Project description:Yeast form complex highly organized colonies in which cells undergo spatiotemporal phenotypic differentiation in response to local gradients of nutrients, metabolites, and specific signaling molecules. Colony fitness depends on cell interactions, cooperation, and the division of labor between differentiated cell subpopulations. Here, we describe the regulation and dynamics of the expansion of papillae that arise during colony aging, which consist of cells that overcome colony regulatory rules and disrupt the synchronized colony structure. We show that papillae specifically expand within the U cell subpopulation in differentiated colonies. Papillae emerge more frequently in some strains than in others. Genomic analyses further revealed that the Whi2p-Psr1p/Psr2p complex (WPPC) plays a key role in papillae expansion. We show that cells lacking a functional WPPC have a sizable interaction-specific fitness advantage attributable to production of and resistance to a diffusible compound that inhibits growth of other cells. Competitive superiority and high relative fitness of whi2 and psr1psr2 strains are particularly pronounced in dense spatially structured colonies and are independent of TORC1 and Msn2p/Msn4p regulators previously associated with the WPPC function. The WPPC function, described here, might be a regulatory mechanism that balances cell competition and cooperation in dense yeast populations and, thus, contributes to cell synchronization, pattern formation, and the expansion of cells with a competitive fitness advantage.

Quantitative interactome profiling of rabies virus P-protein isoforms P1 and P3

Project description:RNA viruses must achieve complex replication and immune evasion functions using limited coding capacity. The rabies virus P protein is a multifunctional viral protein expressed in multiple isoforms via alternative translation. In this study, we investigate how structural and functional diversity arises between two isoforms—P1 (full-length) and P3 (a naturally truncated form)—and how these differences influence host protein interactions. We used GFP-tagged P1 and P3 proteins expressed in HEK293T cells to perform quantitative immunoprecipitation-mass spectrometry (IP-MS) and define isoform-specific interactomes. This dataset includes raw and processed IP-MS data for GFP-P1, GFP-P3, and GFP control (each in quadruplicate), and supports the conclusion that conformational and functional diversification of viral proteins enables expanded host interface capacity. This study provides new mechanistic insights into how viruses exploit isoform diversity and RNA-driven interactions to overcome genome limitations.

Differentially expressed genes in H2AX knockdown cells undergoing apoptosis

Project description:H2AX has been characterized as a novel tumor suppressor protein. Difficiency of H2AX will result in apoptotic inhibition of cancer cells. However, how H2AX epigenetically regulates apoptosis of cancer cells is still unclear. To reveal the genes expression regulated by H2AX and involved in apoptosis, the microarray profiling analysis was employed to identify differentially expressed genes in H2AX knockdown lung cancer cells and control ones after apoptotic induction. H2AX was knockdown by miRNA interfering system in human lung cancer originated cell A549 and the stable cell line named P1, while the control stable cell line named C. Genes with greater than 1.2-fold change and P-value ï¼0.05 were identified as differentially expressed genes between C and P1 cells both with apoptosis induction. The two groups including control (C) and H2AX knockdown lung cancer cells (P1) were harvested 48 h after VP-16 treatment. Three independent experiments were performed for each group.
