HPLC-MS/MS analysis of proteins in serum samples

ABSTRACT: HPLC-MS/MS analysis of proteins in serum samples using high-resolution mass spectrometry. Dataset of 50 samples from patients with schizophrenia (Series "SCH") and 50 samples from healthy volunteers (Series "CNT") and the data analysis were carried out using shotgun ultra-high resolution mass spectrometry.

Project description:We sought to identify a characteristic profile of proteins in blood plasma samples taken from patients with colorectal cancer using mass spectrometry. We obtained mass spectrometry data of 38 samples from patients with colorectal cancer and 41 samples from healthy volunteers and the data analysis were carried out using panoramic ultra-high resolution mass spectrometry. We carried out a comparative analysis for two series "case" and "control" of protein profiles, differently expressed proteins and proteins with posttranslational modifications.

Project description:Dataset for a comparative analysis of protein profiles of three series of mouse macrophage cell line PMJ2R samples including the control uninfected series (sample M) and infected with Tick-borne encephalitis virus, strain Hypr original isolate on the second (H2) and sixth day (H6) after infection was carried out using panoramic ultra-high resolution mass spectrometry. The analysis of measurements was performed using the proteomics search engine. The findings of the panoramic mass spectrometric analysis indicated that the protein composition of the cell samples is quite similar with respect to the number of identified proteins, protein composition and relative abundance. We identified a group of proteins specific for cell series H2 and H6.

Monocytic ontogeny of regenerated macrophages characterizes the mesotheliomagenic responses to carbon nanotubes

Project description:Macrophages not only derive from circulating blood monocytes, but also originate from embryonic progenitors that can proliferate. This dichotomic origin has an impact on macrophage functions in steady state and pathological conditions. Long needle-like carbon nanotubes (CNT) damage serous membranes and induce mesothelioma like asbestos fibers. Macrophages predominantly infiltrate CNT-induced injured tissue and contribute to cancer development. Here, we revealed the exact ontogeny of macrophage populations after CNT exposure by comparing the response to needle-like mesotheliomagenic CNT-7 with tangled-like non-mesotheliomagenic CNT-T. In a rat peritoneal cavity model, both CNT induced a rapid and complete depletion of MHCIIlow resident macrophages generating an empty niche available for macrophage repopulation. Macrophage disappearance after mesotheliomagenic CNT-7 was followed by a substantial inflammatory reaction, and macrophage replenishment completed after 7 days. Fifteen days after non-mesotheliomagenic CNT-T, macrophage repopulation was incomplete and accompanied by a limited inflammatory reaction. Cell depletion experiments, flow cytometry and RNA-seq analysis demonstrated that, after CNT-7, resident macrophages were mainly replaced by an influx of monocytes, which differentiated locally into MHCIIhigh inflammatory macrophages. In contrast, the low inflammatory response induced by CNT-T was linked to the activation of self-renewing MHCIIlow macrophages. In conclusion, the mesotheliomagenic response to CNT specifically relies on macrophage niche recolonization by monocytic progenitors. The homeostasis returns after exposure to non mesotheliomagenic CNT-T involves, in contrast, a macrophage maintenance by proliferation. Macrophage depletion and repopulation are thus decisive events characterizing the carcinogenic activity of fibrous particles.

Gene expression profiles in human alveolar epithelial cell line exposure to single carbon nanotubes

Project description:To further development of our gene expression approach to assess the effects of manufactured nanomaterials at the molecular level, we have employed whole genome microarray expression profiling as a discovery platform to identify genes with the potential to distinguish characterization of physico-chemical properties of single-wall carbon nanotubes (SWCNTs). Three kinds of single-wall carbon nanotubes (SWCNTs), CNT-1, CNT-2 or CNT-3 induced gene expression in human alveolar epithelial cell lines were measured at 24 and 48 hours exposure to doses of 90.8Î¼g/mL (CNT-1), 109.0Î¼g/mL (CNT-2), and 77.0Î¼g/mL (CNT-2). Three independent experiments were performed at each time (24 or 48 hours) using different untreated control for each experiment (n=4).

Project description:Rye, wheat and barley contain gluten, proteins that trigger immune-mediated inflammation of the small intestine in people with coeliac disease (CD). The only treatment for CD is a lifelong gluten-free diet. To be classified as gluten-free by the World Health Organisation the gluten content must be below 20 mg/kg, but Australia has a more rigorous standard of no detectable gluten and not made from wheat, barley, rye or oats. The purpose of this study was to devise an LC-MS/MS method to detect rye in food. An MS-based assay could overcome some of the limitations of current immunoassays, wherein antibodies often show cross-reactivity and lack specificity due to the diversity of gluten proteins in commercial food and the homology between rye and wheat gluten isoforms. Comprehensive proteomic analysis of 20 rye cultivars originating from 12 countries enabled the identification of a panel of candidate rye-specific peptide markers. The peptide markers were assessed in 16 cereal and pseudo-cereal grains, and in 10 breakfast cereals and 7 snacks foods. Spelt flour was contaminated with rye at a level of 2% and trace levels of rye were found in a breakfast cereal that based on its labelled ingredients should be gluten-free.
