Project description:Vaccination with a new vaccine against bovine viral diarrhea (BVD) caused unwanted immune reactions through alloreactive antibody production in 5–10% of vaccinated cows. Transfer of these alloreactive antibodies via colostrum caused highly lethal (90%) bovine neonatal pancytopenia (BNP) in calves. Despite the assumption of an immune deviant lymphocyte (PBL) phenotype, the exact immunological mechanisms driving these unwanted immune reactions are not fully understood to date. To gain deeper insight into these mechanisms, we analyzed the PBL secretome from alloreactive antibody producers (BNP cows) and non-alloreactive responders (controls) after stimulation with pokeweed mitogen (PWM).

Project description:Bovine paratuberculosis is a serious chronic disease of the gastrointestinal tract caused by Mycobacterium avium subspecies paratuberculosis (MAP). The aim of this study was to detect proteomic changes in peripheral blood mononuclear cells (PBMC) from cows of the same herd with different MAP infection status after co-incubation with viable MAP in vitro using label-free LC-MS/MS. Our data contribute to a better understanding of the bovine immune response and mechanisms of susceptibility to MAP.

Project description:The heart tissue is a potential target of various noxae contributing to the onset of cardiovascular diseases. However, underlying pathophysiological mechanisms are largely unknown. Human stem cell-derived models are promising, but a major concern is the cell immaturity when estimating risks for adults. In this study, 3D aggregates of human embryonic stem cell-derived cardiomyocytes were cultivated for 300 days and characterized regarding degree of maturity, structure, and cell composition. Furthermore, effects of ionizing radiation (X rays, 0.1–2 Gy) on matured aggregates were investigated, representing one of the noxae that are challenging to assess. Video-based functional analyses were correlated to changes in the proteome after the irradiation. Cardiomyocytes reached maximum maturity after 100 days in cultivation, judged by α-actinin lengths, displayed typical multinucleation and branching. At this time, aggregates contained all major cardiac cell types, proven by the patch-clamp technique. Matured and X-ray-irradiated aggregates revealed a subtle increase in beat rates and decrease in rhythmicity in a dose-dependent manner compared to non irradiated sham controls. The proteome analysis provides first insights into signaling mechanisms contributing to cardiotoxicity. Here, we propose an in vitro model suitable to screen various noxae to target adult cardiotoxicity by preserving all benefits of a 3D tissue culture.

Project description:Laron Syndrome (LS) is a rare hereditary disease in humans caused by a mutation in the growth hormone receptor gene which leads to high levels of circulating GH [1]. The disease is characterized by short statue, obesity, transient hypoglycemia during youth and low incidences of cancer and diabetes [2]. Thus making it interesting to study the underlying mechanisms. Recently a porcine model to mimic human LS was developed [3]. Growth hormone receptors are expressed on various cells of the immune system yet the extent to which dysregulation of GH might affect immune function in LS is poorly understood [4]. Therefore we decided to characterize the influence of growth hormone receptor knockout on the immune function of these animals by analyzing the proteome of their CD4+ and CD4- peripheral blood mononuclear cells (PBMC). 1. Project description:Primary cilium is a specialized sensory organelle protruding from apical surface of most cells that transmits environmental information into cells. Its length is tightly controlled by various mechanisms such as the frequency or the cargo size of the intraflagellar transport trains which deliver the building materials essential for the growing cilia. We demonstrate that the sialoglycan interacting galectin 8 regulates the process of primary ciliogenesis, and as the epithelia become polarized, there are more galectin 8 being apically secreted. These extracellular galectin 8 molecules apparently bind to a lipid raft enriched domain at the base of the primary cilia, and binding of galectin 8 at this critical region triggers rapid growth of primary cilia by perturbing the barrier function of transition zone. To identify the ciliary molecule which mediates the cilia-lengthening effect of galectin 8, we approached by adopting sequential apical domain selective biotinylation, surface GST-galectin 8 fusion protein incubation, followed by two-step affinity purification scheme. We then processed these apical surface galectin 8-interacting candidates by mass spectrometry. We totally identified 254 reproducible candidates in two independent experiments. There were 96 and 117 plasma membrane candidates noted from the first and second experiments respectively. We focused on 66 candidates categorized as plasma membrane proteins (~25% of total candidates) which were repetitively identified in two separate experiments.

Project description:Host cell proteins are inevitable contaminants of biopharmaceuticals. Here, we performed detailed analyses of the host cell proteome of moss (Physcomitrella patens) bioreactor supernatants using mass spectrometry and subsequent bioinformatics analysis. Distinguishing between the apparent secretome and intracellular contaminants, a complex extracellular proteolytic network including subtilisin-like proteases, metallo-proteases and aspartic proteases was identified. Further, we confirmed predicted cleavage sites of 40 endogenous signal peptides employing an N-terminomics approach.

Project description:Osmotic stress is detrimental for the plant which survival relies heavily on proteomic plasticity. Protein ubiquitination is a central post-translational modification (PTM) in osmotic mediated stress. Plants use the ubiquitin proteasome system to modulate protein contents required to respond to and tolerate adverse growth conditions and a role for ubiquitin to mediate endocytosis and trafficking of plant plasma membrane proteins has recently emerged. In this study, using the K-Ɛ-GG antibody enrichment method integrated with high-resolution mass spectrometry, a list of 786 ubiquitinated lysine (K-Ub) residues in 451 proteins of which 50 % were transmembrane proteins was compiled for Arabidopsis root membrane proteins, increasing the database of ubiquitinated substrates in plants. Such analysis allowed to evidence specific ubiquitination motifs, a role for ubiquitination in the internalization and sorting of cargo proteins and ubiquitination dialog with other post-translational modifications (PTMs). In silico interactomics analysis allowed to pinpoint two E2 ligases, UBC32 and UBC34 targeting membrane proteins and putatively involved in response to osmotic stress. The simultaneous quantification of proteome and ubiquitinome after plant treatment with mannitol, suggested a minor role for ubiquitination in protein degradation. Then, this study revealed that a major plasma membrane aquaporin (PIP2;1) harbors two ubiquitination sites that dialog with N-terminal acetylation and C-terminal phosphorylation, to putatively limit PIP2;1 degradation and to favor its internalization contributing to a decreased root hydraulic conductivity (Lpr) while maintaining its cellular abundance.

Project description:Acute postsurgerical pain and it's management represent a major clinical challenge. In particular, severe and prolonged pain impairs immediate recovery and might lead to long-term consequences like chronic pain, opioid abuse, and reduced quality of life. Rodent incision models greatly contributed to the understanding of cutaneous wound repair, but the underlying mechanisms' knowledge remains incomplete, yet translational data are urgently needed to develop novel treatment options and preventive measures. We combined extensive sensory phenotyping with unbiased quantitative proteomics in human and mouse skin after an incision to assess interspecies protein signatures to overcome this gap. Additionally, stratification of human volunteers based on the hyperalgesic area after surgery in correlation with the corresponding proteomic fingerprint revealed novel insides into human-specific mechanisms.Unbiased skin proteome analysis of both species unveiled comparable protein signatures after incision, notably for inflammatory activity and actin polymerization. Upon incision, we could discern 50 commonly regulated proteins, amounting to 61% of all regulated human and 10% regulated mouse proteins. Notably, the direction of regulation upon incision, i.e. up-or downregulated, was conserved in humans and mice to a great extent. Integrative analysis of pain-related phenotyping with quantitative mass spectrometry identified hitherto unknown skin protein signatures, providing a tool for stratification on the protein level. Protein-protein interaction (PPI)-networks differed between volunteers with small incision-related hyperalgesic areas (termed"low responder") versus those with large areas ("high responder"). In particular, PPIs of volunteers exhibiting a large hyperalgesic area were characterized by a predominant dysregulated proteolytic environment associated with persistent inflammation reponse. In contrast, PPI- networks of low responders point to anti-inflammatory processes. Here, we present a framework for specific-specific alterations in human and mice upon incision, highlighting similarities and differences on a protein network level. Moreover, the detection of developing hyperalgesia and pain after surgery in volunteers and it's correlation with corresponding molecular fingerprints indicates the need for tailored treatments to prevent chronification processes in humans after surgery.
