Tbeta4 identification CaCo2 cells

ABSTRACT: The paper describes new experiments about thymosin beta 4 nuclear translocation using different in vitro cellular stress in CaCo2 cell line adding new details regarding the role of this peptide during adaptive and defensive response. To this purpose, CaCo2 cells were submitted to different stress treatments, including serum starvation, DMSO and Butyrate treatment. The expression of Tβ4 was studied in culture cells before and after stress treatment in the cytosolic and nuclear fractions, with the use of different apporaches: immunohistochemistry with anti-Tβ4 antibody; mass-spectrometry characterization and quantification, and mRNA analysis.

Project description:In the HPLC high-resolution ESI-MS profiles of Gingival Crevicular Fluid, four proteins, with exp. monoisotopic ion [M+H]+ at 3440.54 ± 0.06 m/z, 3369.50± 0.06 m/z, 3484.51 ± 0.06 m/z and 3707.77 ± 0.06, eluting between 24.5-26.6 minutes, were detected. The monoisotopic mass values and the manual inspection of the high-resolution MS/MS spectra of the ions at [M+5H]+5 at 689.31, 675.10, 698.11 and 742.76 m/z, and of the ions at [M+4H]+4 at 843.63, 872.39 m/z allowed to establish that the four proteins were alpha defensins 1, 2, 3 and 4.

Quantitative characterization of the neuropeptide level changes in dorsal horn and dorsal root ganglia regions of the murine itch models

We tested two different itch models-the AEW model (known commonly as the dry skin model) and the MC903 model. Performed peptide extraction from the dorsal horn (DH) and dorsal root ganglia regions of these two models and compared their levels with that of control mice.

Human Salivary Prolactine Inducible Protein (PIP)

Project description:In the HPLC high-resolution ESI-MS profiles of both multiple sclerosis subjects and healthy controls saliva samples three proteins, with exp. monoisotopic ion [M+H]+ at 13493.9 ± 0.2 m/z, 13696.9 ± 0.2 m/z (+203 Da, with respect to 13493.9), and 13843.1 ± 0.2 m/z (+147 Da, with respect to 13696.9) eluting between 37.8-38.2 minutes, were detected. The mass difference of 203 Da suggested that the protein with exp. monoisotopic [M+H]+ at 13696.9 ± 0.2 m/z could correspond to the N-acetylhexosamine (theor. monoisotopic ion at 203.1 m/z) derivative of the 13493.9 ± 0.2 m/z protein. On the other hand, the mass difference of 146 Da between the proteins with exp. monoisotopic [M+H]+ at 13843.1 ± 0.2 m/z and 13696.9 ± 0.2 m/z was in agreement with an additional deoxyhesose moiety (theor. monoisotopic ion at 146.1 m/z). Manual inspection of the high-resolution MS/MS spectra of the [M+10H]+10 ions at 1351.00, 1371.32 and 1386.11 m/z allowed to establish that the three proteins were different proteoforms of PIP with the N-terminal glutamine residue converted to pyro-glutamic acid and with two disulfide bonds, but not to assign the glycosylation site. The presence in the MS/MS spectra of low-molecular weight saccharide oxoniun ions at 204.087 and 138.106 m/z confirmed the presence of an N-acetylhexosamine moiety in the protein with [M+H]+ at 13696.9 ± 0.2 m/z.

SWATH-LC/MS/MS of the proteomic response in Campylobacter jejuni 81-176 to sublethal concentrations of seven different bile acids

Project description:To characterize the adaptation processes of C. jejuni to sublethal concentrations of bile acids, that are a crucial component of the intestinal antimicrobial defense, including cholic- , chenodeoxycholic-, taurocholic-, glycocholic-, deoxycholic-, lithocholic-, and ursodeoxycholic acid, we used proteome profiling by label-free mass spectrometry (SWATH-MS). The most toxic bile acids, deoxycholic- and chenodeoxycholic acid induced the most significant changes in protein expression. In detail we found a significant down-regulation of in principle all basic biosynthetic pathways and a general decrease of the transcription machinery. Concurrently, an induction of factors involved in detoxification of reactive oxygen species, protein folding, and bile acid exporting efflux pumps was detected. The metabolism was optimized in such a way that the more energy-efficient aerobic respiration pathway was increased, while the energy-inefficient anaerobic branches of the electron transport chain were down-regulated. Uptake of amino acids in order to catabolize them was also increased. These results show that C. jejuni has a well-built, differentiated system of adaptation to bile acids stress. The findings of this study will enhance the understanding of the biology on the interaction of C. jejuni with the human intestine, and may provide clues to future medical treatment and prevention of C. jejuni infections.

Shedding light on biofilm formation of Halobacterium salinarum R1 by SWATH-LC/MS/MS analysis of planktonic and sessile cells

Project description:Early and mature biofilm formation in the extremely halophilic euryarchaeon Halobacterium salinarum strain R1 was characterized by SWATH-LC/MS/MS. Using a simple surfactant-assisted protein solubilization protocol and one-dimensional ultrahigh performance nanoflow chromatography on the front end, 63.2% and 58.6% of the predicted Hbt. salinarum R1 proteome could be detected and quantified, respectively. Analysis of biophysical protein properties, functional analysis and pathway mapping indicate that we achieved a comprehensive characterization of the proteome. 60.8% of quantified proteins (or 34.5% of the predicted proteome) exhibited significant abundance changes between planktonic and biofilm states, demonstrating that haloarchaeal biofilm formation represents a profound “lifestyle change” on the molecular level. Taken together our results and analysis constitute the first comprehensive study to track the molecular changes from planktonic cells to initial and mature archaeal biofilms on the proteome level. Proteins exemplifying different protein expression level profiles were selected, and their corresponding gene transcripts targeted by qRT-PCR to test the feasibility of establishing rapid PCR-based assays for archaeal biofilm formation.

Top Down Proteomic analysis of Gingival Crevicular Fluid in deciduous, exfoliating and permanent teeth in children

Project description:Gingival Crevicular Fluid, a plasma-derived exudate present in the gingival crevice was collected from deciduous, exfoliating and permanent teeth from 20 children (60 samples) with the aim to characterize and quantify by a mass spectrometry based top-down proteomic approach, the peptide/proteins in the fluid and verify possible variations occurring during the exfoliating process. The results obtained confirmed the presence in Gingival Crevicular Fluid of α-Defensins 1-4, Thymosin β4 and Thymosin β10, as described in previous works and revealed the presence of other interesting peptides never described before in Gingival Crevicular Fluid, such as specific fragments of α-1-antitrypsin, α-1-antichymotrypsin, Thymosin β4 and Thymosin β10 fragments, Fibrinopeptide A, Fibrinopeptide B, S100A, LVV Hemorphin-7 (hemoglobin chain β fragment), as well as some other peptides deriving from α and β subunits of hemoglobin. Statistical analysis evidenced different levels in 5 proteins/peptides in the three groups in particular with higher level in exfoliating teeth. Our study demonstrate that an in-depth analysis of a biological fluid like Gingival Crevicular Fluid, present in small amount, can provide useful information for the understanding of different biological processes like teeth eruption.

Transcriptomic profiling of human BLaER1 pre-B cells transdifferentiation into macrophages

Project description:BLaER1 is a human B cell precursor leukemia cell line derived from the RCH-ACV cells. These cells are stably infected with a construct that overexpresses the transcription factor C/EBPa fused with the estrogen receptor hormone binding domain (ER) and GFP. Upon induction with beta-estradiol, C/EBP is internalized into the nucleus, promoting massive transcriptional changes and inducing the transdifferentiation of these pre-B cells into functional macrophages. This process, that lasts 7 days, can be monitored by the detection of specific B cell and macrophage surface markers by flow cytometry. With the goal of understanding the interplay between chromatin and transcription, we have generated transcriptomic data by RNA-Seq in twelve time points along the transdifferentiation process, in two biological replicates.
