Project description:Shells of pearl oyster are natural biominerals with remarkable properties and can be repaired after damage, which are regulated by biomacromolcules especially shell matrix proteins (SMPs). Identifying SMPs is critical for further understanding the process. Although proteomic methods have been used to reveal the complex protein mixture in mature shells, the proteomics of repaired shells after shell damage have not been reported before. Here, we studied the SMPs of the repaired shells (prismatic layers) 5-10 days after shell damage in Pinctada fucata by integrating transcriptomics and proteomics and compared the microstructure difference between repaired and mature shells. Although the repaired shells are calcite, similar to mature shells, the microstructure of repaired shells has holes in the center of prisms with irregular and curved edges. In total, we found 49 SMPs from the repaired shells including some proteins only existing in mature nacreous layers. Peroxidase-like protein and beta-N-acetylhexosaminidase may be important players in repaired shells. In addition, they have the capability to affect CaCO3 crystallization process in vitro, altering the packing and reducing the crystallinity of crystals. This study could improve our understanding of shell repair process and lay the foundation for studying SMPs-controlled biomineralization.

Project description:The development of protein anti-degradation strategy is important for their storage at ambient conditions. Despite that it is known that biominerals, typical inorganic-organic composites, can preserve proteins at room temperature for a long time, it is unclear the extent of protein degradation under high temperatures. In this study, we examined protein remaining in the toasted abalone shell under high temperatures (200 and 300 °C) by biomineral proteomics method. Surprisingly, 24 proteins including carbonic anhydrase, hemocyanin, actin can still be identified from shells even after toasting under 300°C, not much decreased compared to that in the 200°C-treated and the native shell. However, the microstructure and composition (both mineral and organic matrix) of shells were altered significantly revealed by scanning electron microscopy, infrared spectroscopy, and X-ray diffraction. The well-preserved of proteins may be partially due to the sacrifice of mineral/organic interfaces and the formation of nanopores in the shell at high temperatures. Moreover, the extracted proteins from both groups were able to affect calcium carbonate in vitro. This study has potential implications in various fields such as protein storage at high temperatures and palaeoproteomics.

Project description:This work reports a comprehensive and integrated microstructural, biochemical and proteomics study on the shell matrix of Spondylus gaederopus, the Mediterranean thorny oyster. We investigate the skeletal matrix proteins which are involved in biomineralization and compare the identified Spondylus sequences with other shell proteins, that are publicly available in databases. Using high-resolution liquid chromatography tandem mass spectrometry (LC-MS/MS) we characterized several shell protein fractions, isolated by different bleaching treatments. We identified six shell proteins, which also displayed features and domains typically found in biomineralized tissues, including the prevalence of intrinsically disordered regions. However, many reconstructed peptide sequences (de novo) could not be matched to any known shell proteins and we suggest that these probably represent lineage-specific sequences. The proteomic data implies that Spondylus may have evolved a distinct molecular toolkit for biomineralization. Project description:Background biology: Global warming has accelerated in recent decades, with the Arctic warming 2–3 times faster than the global average. As a result boreal species are expanding into the Arctic, at a pace reflecting environmental warming. Nevertheless, the poleward expansion of boreal marine species is restricted by their ability to tolerate low water temperatures, and in the case of intertidal species, sub-zero air temperatures during winter. In Greenland, however, the number of days with extreme sub-zero air temperatures has decreased by more than 50% since the 1950’s, suggesting that the low air temperature constraint is weakening. Although boreal intertidal species could potentially benefit from this warmer climate to establish populations in the Arctic, recent work has shown that local intertidal summer air temperatures in Greenland can exceed 36°C. This temperature is above the thermoregulatory capacity of many boreal intertidal species, including the highly abundant blue mussel Mytilus edulis. Therefore will further colonisation of M. edulis in Greenland be inhibited by the increasingly warm summer temperatures. Aim of experiment: Intertidal animals (Greenland blue mussel M. edulis) were sampled in situ on the first warm days of the year from the inner (warmer) and outer (cooler) regions of the Godthåbsfjorden around Nuuk (64°N) to examine the fjord temperature gradient effect. In addition, subtidal M. edulis were also collected and subjected to two acute temperature shocks of 22 and 32°C, which represented common and extreme summer air temperatures for intertidal habitats near Nuuk.

Project description:The contamination of marine ecosystems with microplastics, such as the polymer polyethylene, a commonly used component of single-use packaging, is of global concern. Although it has been suggested that biodegradable polymers, such as polylactic acid, may be used to replace some polyethylene packaging, little is known about their effects on marine organisms. Blue mussels, Mytilus edulis, have become a “model organism” for investigating the effects of microplastics in marine ecosystems. We show here that repeated exposure, over a period of 52 days in an outdoor mesocosm setting, of M. edulis to polyethylene microplastics reduced the number of byssal threads produced and the attachment strength (tenacity) by ~50%. Exposure to either type of microplastic altered the haemolymph proteome and, although a conserved response to microplastic exposure was observed, overall polyethylene resulted in more changes to protein abundances than polylactic acid. Many of the proteins affected are involved in vital biological processes, such as immune- and stress- regulation, metabolism and cellular and structural development. Our study highlights the utility of mass spectrometry-based proteomics to assess the health of key marine organisms and identifies the potential mechanisms by which microplastics, both conventional and biodegradable, could affect their ability to form and maintain reefs.

Project description:Nacre, the iridescent material found in pearls and shells of molluscs, is formed through an extraordinary process of matrix-assisted biomineralization. Despite recent advances, many parts of the biomineralization process and its evolutionary origin remain a mystery. The pearl oyster Pinctada fucata martensii is a well-known master of biomineralization, but the molecular mechanisms underlie its production of remarkable shells and pearls is not fully understood. We sequenced the highly polymorphic genome of the pearl oyster and conducted multi-omic and biochemical studies to probe nacre formation. We identified a large set of novel proteins participating in matrix-framework formation, many in expanded families, including components similar to that found in vertebrate bones such as collagen-related VWA-containing proteins (VWAP), chondroitin sulfotransferases and regulatory elements.Considering that there are only collagen-based matrices in vertebrate bones and chitin-based matrices in most invertebrate skeletons, the presence of both chitin and elements of collagen-based matrices in nacre matrices suggests that elements of chitin- and collagen-based matrices are deeply rooted and might be part of an ancient biomineralizing matrix. Our results expand the current shell matrix-framework model and provide new insights into the evolution of diverse biomineralization systems.

Project description:For MS characterization of major ampullate spidroin-1 from the three species of Nephila clavipes, edulis and madagascariensis, an experimental approach was developed combining 2-DE with multiple proteolytic in-gel digestion (trypsin, chymotrypsin, pepsin, proteinase K, subtilisin and proteinase 10) followed by NanoLC and mass spectrometry analysis using collision-induced dissociation (CID) and electron-transfer dissociation (ETD) for fragmentation. MASCOT searches were done by using the MASCOT 2.2.06 (Matrix Science, London, UK) against database for protein identification and post-translational modifications.

Project description:To better understand plant-pathogen interactions and the activation of defense mechanisms, this study proposes to identify the differentially abundant proteins in an interspecific hybrid of passion fruit and its parents, Passiflora edulis and P. setacea, inoculated with CABMV compared to their non-inoculated counterparts. Leaves from the three populations were inoculated and collected 72 h after inoculation. The experiment was set up as a completely randomized design with three replicates. Of the over 500 identified proteins, 100, 290, and 96 were differentially abundant for the hybrid, P. edulis, and P. setacea, respectively, in response to CABMV. In the interspecific hybrid, 41% of the proteins were down-regulated and 59% were up-regulated, compared to control. In the P. setacea parent, 39% of the protein being down-regulated. The P. edulis parent, in turn, showed a distinct profile, in which 82% of the protein were down-regulated and only 18% were up-regulated compared to control. It is suggested that CABMV suppresses accumulation of proteins in susceptible species that may have a key function in plant defense response, such as those linked to proteasome regulation and heat shock proteins. At the same time, it should regulate the accumulation of proteins that may impair viral signaling within the host such as glutathione peroxidase. Despite responding to the infection, P. edulis was not capable of stopping the disease establishment. Results suggest that the disease manifests due to a failure in the signaling system of susceptible species, whose activated response pathways are not sufficient to control it.

Project description:Hypoxia is a characteristic feature of marine environments and a major stressor for marine organisms inhabiting benthic and intertidal zones. Several studies have explored the responses of these organisms to hypoxic stress at the whole organism level with a focus on energy metabolism and mitochondrial response, but the instrinsic mitochondrial responses that support the organelle’s function under hypoxia and reoxygenation (H/R) stress are not well understood. We studied the effects of acute H/R stress (10 min anoxia followed by 15 min reoxygenation) on mitochondrial respiration, production of reactive oxygen species (ROS) and posttranslational modifications (PTM) of the proteome in a marine facultative anaerobe, the blue mussel Mytilus edulis. The mussels’ mitochondria showed increased OXPHOS respiration and suppressed proton leak resulting in a higher coupling efficiency after H/R stress. ROS production decreased in both the resting (LEAK) and phosphorylating (OXPHOS) state indicating that M. edulis is able to prevent oxidative stress and mitochondrial damage during reoxygenation. Hypoxia did not stimulate the rearrangement of the mitochondrial supercomplexes but impacted the mitochondrial phosphoproteome including the proteins involved in OXPHOS, amino acid and fatty acid catabolism, and protein quality control. This study indicates that mussels’ mitochondria possess intrinsic mechanisms (including regulation via PTM mechanisms such as reversible protein phosphorylation) that ensure high respiratory flux and mitigate oxidative damage during H/R stress and contribute to the hypoxia-tolerant mitochondrial phenotype of this metabolically plastic species.

Project description:Marine biomonitoring programs in the U.S. and Europe have historically relied on monitoring tissue concentrations of bivalves to monitor contaminant levels and ecosystem health. By integrating M-bM-^@M-^Xomic methods with these tissue residue approaches we can uncover mechanistic insight to help link tissue concentrations to toxic effects. In an effort to identify novel biomarkers and better understand the molecular toxicology of metal bioaccumulation in bivalves, we exposed the blue mussel, Mytilus edulis L., to sub-lethal concentrations (0.54 M-oM-^AM--M) of cadmium, lead, and a Cd+Pb mixture. Metal concentrations were measured in gill tissues at 1, 2, and 4 weeks, and increased linearly over the 4 week duration. In addition, there was evidence that Pb interfered with Cd uptake in the mixture treatment. Using a 3025 sequence microarray for M. edulis, we performed transcriptomic analysis, identifying 57 differentially expressed sequences. Hierarchical clustering of these sequences successfully distinguished the different treatment groups demonstrating that the expression profiles were reproducible among the treatments. Enrichment analysis of gene ontology terms identified several biological processes that were perturbed by the treatments, including nucleoside phosphate biosynthetic processes, mRNA metabolic processes, and response to stress. To identify transcripts whose expression level correlated with metal bioaccumulation, we performed Pearson correlation analysis. Several transcripts correlated with gill metal concentrations including mt10, mt20, and contig 48, an unknown transcript containing a wsc domain. In addition, three transcripts directly involved in the Unfolded protein response (UPR) were induced in the metal treatments at 2 weeks and were further up-regulated at 4 weeks. Overall, correlation of tissue concentrations and gene expression responses indicates that as mussels accumulate higher concentrations of metals, initial stress responses are mobilized to protect tissues. However, given the role of UPR in apoptosis, it serves as an early indicator of stress, which once overwhelmed will result in adverse physiological effects. A total of 52 samples including initial samples and 3 time-points following exposure to cadmium, lead, or a mixture. There were four replicate samples for each treatment and time-point. An additional three replicate samples were used for RT-qPCR.
