Modulation of glioma cell proteome by Delta-24-RGD adenovirus (24hpi)

Project description:Glioblastoma multiforme (GBM) is the most common and aggressive type of malignant glioma. Oncolytic adenoviruses are being modified to exploit the aberrant expression of proteins in tumor cells to enhance tumor tropism and glioma-selective replication. E1A mutant adenovirus Delta-24-RGD has shown favorable toxicity profile and remarkable efficacy in a first-in-human phase I clinical trial. However, the comprehensive modulation of glioma metabolism in response to Delta-24-RGD infection is poorly understood. Integrating mass spectrometry based-quantitative proteomics, physical and functional interaction data, and biochemical approaches, we conducted a cell-wide study of intracellular and secreted glioma proteomes at late stage of Delta-24-RGD infection, when prominent autophagy has been described.

Genetic regulation of the ATP synthase in neurons modulates cognition

Project description:The mitochondrial ATP synthase produces ATP by oxidative phosphorylation and integrates different signals to regulate cellular functions and fate. The ATPase inhibitory factor 1 (IF1) is a structurally-disordered protein that inhibits the ATP synthase, contributing to metabolic reprogramming and signalling through mitochondrial reactive oxygen species (mtROS). mtROS regulate kinases and transcription factors in mitohormetic responses that favour adaptation to toxic insults2. IF1 is tissue-specifically expressed and in human and mouse brain is in molar excess over the ATP synthase. Herein, we have used genetic approaches to ablate or overexpress IF1 in neurons to investigate its role in brain functions. IF1 inhibits a fraction of the ATP synthase under physiological conditions and regulates respiration, mtROS production and mitochondrial structure. Transcriptomic, proteomic and metabolomic analyses indicate that IF1 regulates synaptic transmission and cognition. Ablation of IF1 impairs short-term memory whereas IF1 overexpression increases basal synaptic transmission and learning by mtROS-dependent activation of the extracellular signal-regulated kinases 1/2 (ERK 1/2). Overall, we show that IF1 dose plays a fundamental role in the regulation of neuronal function by controlling the fraction of inhibited ATP synthase that acts as source of mitohormetic mtROS, further emphasizing the ATP synthase/IF1 as promising targets to treat cognitive disorders.

Identification of protein-protein interactions involving the eIF4E binding proteins Caf20p and Eap1p on yeast via affinity purification and LC-MSMS.

Project description:There are multiple translational control pathways. These include pathways involving eIF4E binding proteins (4E-BPs), which inhibit translation by binding and sequestering the 5’ cap binding protein eIF4E away from its partner eIF4G. Saccharomyces cerevisiae has two 4E-BPs: Caf20p and Eap1p. Previous analyses had shown that each 4E-BP regulates different subsets of mRNAs. In order to assess whether binding different proteins caused their different regulatory role, we used tandem affinity purification followed by label-free mass spectrometry to compare the proteomes pulled-down with the TAP-tagged 4E-BPs, and the global proteome. These analyses point out that Caf20p and Eap1p share most interaction partners, including ribosomes, and that both bind several other RNA-binding proteins.
