify the proteins against a species specific database in NCBI.

Project description:The response to bile of Lactobacillus casei BL23 was investigated. Lactobacillus casei BL23 was grown in MRS medium at 37C without shaking until O.D. 595 nm. reached 0.5. The cultures were then two fold diluted with prewarmed MRS medium (control) or MRS supplemented with 0.2% bovine bile (treatment) and incubation continued for 45 min. Cells were then harvested and total RNA purified.

Comparative genome hybridization for Lactobacillus casei

Project description:Lactobacillus casei is remarkably adaptive to diverse habitats. To understand the evolution and adaptation of Lb. casei strains isolated from different environments, the gene content of 22 Lb. casei strains isolated from various habitats (cheeses, n=8; plant materials, n=8; and human sources, n=6) were examined by comparative genome hybridization with an Lb. casei ATCC 334-based microarray. Comparative genome hybridization was performed against an Affymetrix custom microarray designed to include 2,661 (97%) chromosomal and 17 (85%) plasmid CDSs predicted to occur in Lb. casei ATCC 334, as well as all predicted CDSs in the draft Lb. helveticus CNRZ 32 genome. CDSs that were not included in the microarray design were all transposase-encoding genes.

Gene expression profile of Lactobacillus casei Zhang in fermented milk during transit in simulated gastrointestinal tract

Project description:The survival of probiotics could be strongly enhanced by delivery vehicles, but the mechanism was unknown. In this study, whole genome microarray technology was used to detect the gene expression profiles of probiotic Lactobacillus casei Zhang in the absence and presence of fermented milk in simulated gastrointestinal tract. The results showed the gene expression profiles were significantly different under these two different conditions. The change of the gene expression profile may be helpful to comprehend the role of delivery vehicles in enhancing the survival of probiotics. Twelve samples of bacterial cells in the absence and presence of fermented milk were collected at 3 h in simulated gastric juice, 4 h and 8 h in intestinal juice, 2 biological replicates were obtained in each time point. The average of the normalized expression values of the 2 biological replicates for each probe was regarded as the expression value of a predicted gene. To identify the expression pattern of each gene across different treatments, n-fold change ratios were calculated for every gene in pure L. casei Zhang versus L. casei Zhang in fermented milk.
