Grb2 interactome data with and without EGF stimulation

ABSTRACT: Growth factor receptor-bound protein 2 or Grb2 is an adaptor protein involved in signal transduction. It is widely expressed and integral to several cellular functions, including cell proliferation. We designed this study to capture interacting partners of Grb2 in absence and presence of epidermal growth factor (EGF). HEK 293 cells, expressing tagged Grb2, were cultured with SILAC labels in presence and absence of EGF. Grb2 interactome was enriched by immunoprecipitation and subjected to mass spectrometry. This data will help in understanding the regulatory effect of EGF stimulation on Grb2 protein and its interactors.

Growth factor receptor-bound protein 2 (Grb2) is an adaptor protein involved in the signal transduction pathways. Project description:The biological significance of the down-regulation of miR-376c in HuCCT1 cells is unknown. We revealed that miR-376c significantly reduced EGF-dependent cell migration through its target GRB2 in HuCCT1 cells. To elucidate key molecules in EGF-dependent HuCCT1 migration, in which GRB2 upregulation is caused by abnormal suppression of miR-376c, we compared expression profiles of mRNAs affected by pre-miR-376c and siRNA targeting GRB2. Comparison of E14.5 embryonic kidney transcriptomes (Grb2 ureteric-specific knockout vs wild-type)

Project description:Grb2 is a small SH2-SH3 adaptor molecule that interacts with activated tyrosine kinase receptors (RTKs), providing a crucial link towards downstream activation of pro-proliferative and pro-survival ERK and Akt signaling pathways. Ret and FGFR2, two RTKs that interact with Grb2, play important roles in ureteric branching and the proper establishment of the renal collecting duct system, but it is unclear whether Grb2 is required in this process. In this study, we selectively ablated a conditional floxed allele of the Grb2 gene within the ureteric epithelial lineage in mice and demonstrate that Grb2 signaling is essentially required for proper collecting duct development. Ureteric Grb2 deficiency results in perinatal lethality and severe renal hypodysplasia closely reminiscent of the rudimentary kidney phenotypes observed in Ret-null mutant mice. Grb2 loss attenuates ERK and Akt activation in the ureteric epithelia resulting in pronounced impairment of ureteric branching. Gene expression analysis reveals that Grb2 deficiency results in defective induction of genes implicated in both ureteric branching and reciprocal mesenchymal metanephric induction. Our findings therefore strongly indicate that Grb2 is a physiologically-relevant major RTK signaling relay partner that promotes renal branching morphogenesis and the proper development of the collecting duct system and the urogenital tract. Delineating the Rb1 interactome data and its modulations during cell cycle progression.

Project description:The retinoblastoma (Rb) protein is a potent tumor suppressor which is known to negatively regulate the cell cycle as well as tumor progression. Phosphorylated Rb protein (pRb) has been demonstrated to be in-charge for the key G1 checkpoint, blocking entry into S-phase and thereby the cell growth. This study was designed to capture interacting protein partners of Rb1 as the cell cycle progresses. Rb1 expressing HEK-293 cells were cultured in light, medium and heavy SILAC labels to capture the changes in Rb1 interactome as the cell cycle progressed from G0 to G1S and then to G2 phase, respectively. This data might help in understanding the cell cycle regulatory effect of Rb1 protein and complement the available information on its interacting partners.

Mass spectrometry analysis of the Themis1 interactome in murine thymocytes

Project description:Themis1, a recently described T-lineage specific protein, is essential for thymic positive and negative selection. Although Themis1 has been clearly identified as a component of the T cell antigen receptor (TCR) signalosome, its precise role in TCR signaling remains unclear. Here, we used quantitative proteomic and TCR signaling reporter mice to gain insight into Themis1 signaling function. Mass spectrometry analysis of the Themis1 interactome identified Grb2, SHP1 and Vav1 as the principal interacting partners of Themis1 in thymocytes. The dataset contains mass spectrometry results from the analysis of 6 different kind of AP-MS purifications (based on immunoprecipitation using a Themis1 antibody) starting from the following samples: - thymocytes from WT mice, non stimulated (noted WT NS) - thymocytes from WT mice, stimulated with pervanadate (noted WT P) - thymocytes from GRB2 +/- mice (with decreased expression of GRB2), non stimulated (noted GRB2 NS) - thymocytes from GRB2 +/- mice (with decreased expression of GRB2), stimulated with pervanadate (noted GRB2 P) - thymocytes from Themis1 -/- mice (knock-out for Themis1), non stimulated (noted KO NS) - thymocytes from Themis1 -/- mice (knock-out for Themis1), stimulated with pervanadate (noted KO P) Three biological replicates were prepared for these 6 different conditions (noted, 1,2,3), yielding 18 analyzed samples. Three technical nanoLC-MS runs were acquired for each sample (noted R1, R2, R3), leading to the 54 nanoLC-MS raw files contained in the dataset.

Characterization of PEAK3 pseudokinase as a pro-migratory and -invasive member of New Kinase Family 3

Project description:The New Kinase Family 3 (NKF3) of pseudokinases comprises PEAK1 and PEAK2 as well as the recently-identified PEAK3. PEAK1/2 play fundamental roles in regulating tyrosine kinase signal output and oncogenesis, while PEAK3 remains poorly-characterized. Here we demonstrate that PEAK3 undergoes homotypic association as well as heterotypic interaction with PEAK1/2. PEAK3 also recruits ASAP1/2, Grb2, CrkII, Cbl and PYK2 with effector recruitment being dependent on PEAK3 dimerization. PEAK3 tyrosine phosphorylation on Y24 is also dependent on dimerization as well as Src family kinase activity, and interestingly, is decreased via PTPN12 in response to EGF treatment. Both phosphorylation of Y24 and an intact N-terminal SH3 binding motif are required for optimal binding of Grb2, CrkII and ASAP1. Overexpression of PEAK3 in MDA-MB-231 breast cancer cells enhanced cell elongation and cell motility, while knockdown of endogenous PEAK3 decreased cell migration. In addition, overexpression of PEAK3 in PEAK1/2 compound knock-out MCF-10A breast epithelial cells enhanced acinar growth and invasion in 3D culture, with the latter phenotype dependent on PEAK3 tyrosine phosphorylation and ASAP1 binding. These findings characterize PEAK3 as an integral member of NKF3 with scaffolding roles that promote cell proliferation, migration and invasion.

Mapping the SLP76 interactome in T cells lacking each of the GRB2-family adaptors reveals molecular plasticity of the TCR signaling pathway

Project description:The propagation of intracellular T cell receptor (TCR) signals involved various adapter proteins that are important molecular switches to connect proteins together. The global characterization of changes in protein-protein interactions following genetic perturbations is critical to understand the reorganization of protein networks associated with the observed phenotypes. Here, by combining genome editing techniques and AP-MS analysis we determined and quantified the molecular reorganization of the SLP76 interactome resulting from the inactivation of each of the three GRB2-family adaptors. Our data define disruption context-specific and time-dependent networks that form around SLP76 after TCR stimulation, and selectively preserved or highly affected protein complexes that regulate T cell activation.

Phosphorylation of the multi functional signal transducer B-cell adaptor protein (BCAP) facilitates recruitment of multiple SH2/SH3 proteins including Grb2.

Project description:B-cell adaptor protein (BCAP) is a multimodular, multi-functional signal transducer that regulates signal transduction processes in leukocytes including macrophages, B-cells and T-cells. In particular BCAP acts as a negative regulator of inflammatory signaling by the Toll-like receptors. Here we characterize the complex phosphorylation patterns of BCAP and use a novel protein complex trapping strategy (virotrap) to identify interaction partners. This analysis identifies known interactions with BCAP with PI3K p85 subunit and Nck, together with novel SH2 and SH3 domain adaptor proteins notably Grb2 and CRKL. We show that the SH3 domain of Grb2 can bind to BCAP independently of phosphorylation, suggesting that the SH2 domains mediate interaction with activated receptor tyrosine kinase complexes including the CD19 subunit of the B-cell receptor. Our data also suggest that PI3K p85 subunit binds to the BCAP via SH3 domains forming an inactive complex that is activated subsequently by sequential binding with the SH2 domains. Taken together our results indicate that BCAP is a complex hub that processes signals from multiple pathways in diverse immune system cells.
