Project description:Sevoflurane is the most commonly used general anesthetic in pediatric surgery, but it has the potential to be neurotoxic. Previous research found that long-term or multiple sevoflurane exposures could cause cognitive deficits in newborn mice but not adult mice, whereas short-term or single inhalations had little effect on cognitive function at both ages. The mechanisms behind these effects, however, are unclear. In the current study, 6- and 60-day-old C57bl mice in the sevoflurane groups were given 3% sevoflurane plus 60% oxygen for three consecutive days, each lasting 2 hours, while those in the control group only got 60% oxygen. The cortex tissues were harvested on the 8th or 62nd day. The tandem mass tags (TMT)pro-based quantitative proteomics combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysi were applied to analyze the influences of multiple sevoflurane anesthesia on the cerebral cortex in mice with various ages. A total of 6247 proteins were measured using the combined quantitative proteomics methods of TMTpro-labeled and LC-MS/MS, 443 of which were associated to the age-dependent neurotoxic mechanism of repeated sevoflurane anesthesia. Our findings would help to further the mechanistic study of age-dependent anesthetic neurotoxicity and contribute to seek for effective protection in the developing brain under general anesthesia.

Project description:Cerebral ischemia/reperfusion injury (CI/RI), including neurological behavior deficits, cerebral infarction, blood-brain barrier (BBB) dysfunction, and neuroinflammation, et al. is a severe challenge in treatment of ischemic stroke. Traditional Chinese medicine (TCM) with the characteristics of multi-components and multi-functions for multiple targets/pathways has been historically used in the treatment of stroke and post stroke recovery in China. QiShenYiQi (QSYQ), a representative component-based Chinese medicine, is capable of reducing organ injury caused by ischemia/reperfusion via multiple mechanisms. Recently, we have previously reported that the positive action of QSYQ against the acute phase of CI/RI is partly via modulation of neuroinflammatory response. However, the effects and underlying mechanisms of QSYQ on subacute phase of CI/RI remain unknown, which are aimed to investigate in present study. The pharmacological action of QSYQ treatment on brain damage was estimated in the experimental stroke rats underwent 90 min ischemia and 8 days reperfusion by assessing the neurological and locomotor deficits, cerebral infarction, brain edema, and BBB integrity. Furthermore, we used TMT-based quantitative proteomics technology to identify significantly differentially expressed proteins following QSYQ treatment, which could give important clues for revealing the possible mechanism underlying the positive role of QSYQ in CI/RI. Besides, immunohistochemistry, western blot analysis, RT-qPCR, and rat ELISA kits were executed to further verify the accuracy of proteomics analysis results. As a consequence, we found that treatment with QSYQ (600mg/kg) for 7 days obviously improved neurological and behavioral impairment, attenuated infarct volume and brain edema, and alleviated BBB breakdown in stroke rats. Bioinformatics analysis suggested that differentially expressed protein galectin-3 mediated inflammatory response was closely related to the beneficial effect of QSYQ. Results of immunohistochemistry, western blot analysis and RT-qPCR showed that the model rats treated with QSYQ (600mg/kg) markedly downregulated the mRNA and protein expression level of galectin-3 in brain tissues compared to the untreated stroke rats. In addition, the decrease of mRNA level in brain tissues and contents in serum of TNF-α and IL-6 following QSYQ treatment were also observed. These interesting finding manifested that the effective action of QSYQ against subacute phase of CI/RI was partly via regulating galectin-3 mediated inflammatory reaction.

Bioinformatics analyses of piRNAs in Drosophila ovaries suggested that piRNAs are produced by two systems, the primary processing pathway and the amplification loop, from repetitive genes and piRNA clusters in the genome. The amplification loop occurs in a Dicer-independent, PIWI-Slicer-dependent manner. However, the primary processing pathway remains largely conceptual. Here we show that in ovarian somatic cells, which lack Aub and AGO3 but express Piwi, the primary processing pathway for piRNAs indeed exists. Project description:The immune system is thought to be fragile in the neonate, which is susceptible to pathogens. Exosomes are a type of vehicles existing in the body fluid and participate in many biological processes, especially the immune response. Inorder to investigate the roles that exosomes may play during virus infection in the neonate, porcine epidemic diarrhea virus (PEDV), a devastating enteric virus to newborn piglets, was selected for infection. Serum exosomes were then isolated from the newborn-piglets infected or mock-infected with PEDV and followed by a label-free LC-MS/MS based comparative quantitative proteomic analysis. Among 441 proteins detected in the serum exosomes, there were lots of complement proteins. The expression level of the complement C3, C6 and CFB suffered drastic changes due to PEDV infection. After the confirmation by western-blot assay, we then investigated the function of these exosomes on PEDV infection and discovered that the exosomes from mock-infected newborn piglets restricted PEDV infection but this inhibition disappeared after exosomes were heat-inactivated, suggesting that the complement is one of the key antiviral molecules. These findings will facilitate the understanding of the antiviral response of the neonate mediated by exosomes

Project description:Scope: Adipose tissue is regarded as a true endocrine organ. Recent studies showed that adipose tissue derived exosomes could serve as carrier of circulating miRNAs to regulate distant targets. However, the characteristics of exosomal proteins released from adipose tissue have not been investigated yet. Methods and Results: In this study, we conducted a complementary protein profiling on exosome-like vesicles derived from adipose tissue (ELV-AT) with Label-free Quantitative Proteomic Analysis. A total of 3229 ELV-AT proteins were identified, among which only 266 proteins have been annotated as adipokines. 3 undefined adipokine candidates (NPM3, STEAP3, and DAD1) were selected for further validation. These 3 proteins were expressed in both white and brown adipose tissue, upregulated during adipogenic differentiation of both 3T3-L1 cells and adipose derived stem cells (ADSCs). Expressions of NPM3, DAD1 in ELV-AT were significantly decreased in obese subjects compared with lean controls while obesity could not alter the expression of STEAP3. Conclusions: Our profiling study of the ELV-AT proteins expands the list of adipokines and highlights the pivotal role of exosomal adipokines in the regulation of multiple biological processes within adipose tissue

Project description:Extracellular vesicles (EVs) have been extensively studied in animal cells, and play an important role in cell-to-cell communications. Emerging evidence shows that EVs also act as important agents in plant-microbe interactions. However, the mechanisms by which EVs mediate cross-kingdom interactions between plants and microbial pathogens remain largely elusive. Here, proteomic analyses of soybean root rot pathogen Phytophthora sojae EVs identified tetraspanin family proteins, PsTET1 and PsTET3, that can be recognized by Nicotiana benthamiana to trigger plant immune responses. PsTET1 and PsTET3 were redundantly required for the full virulence of P. sojae. Further analyses revealed that the large extracellular loop (EC2) of PsTET3 is the key region recognized by N. benthamiana and also by Glycine max, and that recognition depends on the plant receptor-like kinase BAK1. TET proteins from oomycete and fungal plant pathogens could be recognized by N. benthamiana and induce immune responses. However, plant-derived TET proteins failed to do so, due to the divergent sequences of the final 16 amino acids of EC2, which ultimately makes plants distinguish self and non-self EVs, triggering active defenses against pathogenic eukaryotes.
